4.Efficacy of ultra-early endovascular embolization for ruptured intracranial aneurysms:a Meta-analysis
Chinese Journal of Cerebrovascular Diseases 2017;14(2):64-70
Objective To compare the effectiveness and safety of ultra-early (< 24 h)and delayed (≥ 24 h ) endovascular embolization of ruptured intracranial aneurysms with systematic review. Methods PubMed,Embase, the Cochrane Library,VIP,Wanfang Data, and China National Knowledge Internet (CNKI)were retrieved according to inclusion, exclusion criteria and retrieval strategies, and the clinical literature of ultra-early and delayed endovascular embolization for the treatment of ruptured intracranial aneurysms were obtained. The Review Manager 5. 3 software was used to conduct Meta-analysis for good prognosis rate, postoperative mortality, and postoperative rebleeding rate. Results A total of 10 articles were included,9 of them were retrospective control studies and 1 was prospective control study. A total of 2 021 patients were enrolled, including 970 patients treated with ultra-early treatment;1 051 patients treated with delayed treatment. There was significant difference in the good prognosis rate (OR,2. 67,95% CI 2. 07 -3. 44,P < 0. 01)and the postoperative rebleeding rate (OR,0. 23,95% CI 0. 11 -0. 47)between the ultra - early embolization group and the delayed embolization group (all P < 0. 01). There were no significant difference in the mortality between the two groups (OR,0. 76,95% CI 0. 51 -1. 13,P = 0. 17). The subgroup analysis showed that there were significant differences in the good prognosis rate in the ultra-early group compared with the early group (< 3 d,OR,1. 98,95% CI 1. 33 -2. 95)and the middle and late group (≥3 d,OR,4. 66,95% CI 2. 21 -9. 81,all P < 0. 01). Conclusion Compared with the delayed group, ultra-early embolization of ruptured intracranial aneurysms may improve the good prognosis rate, reduce the rebleeding rate, and not increase the mortality after procedure at the same time. However, more high quality and large sample randomized controlled trials are needed to confirm them.

Objective To identify the potential DNA vaccine candidate which can induce the protective immune response to Toxoplasma gondii by inoculating mice with plasmid DNAs encoding three different forms of P30 antigen (membranous secretory, and intracellular). Methods Three forms of recombinant plasmid: pcDNA3 P30Mb(contain the whole P30 gene sequence, including the gene encoding signal peptide and hydrophobic tail),pcDNA3 P30Se(contain the whole P30 gene sequence, without the gene encoding hydrophobic tail) and pcDNA3 P30In(contain the whole P30 gene sequence, without the gene encoding signal peptide) were constructed by PCR and subcloning technique. The mice were immunized with different forms of recombinant plasmids and IgG antibodies in the mice were detected by ELISA and Western blotting. Results Three forms of expression recombinant plasmid of Toxoplasma gondii P30 gene were successfully constructed. The P30 inserts were identified by restrictive enzyme digestion and sequencing. ELISA and Western blotting analysis demonstrated that specific IgG antibody could be induced in three immunized groups, but there was some difference in appearence time and intensity of IgG.Conclusion Genetically immunization of mice with the recombinant plasmids could elicit specific IgG antibodies. In respect to IgG response, the immune efficiency of the three forms of recombinant plasmids was different at the beginning (2 wk),but 4 wk later approximately same.

