3.Ki-67 expression in T1 primary breast cancer tissue and its relationship with Her-2 and ER

Objective To study the expression of Ki-67 in T1 primary breast cancer tissue and its relationship with Her-2 and ER,evaluate the prognosis of early breast cancer.Method s Sixty cases of women T1 primary breast cancer tissues of 60 female cases from January 2008 to June 2011 were selected.The expressions of Ki-67 were detected by immunohistochemieal method and the relationships between Ki-67 and clinical-pathological fcatures,Her-2 and ER were analysed.Results The expression of Ki-67 in T1 primary breast cancer was 55% among the 60 patients.The expression of Ki-67 was not related with the clinical-pathological type of breast cancer and menopause (P>0.05).The expression of Ki-67 in group with lymph node metastasis was higer than in group without lymph node metastasis(P =0.047 ,P <0.05).The expression of Ki-67 in Her-2positive group was higher than Her-2 negative group (P= 0.021,P<0.05),and the expression of Ki-67 in ER positive group was lower than that of ER negative group (P = 0.037,P < 0.05).Conclusions The expression of Ki-67 in T1 primary breast cancer indicates the possibility of lymph node metastasis,and its positive correlation with Her-2 positive expression,ER negative expression indicates poor prognosis.

5.Clinical application of anti-carbamylated protein antibody

Rheumatoid arthritis ( RA) was a common autoimmune disease characterized with main clinical manifestations of chronic inflammation in joints.Anti-carbamylated proteins(anti-CarP)antibody, as a promising bio-marker, played a crucial role in the diagnosis of RA patients and the screening of pre-RA patients .In this article , recent progress on the study of anti-CarP antibody and RA related diseases has been reviewed , which was expected to provide new references for the diagnosis and treatment of related clinical diseases .
6.Comparison of conventional and new mesh hydroxyapatite in repair of cleft lip and palate

BACKGROUND:Despite regular mesh hydroxyapatite has certain biocompatibility and bone conductivity, it cannot be absorbed by human body after transplantation, and wil affect the growth and eruption of maxila and fangs when using for filing the injured alveolar cleft. OBJECTIVE: To compare the effect of conventional and new mesh hydroxyapatite composites in repair of alveolar cleft defects. METHODS: Totaly 46 patients with cleft lip and palate were randomly and evenly divided into test and control groups, and al were arranged according to cleft lip and palate sequence treatment. After nasal soft tissue defect repair, new mesh hydroxyapatite composite bone material (hydroxyapatite and bone morphogenetic protein compound). Conventional mesh hydroxyapatite material was adopted in the test and control groups, respectively. Within 6 months after the repair, the vertical distance from the newly formed bone tissue plane to the top of alveolar ridge was observed. The clinical repair effect was comprehensively evaluated from the aspects of complications, alveolar cleft shape and size, calus formation and material absorption. RESULTS AND CONCLUSION: At 2, 4, 8, 12 and 24 weeks, the vertical distance from the newly formed bone tissue plane to the top of alveolar ridge in the test group was higher than that in the control group (P < 0.05). The clinical repairing significant efficiency and total effective rate were al higher than those in the control group (P< 0.05). These results show that new mesh hydroxyapatite composite bone materials in the repair of alveolar cleft defects have good bone osteoinductivity and biocompatibility, rare complications and rapid calus formation.
7.Research progress of application of immunotherapy by chimeric antigen receptor-engineered T cells

The technique of chimeric antigen receptor-engineered T cells (CAR-T) is combined single chain antibody and T-cell activation modification and has specific and durable killing effect on tumors.It had achieved stunning results on clinical trials of B lymphocytic leukemias,B lymphomas and solid tumors.Meanwhile it also had potential risk on the off target effects,cytokine release syndrome,graft versus host disease and so on.This paper briefly reviews the principal of CAR-T cellular mechanism,advantages and clinical applications.
8.Preparation and process optimization of notoginsenoside R1 chitosan nanoparticles

