2.Recent progress in metastasis mechanism of malignant tumors and their treatment strategies

Although great achievement has been made on the diagnosis and treatment of malignant tumors,metastasis of tumor remains to be the major reason for treatment failure and death of their victims.The development of systematic biology and functional genomics give us a deeper understanding of the nature of metastasis at the molecular level.Based on the consensus reached on the metastasis mechanisms of cancer,including the genetic heterogeneity of cancer,pre-metastatic niche,epithelial-mesenchymal transition,anoikis resistance,angiogenesis and lymphangiogenesis,etc,the targeted therapy at molecular level should be emphasized,the treatment should target the tumor stem cells,and RNAi technique and other techniques should be used in clinical anti-metastatic therapy.
3.Change of niclosamide concentration in soil through heaping and the effect of Oncomelania control

Objective To analyze the concentration, distribution and decreasing trend of niclosamide in soil through soil heaping mixed with niclosamide and the effect of Oncomelania control, and to explore the appropriate dosage for the 'heaping' method. Methods The soil samples were collected from six groups in Tezi township, Puge county, Sichuan Province, and the dosages were 16, 8, 4, 2, 1 and 0 g/m~2, respectively. After ultrasonic extraction, centrifugence and concentration, the samples were finally determined by high-performance liquid chromatography (HPLC). The mortality rate of snails was observed after 3 days and 7 days. Results The standard error and range of niclosamide in soil were large. There was no significant difference in concentration of niclosamide between the surface layer and deep layer of soil (P>0.05). After 5 months, niclosamide still could be determined in groups of 4 g/m~2. The mortality rate of snails decreased as the concentration of niclosamide decreased (P<0.05). After 5 months, in the group of 4 g/m~2 dosage, the 3 and 7 days mortality rate of snails were 5.33% and 9.33% in the surface layer, higher than the control group (P<0.05). Conclusions The heaping method is an efficacious measure of controlling snails and its recommended dosage is 4 g/m~2.
4.Expression of herpesvirus-associated ubiquitin specific protease(HAUSP) in breast carcinoma and its clinical significance

Background and purpose: Herpesvirus-associated ubiquitin-specific protease (HAUSP) is a new deubiquitinating enzyme that was recently discovered. It has been demonstrated that HAUSP could deubiquitinate p53 both in vitro and in vivo. These results suggested that HAUSP might act as a tumor suppressor through the stabilization of p53 protein. The aim of this study was to investigate the expression of HAUSP in breast carcinoma, and its association with p53 protein as well as their relationship to prognosis. Methods: The expression of HAUSP mRNA was detected by real-time PCR. HAUSP protein and p53 protein were detected by immunohistochemistry with EnVision system in breast carcinoma tissues and noncancerous tissues. The relationship between their expressions and clinical pathological parameters were analyzed. Results: The expression of HAUSP mRNA was significantly lower in breast cancer tissue than noncancerous tissue (1.85±0.04 vs. 2.74±0.03, P<0.01). The positive rates of HAUSP protein were significantly lower in breast cancer tissue in noncancerous tissue (59.4% vs. 75.0%, P<0.01), and expression of HAUSP protein had no significant correlation with the clinical pathological parameters. There was no significant correlation between HAUSP and p53 protein (P>0.05). The expression of HAUSP mRNA was positively associated with that of HAUSP protein (P<0.01). There was no significant correlation between HAUSP and p53 protein. DFS of patients with both HAUSP positive and p53 negative was significant higher than the controls (P<0.01). Conclusion: Down-regulation of HAUSP protein and HAUSP mRNA in breast carcinoma indicated that HAUSP gene might correlated to tumor carcinogenesis. In addition, the simultaneous evaluation of both HAUSP expression and p53 expression status may be helpful to evaluate the prognosis of breast cancer patients.
5.Optimization of Dosage Regimen of Amoxicillin and Clavulanate Potassium Extended Release Tablets Based on PK/PD Parameters

