1.Role of synovium macrophage during adjuvant arthritis in rat articular damage

AIM: To explore the role of synovium macrophage of adjuvant arthritis (AA) rats in articular damage. METHODS: AA was induced in female Wistar rats by means of intradermal injection with complete Freud's adjuvant. X-ray on hind paws of AA rats was took at different time points and radiograph index (RI) was then evaluated. Synovium macrophages were detected through immunohistochemistry with ED 1 monoclonal antibody. The correlations between RI and synovial lining layer count/synovium macrophage count were also analysed. RESULTS: Synovial lining layer count, both macrophage count in synovial lining layer and sublining layer were positively correlated with RI. CONCLUSION: Synovium macrophages of AA rats may play an important role in articular damage.
3.Inhibition effect of 6-gingerol on hair growth.

OBJECTIVETo investigate the effect of 6-gingerol, the main active component of ginger, on hair shaft elongation in vitro and hair growth in vivo.
METHODSFirstly, Hair follicles were co-cultured with 3 different concentration of 6-gingerol for 5 days and hair elongation in three groups was measured. Secondly, The proliferative effect of 6-gingerol on DPCs was measured using MTT assay. Thirdly, the expression of Bcl-2 and Bax in DPCs were measured using Western blotting. In vivo study, the influence of 6-gingerol on hair growth in C57BL/6 rats was measured through topical application of 6-gingerol on the dorsal skin of each animal.
RESULTSThe length of hair shaft in 20 microg/ml 6-Gingerol group (0.50 +/- 0.08 mm) is less than 0 microg/ml (0.66 +/- 0.19) mm and 10 microg/ml (0.64 +/- 0.03) mm 6-Gingerol group (P < 0.05). In cell culture, compared to 0 microg/ml and 5 microg/ml 6-Gingerol, 10 microg/ml 6-Gingerol can significantly inhibited the proliferation of DPCs (P < 0.05). Along with the growth inhibition of DPCs by 6-gingerol, the Bax/Bcl-2 ratio increased obviously. In vivo study, the hair length and density decreased a lot after using 1 mg/ml 6-gingerol.
CONCLUSIONS6-Gingerol can suppress human hair shaft elongation because it has pro-apoptotic effects on DPCs via increasing Bax/Bcl-2 ratio. It might inhibit hair growth by prolonging the telogen stage in vivo.

4.Clinical observation on two-way quintuple puncture in the treatment of ganglion.

OBJECTIVETo explore the better therapy in the treatment of ganglion.
METHODSNinety cases of ganglion were randomized into a two-way quintuple puncture group, a common quintuple puncture group and a fire needling group, 30 cases in each one. In the two-way quintuple puncture group, the "9-in-1" multiple penetrating needling technique was used. In the common quintuple puncture group, the traditional "5-in-1" multiple penetrating needling technique was applied. In the fire needling group, the traditional multiple fire needling technique was adopted. The treatment was given once a day, 3 treatments made one session and the efficacy was analyzed statistically after 1 session treatment in the three groups.
RESULTSAll of the three therapeutic methods achieved the efficacy on ganglion. The curative rate was 96. 7% (29/30) in the two-way quintuple puncture group, which was better obviously than 66.7% (20/30) in the common quintuple puncture group and 60. 0% (18/30) in the fire needling group (both P<0. 01).
CONCLUSIONThe two-way quintuple puncture technique achieves the remarkably superior efficacy on ganglion as compared with the common quintuple puncture technique and fire needling technique.

