2.Analysis of schistosomiasis epidemic situation in Xinjiangkou Township, Sonzi City,from 2004 to 2013

Objective To master the status of schistosomiasis transmission in Xinjiangkou Township,Sonzi City,from 2004 to 2013. Methods The data of schistosomiasis epidemic situation were collected and analyzed statistically in Xinjiangkou Town-ship,Sonzi City,from 2004 to 2013. Results From 2004 to 2013,the areas with Oncomelania hupensis snails were 193.2-185.3 hm2,and the highest density of infected living snails was 0.000 70 snail/0.1 m2 in 2007. However,after 2011,no infected living snails were found. The positive rate of blood tests was 9.46%and schistosomiasis infection rate was 7.36%in 2005,and the posi-tive rate of blood tests was 3.27%and schistosomiasis infection rate was 0.57%in 2013. There were 7 cases of acute schistosomia-sis in 2004 and 2005,respectively,and there were no acute schistosomiasis patients after 2006. The schistosome infection rate of cattle was 18.27%in 2004,and after 2007,the infection rate decreased below 5%. In 2013,all the cattle were obsoleted. Conclu-sion The schistosomiasis transmission has reduced to a low level in Xinjiangkou Township. However,to realize the aim of schisto-somiasis transmission controlled,the comprehensive control measures mainly based on infectious source control should still be strengthened.
3.Recent developments in pathogenesis of Hirschsprung's disease

Hirschsprung's disease (HD) is a conginental disorder of enteric nervous system (ENS) and is associated with the abnormal migration of gut neural cerst cell (GNCC).Recent studies showed that the regulation of this migration is a complex gene network.By the development of epigenetic and stem cells,this network is becoming much more clear.Transplantation of the modified gut neural crest stem cells/gut neural crest precursors(GNCSC/GNCP) acquired from the HD bowl is becoming the potential therapy of HD.This paper summerizes the recent advances and is to promote better understanding the pathogenesis of HD.
4. Endothelial progenitor cells: Functions and clinical application in cardiac and vascular diseases

Circulating endothelial progenitor cells (EPCs) derived from bone marrow are capable of endocrine, reendothelialization of injured blood vessels and neovascularization of ischemic lesions; besides, they also play a major role in the pathogenesis of atherosclerosis. However, the number and functions of circulating EPCs are greatly declined in patients suffering from cardiovascular diseases. Functions of EPCs are associated with not only pathophysiological factors such as aging, hypertension, diabetes and hyperlipidemia, but also personal lifestyle such as exercise, body weight and smoking, which directly influence the development of cardiovascular diseases. In this review, we mainly focus on the relationship between EPCs and cardiovascular risk factors and the potential roles of EPC therapy for cardiovascular diseases.
5. In vitro skin transplantation and TUNEL assay in evaluation of cellular immunity caused by infusion of rhesus mesenchymal stem cells

Objective: To transfuse rhesus bone marrow derived mesenchymal stem cells (MSCs) to rhesus leucocytes antigen (RhLA) mismatched recipients, in attempt to study the immunological reaction between recipients' T lymphocytes and the skin of donor or a third party. Methods: Bone marrow aspirates were collected from femurs of donor rhesus, and the mononuclear cells were isolated and resuspended in low glucose Dulbecco's Modified Eagles Medium (DMEM) containing 2% fetal bovine serum. Phenotype of MSCs was analyzed by flow cytometry and the differentiated cells were identified by relevant specific staining. Allogeneic MSCs were given to recipients (single infusion) without preconditioning. Peripheral blood lymphocytes from recipients were co-cultured in vitro with the donor's skin or a third rhesus for 72 h before and 20 days, 40 days, and 60 days after infusion. The pathological lesions were evaluated and the keratinocyte apoptosis of the skin sections was determined by TUNEL assay. Results: Twenty days after infusion, the pathological lesion in in vitro skin explants from MSCs donor was less and the apoptosis rate was declined to (12.2±1.9)% from a pre-infusion level of (23.0±4.9)%. The apoptosis of cells in skin explants from the third party also declined to (11.8±1.4)% from a pre-infusion level of (22.0±2.0)% (P<0.05). Intensive immunological reaction was observed between recipient and donor/the third party on day 40 and day 60 after transplantation, with the apoptosis rate being (22.0±3.2)%, (22.0±2.5)% (donor) and (23.2±2.4)%, (24.0±2.6)% (the third party). Conclusion: Allogenic MSCs transfusion can decrease the immunological reaction between donors and recipients in rhesus, but this decreased reaction is temporary and reversible.

7. Characterization and in vitro release of Periplaneta americana-loaded nanoparticles

Objective: To prepare Periplaneta americana extract CII-3-loaded nanoparticles for oral administration, and to investigate the in vitro release profile of CII-3-loaded nanoparticles and the protection of nanoparticles for CII-3 extract in vitro. Methods: The in vitro release behavior of the CII-3-loaded nanoparticles was carried out in the artificial gastric juice, artificial intestinal juice, and pH 7.4 PBS, and the fitting of different models was performed based on the accumulative drug release percentages observed by Folin-reagent method; The amino acid content of CII-3 and CII-3-loaded nanoparticles at different time points in artificial gastric juice was determinated by ninhydrin colorimetry and degradation rates of the two drugs were compared. Results: The mean size of the resulted nanoparticles was about (109.9 ± 0.6) nm and the Zeta potential was (-37.5 ± 3.5) mV; The accumulative release level of CII-3-loaded nanoparticles Qt was (22.63 ± 1.17)% in the artificial gastric juice in the first 2 h, and then, in the artificial intestinal juice, the accumulative release level of CII-3-loaded nanoparticles over a period of 60 h was (72.35 ± 1.90)%, which was in line with Higuchi model release equations, Qt = 8.287 2 t1/2 + 7.758 6. The accumulative release level of CII-3-loaded nanoparticles was (72.67 ± 1.65)% over a period of 10 d, which was in line with Weibull equation, lnln[1/(1-Qt)] = 0.403 7 ln(t-0.411 9)-1.713 3; The CII-3 was completely degraded in 4 h in the artificial gastric juice, while about 70% of CII-3 contained in nanoparticles was degraded. Conclusion: CII-3-loaded nanoparticles have a satisfactory sustained in vitro release effect, and the stability of CII-3 contained in the nanoparticles is improved in the artificial gastric juice.
8.New Strategy for anti-HBV therapy: blocking P-8 interaction.

Clinically being applied treatment against chronic hepatitis has three limitations: low response rates, severe adverse effects and a high rate of drug resistance. Hence, novel targets for antiviral therapy need to be developed so as to provide an armory of different strategies. During the replication of hepatitis B virus, the interaction of viral polymerase (P protein, also called P) and epsilonRNA is indispensable for the initiation of reverse transcription via protein priming and the pregenome RNA (pgRNA) packaging. Three strategies are currently developed for blocking P-epsilon interaction: heat shock protein inhibitors, epsilonaptamers and chemical compounds for blocking formation of P-epsilon complex. Previously, our group has for the first time worldwide in vitro screened several aptamers, which are able to interfere with the P-epsilon interaction. A strong inhibition against HBV was observed in vitro and in vivo experiments, respectively. In conclusion, the so far developed chemicals suppressing the P-epsilon interaction may bypass or overcome the viral resistance problems during clinic treatment and represent a highly attractive option for therapeutic intervention.