Objective:To optimize the clinical dosage regimen of amoxicillin and clavulanate potassium extended release ( ER) tablets based on the PK/PD parameters. Methods:Totally 30 healthy subjects ( half male and half female) were randomly divided into three groups, and orally administered the ER tablets respectively under fasting condition, before the meal and after the meal, and the optimal administration time was determined by comparing the pharmacokinetic characteristics. The subjects in the three groups were ad-ministered the ER tablets respectively at low, medium and high dosage, and the optimal dosage and dosing interval were determined based on the PK/PD parameters. Results:Under fasting condition, the AUC of amoxicillin [(32.2 ±15.0) μg·h·ml-1] was sig-nificantly lower than that before the meal [(41.7 ±1.92) μg·h·ml-1] and that after the meal [(42.6 ±17.7) μg·h·ml-1]. In contrast, the AUC of clavulanate acid after the meal [(1.89 ±0.54) μg·h·ml-1] was significantly lower than that under fasting condition [(2.55 ±0.76) μg·h·ml-1] and that before the meal [(2.58 ±0.76) μg·h·ml-1] (P<0.05). Amoxicillin and clavulanic acid displayed linear pharmacokinetics within the range of 1000-4000 mg and 62. 5-250 mg, respectively. After a single o-ral administration of amoxicillin and clavulanate potassium ER tablets at low, medium and high dose, the duration of blood concentra-tion above the minimum inhibitory concentration (MIC, 2. 0 μg·ml-1) (T> MIC) in 12 h was 5. 5, 7 and 10 h, and the percentage was 46%, 58% and 83%, respectively, and T> MIC in 12 h was 4. 5, 6 and 8 h, and the percentage was 38%, 50% and 67%, re-spectively when MIC was 4. 0μg·ml-1 . Conclusion:It is suggested that amoxicillin and clavulanate potassium ER tablets be taken at the start of a standard meal, 2 tablets per time, twice daily, which is sufficient to achieve T> MIC of 40% -50%.
6.Establishment of pancreatic mucinous cystadenocarcinoma cell line MCC1 with stable overexpression of miR-224

Objective: To construct a hsa-microRNA-224(miR-224) lentiviral expression vector and to establish pancreatic mucinous cystadenocarcinoma MCC1 cell line with stable miR-224 over-expression. Methods: Pri-miR-224 gene fragment was designed and amplified by quantitative real-time polymerase chain reaction (qRT-PCR), and then loaded into GV369 lentiviral vectors (GV369-miR224) by gene recombination technology. GV369-miR-224 lentivrial expression vectors were then identified by PCR and DNA sequencing. The GV369-miR-224 vector fluid was then used to infect pancreatic mucinous cystadenocarcinoma MCC1 cell line to establish the MCC1 cell line stably over-expressing miR-224. The transfection efficiency of GV369-NC and GV369-miR-224 was observed under fluorescence microscopy; and the expression levels of miR-224 in MCC1, GV369-miR-224-MCC1 and GV369-NC-MCC1 cell lines were detected by RT-PCR. Results: The GV369-miR-224 lentiviral vectors were successfully constructed. GV369-miR-224-MCC1 and GV369-NC-MCC1 cells all emit green fluorescence under the fluorescence microscope. The expression level of miR-224 in GV369miR-224-MCC1 cell group was significantly higher than that in negative control GV369-miR-224-MCC1 group and blank control MCC1 cell group (23.45±1.94, 1.46±0.1 and 2.11±0.38, P<0.01), however, there was no significant difference between the two control groups (P>0.05). Conclusion: A pancreatic mucinous cystadenocarcinoma MCC1 cell line with stable miR-224 over-expression was successfully established, and this will provide a new cell model for exploring the function and pathogenesis of miR-224 in pancreatic mucinous cystadenocarcinoma.

To promote the development of extracellular vesicles of herbal medicine especially the establishment of standardization, led by the National Expert Committee on Research and Application of Chinese Herbal Vesicles, research experts in the field of herbal medicine and extracellular vesicles were invited nationwide with the support of the Expert Committee on Research and Application of Chinese Herbal Vesicles, Professional Committee on Extracellular Vesicle Research and Application, Chinese Society of Research Hospitals and the Guangdong Engineering Research Center of Chinese Herbal Vesicles. Based on the collation of relevant literature, we have adopted the Delphi method, the consensus meeting method combined with the nominal group method to form a discussion draft of "Consensus statement on research and application of Chinese herbal medicine derived extracellular vesicles-like particles (2023)". The first draft was discussed in online and offline meetings on October 12, 14, November 2, 2022 and April and May 2023 on the current status of research, nomenclature, isolation methods, quality standards and research applications of extracellular vesicles of Chinese herbal medicines, and 13 consensus opinions were finally formed. At the Third Academic Conference on Research and Application of Chinese Herbal Vesicles, held on May 26, 2023, Kewei Zhao, convenor of the consensus, presented and read the consensus to the experts of the Expert Committee on Research and Application of Chinese Herbal Vesicles. The consensus highlights the characteristics and advantages of Chinese medicine, inherits the essence, and keeps the righteousness and innovation, aiming to provide a reference for colleagues engaged in research and application of Chinese herbal vesicles at home and abroad, decode the mystery behind Chinese herbal vesicles together, establish a safe, effective and controllable accurate Chinese herbal vesicle prevention and treatment system, and build a bridge for Chinese medicine to the world.