5.Application of case teaching method in the teaching practice of nursing nutrition

Objective To understand the case method in the teaching application of nursing nutrition.Methods Set up the teaching quality evaluation sheet,and 449 nursing students that received the teaching application of nursing nutrition by the methods of case teaching were investigated.Results Among the students,89.17% of the students thought that teachers can closely teach the keys of knowledge,related case content with theory closely.93.35% students considered that it can help them deeply understanding,82.92% students thought that it combined with nursing professional characteristics,and case content was vivid (86.91%).More than 90.00% of the students thought that it increased their interesting in the learning of nursing nutrition,and the interaction between teachers and students was increased.And 87.60% students considered that it improved their learning initiative,80.39% students enhanced their deep impression.More than 75.00% students thought that it improved their interpersonal communication,and abilities of problems solving.And 96.27% students were more willing to accept the case method of teaching.Conclusions Case teaching method application can training innovation consciousness,practice ability and the team cooperation spirit of students.
6.LC-MSn analysis of metabolites of 1,2-bis (1,2-benzisoselenazolone-3(2H)-ketone)-ethane, a novel anti-cancer agent in rat.

This study is to elucidate the metabolic pathway of 1,2-[bis (1,2-benzisoselenazolone-3 (2H)-ketone)]-ethane (BBSKE) in rats. Rats were administrated with a single dose of BBSKE 200 mg x kg(-1). The metabolites in rat urine, feces, bile and plasma were identified by LC-MSn analysis. The characterization of fragment ions from LC-MSn chromatography and mass spectrometry was applied to the investigation of structures of metabolites. Three phase I metabolites were detected in rat urine and feces. Two of them were also found in plasma and one existed in bile. These products were derived from oxidized, methylated and S-methylated BBSKE, separately. One phase II glucuronide of BBSKE was also found in bile. Therefore, it is possible that BBSKE was metabolized by oxidization, methylation and glucuronidation.

7.Evaluation of Real-time PCR and Gomori-Methenamine Silver Stain for Diagnosing of Pneumocystis Pneumonia

Objective To compare real-time PCR and gomori-methenamine silver stain in the diagnosis of pneumocystis peumonia (PCP).Methods 2 525 unrepeated specimens from suspected PCP patient admitted in Peaking Union Medical College Hospital were collected in 2014.2 492 samples were detected by gomori-methenamine silver stain,33 samples were detected by real-time PCR,and 429 samples were detected by both methods at the meanwhile.With clinical diagnosis as reference standard,the sensitivity,specificity,positive predictive value and negative predictive value of the two methods were analysised.Results Positive rate of gomori-methenamine silver stain was 1.2 % (30/2 492).The first three specimen types were sputum,tracheal intubation suction and bronchoalveolar lavage fluid,the positive rate was 0.70 % (13/1 845),4.00% (10/250) and 2.72% (7/257) respectively.Positive rate of realtime PCR was 34.20% (158/462),and the positive rate of sputum and bronchoalveolar lavage fluid was 30.61% (105/343) and 44.54% (53/119) respectively.The sensitivity were 13.97% vs 72.07%,specificity were 100% vs 94.24%,positive predictive value were 100% vs 92.14% and negative predictive value were 55.36% vs 78.26% for gomori-methenamine silver stain and real-time PCR respectively.All of which were statistically significant analysed by x2 test for paired data.The x2 value and P alue were x2 =68.625,P<0.01;x2 =4.296,P<0.05;x2 =6.380,P<0.01 and x2 =11.873,P<0.01.Conclusion The real-time PCR had higher sensitivity,fewer interference factors and more clinical diagnostic value,so clinicians should make more use of real-time PCR to diagnose PCP earlier.

Recombinant humanized anti-ricin monoclonal antibody (MIL50) is a recombinant humanized monoclonal antibody targeting ricin. In this study, an ELISA method was used to establish a method for the determination of MIL50 in macaque serum, and a cross design method was used. Twelve rhesus monkeys were intravenously injected 1 mg·kg-1 test preparation (MIL50 freeze-died powder injection) and reference preparation (MIL50 liquid preparation) to determine the plasma concentration of MIL50 at different time points, and the pharmacokinetic parameters were analyzed to compare the pharmacokinetic characteristics of MIL50 liquid preparation and freeze-died powder injection in rhesus monkeys. The research on the elder financial exploitation in foreign countries started earlier, while the research on the elder financial exploitation in China is still in its infancy. Therefore, this paper summarizes the concept, theoretical framework, current situation and countermeasures of elder financial exploitation, with a view to providing reference for the study of elder financial exploitation in China.