Objective To prepare optimization of notoginsenoside R1 chitosan nanoparticles,to provide a theoretical basis for clinical application of the drug.Methods Notoginsenoside R1 chitosan nanoparticles were prepared,HPLC method was used to detect the content of notoginsenoside R1 chitosan nanoparticles,preparation technology of nanoparticles were optimized by orthogonal experiment,and the optimized preparation technology of nanoparticles was verified.Results HPLC standard curve equation was A =911.49C -1803.4(r =0.999 9),linear range was from 25 to 900 g/mL.The intra day precision were 1.520%,0.884% and 0.969%(n =6),and the inter day precision were 1.591%,1.447% and 1.269%(n =6).The recovery rates of low,medium and high concentra-tions were (98.11 ±1.16)%,(101.27 ±0.59)% and (100.97 ±0.82)%.4 factors of orthogonal experiment:the concentration of chitosan,the mass ratio of drug and carrier,temperature and rotational speed,and 3 levels of each factor were selected.The average particle size,encapsulation efficiency and drug loading were selected as control indexes.The test results were determined by the method of comprehensive weighted scoring.The orthogonal design was designed according to L9(34)orthogonal design.The optimization process was 2% of chitosan concentration,20% of the weight ratio of drug and carrier,35 ℃ of temperature,600 r/min of rotational speed.According to the optimized process,the average particle size was (123.40 ±7.68)nm,the encapsulation efficiency was (58.41 ±1.59)%,and the drug loading amount was (10.46 ±0.53)%.Conclusion The optimized preparation process of notoginsenoside R1 chitosan nanoparticles is simple and easy to operate,the entrapment efficiency and drug loading amount were high. As a new dosage form,it has a good clinical application prospect.
9.The effects of a single IV lidocaine bolus dose on the minimal alveolar concentration of sevoflurane

Objective To evaluate the effects of a single IV lidocaine bolus dose on the minimal alveolar concentration (MAC)of sevoflurane. Methods Patients (n=90), aged 25-65 years whose Anesthesiologists (ASA) classification wasⅠorⅡand underwent elective surgery on trunk under general anesthesia, were randomly divided into 3 groups with 30 cases in each group:high-dose lidocaine group (group H), low-dose lidocaine group (group L) and control group (group C). They were induced by sevoflurane inhalation, and ventilated by LMA (laryngeal mask airway). After a 15 minutes equilibration period with the above sevoflurane concentration , the medication to be studied (2%lidocaine 1.5 mg/kg for group H , 2%lidocaine 0.75 mg/kg for group L, 0.9%saline 5mL for group C) was administered for 3 minutes before the skin incision. The response to skin incision (movement versus no movement) was recorded in the first minute after skin incision. The MAC for sevoflu?rane was determined using the Dixon′s up and down method. Values of mean arterial pressure (MAP), heart rate (HR), and BIS were recorded at 1 minute and 5 minutes after being monitored (average values were noted as T0), immediately before the administration of medication (T1), immediately before the skin incision (T2) and 1 minute after the skin incision(T3). Results MAC in group H (2.00%± 0.17%) was lower than that in group C (2.22%± 0.18%) by approximately 0.22%,and which was lower than that of group L ( 2.21%± 0.14%) by approximately 0.21%(F=7.054,P<0.05). No significant differ?ence in the MAC of sevoflurane was noted between group L and group C. The values of HR, MAP and BIS all decreased at T 2 and increased at T3 in all 3 groups (all P<0.05). No significant difference in HR, MAP or BIS was observed between T0 and T1 in all three groups. The values of HR and BIS were lower in group H than those in group C and group L at T2 and T3. The values of MAP were lower in group L and group H than those in group C at T2 and T3. The value of MAP were lower in group H than that in group L at T2(all P<0.05). Conclusion A singleⅣ1.5 mg/kg lidocaine decreases MAC of sevoflurane, but the decreased amplitude (11%) does not reach expectation.
10.Clinical therapeutic effect of cattle encephalon glycoside and ignotin on ad-junctive treatment of severe hand-foot-and-mouth disease

Objective To explore clinical therapeutic effect of cattle encephalon glycoside and ignotin (EGI)on adjunc-tive therapy of severe hand-foot-and-mouth disease (HFMD).Methods 94 children with severe HFMD in a hospital were selected and divided into two groups according to the parity of patients’admission numbers,46 children in control group were given conventional therapy;48 children in trial group were given EGI adjunctive therapy based on conventional thera-py,therapeutic effect of two groups was compared.Results The total therapeutic effective rate in trial group was signifi-cantly higher than that of control group (95.83% vs 76.09%,χ2 =16.81,P <0.001 ).Neurological function score was not significantly different between two groups before therapy(P >0.05);after therapy,neurological function score in trial group was significantly lower than control group ([4.07±0.13]vs [5.59±0.18],χ2 =18.19,P <0.05).The recovery time of temperature,heart rate,blood pressure,and breathing,as well as length of stay in hospital in trial group were all significantly shorter than control group(P <0.05);incidence of complications in trial group was lower than control group (4.17%[n=2]vs 21.74%[n=10],χ2 =10.08,P <0.001 ).The proportion of turning into critically ill cases in trial group was significantly lower than control group (2.08% [n = 1 ]vs 13.04% [n = 6 ],χ2 = 8.94,P < 0.001 ). Conclusion EGI has remarkable efficacy in adjunctive therapy of severe HFMD,which can effectively improve clinical symptoms,and is worthy to be popularized and applied.