Objective To summarize the nursing experience of combination therapy of highly agglutinative staphylococcin (HASL) with cisplatin for patients with malignant pleural effusions. Methods After fully draining of pleural fluid in 62 cases with malignant pleural effusion through pleural puncture through central venous catheters and simply suction bottles, we injected HASL and cisplatin into the patients' pleural cavities. During the therapeutic process, firstly, we drained the pleural fluid fully before injection, secondly, we directed the patients to cooperate well and alter the positions properly, and thirdly, we closely observed the patients' condition and adverse reactions. Results 54 of 62 cases were effective after the therapies. The effective rate was 87.1%.Conclusions In order to achieve a satisfled curative effect and reduce the adverse reactions, we need the cooperation of the patients, fully draining of the pleural fluid before injection and proper alternation of the patients' positions during the treatment process.
8.Efficacy of jaw thrust device in the upper airway obstruction
Chinese Journal of Anesthesiology 2009;29(9):814-815
Objective To evaluate the efficacy of jaw thrust device in the upper airway obstruction. Methods Thirty-five ASA Ⅰ or Ⅱ patients aged 18-64 yr with body mass index < 30 kg/m~2 undergoing elective surgery under general anesthesia were included in this study. The anesthesia face mask was placed and held tightly by 4 straps of JTD and connected to the anesthesia machine. Anesthesia was induced with midazolam, propefol, fentanyl and atracurium. When muscle relaxation was achieved, tracheal intubation was not performed. The airway obstruction was assessed by airway peak pressure and stridor score (breathing sounds detected by auscultation over the trachea). And then tracheal intubation was performed, and the patients were mechanically ventilated. Results There was no significant difference in airway pressure and stridor scores between JTD and jaw thrust maneuver. Conclusion JTD can effectively lift the jaw and improve the upper airway obstruction.
9.LC-PDA-MS/MS analysis and anti-depressant study of Zhizi-Gancao-Chi decoction
Journal of China Pharmaceutical University 2009;40(4):342-347
Aim: To establish an analytical method to study the chemical components in Zhizi-Gancao-Chi(ZGC) decoction, and investigate its anti-depressant effect. Methods: An LC-PDA-MS/MS method was established and applied. The separation was performed on an ODS C_(18) column (250 mm ×4. 6 mm, 5 μm) with a gradient elu-tion. The mobile phase consisted of water/acetic acid( 1 %, v/v) and methanol. The main components were identi-fied according to the UV and MS. The anti-depressant effect was evaluated with mice tail-suspension test and forced swimming test. Results: The components of the ZGC decoction were separated isolated and ten of them were identified. These compounds are classified to iridoid glycosides, flavonoids and triterpene saponins. In addi-tion, its anti-depressant effect was observed for the first time. Conclusion: The LC-PDA-MS/MS method can ef-fectively separate and identify the components of ZGC decoction, which has significant anti-depressant effect. The relationship between the chemical component and its anti-depressant effect needs further study.

Chinese Journal of Tissue Engineering Research 2007;0(10):-
BACKGROUND: Presently used biological factors for inducing bone marrow mesenchymal stem cells (BMSCs) do not obtain mature chondrocytes. Cartilage tissue engineering using BMSCs as seeds does not collect tissue-engineered cartilage, which has value in the clinic. The obtained tissue is cartilage-like tissue. OBJECTIVE: To screen differentially expressed genes between rat BMSCs and chondrocytes by microarray. DESIGN, TIME AND SETTING: The cytology gene study was conducted at the Central Laboratory, First Affiliated Hospital, Dalian Medical University in July 2007. MATERIALS: A total of 8 healthy Sprague Dawley rats aged 2 months were obtained from Animal Experimental Center, Dalian Medical University. 27K Rat Genome Array chip was supplied by Bo’ao, Beijing, China. METHODS: Rat BMSCs and chondrocytes in the aural region were sterilely isolated and cultured in vitro. Total RNA was extracted and purified using Trizol one-step method, and transformed into double chain cDNA probe following reverse transcription. Cy5-dCTP and Cy3-dCTP were used to label BMSCs and chondrocytes, which were hybridized and washed. The fluorescent signals were scanned by a scanner. The values were analyzed and calculated by GenePix Pro 4.0 software. MAIN OUTCOME MEASURES: Gene expression spectrum chip hybridization results. RESULTS: Among the differentially expressed genes (2 times difference), BMSCs as controls, upregulated and downregulated genes were 1 226 and 888, respectively. There were many differential expression gene of BMSCs and chondrocytes. Cy5/Cy3 20 genes are defined as significant differential expression gene. Thus, two important cytokines were found: chondromodulin and connective tissue growth factor. CONCLUSION: The gene chip technique provides an ideal method for screening cytokines during study of tissue-engineered cartilage. Cartilage regulin and connective tissue growth factor highly express in chondrocytes, which indicated that the two have closely association with the differentiation of BMSCs into cartilage.
