Objective To explore the effect of the association of low molecular heparin and Galectin-3 on the cell migration and cell proliferation of vascular endothelial cell from mesenchymal stem cells. Methods Depending on the administration, this study was divided into four groups: low molecular weight heparin group, adding 20 mg/L low molecular weight heparin into the cells;Galectin-3 group, adding 5 mg/L Galectin-3 into the cells;combination group, adding 20 mg/L low molecular weight heparin and 5 μg/ml of Galectin-3 into the cells;control group, equal volume of PBS buffer into the cells. The proliferation of vascular endothelial cell was to be detected by EdU incorporation, the cell growth cycle of vascular endothelial cell was to be detected by flow cytometry, and the cell migration of vascular endothelial cell was to be detected by scrath test. Then to investigate the effect of different conditions on the migration and proliferation of vascular endothelial cell. Results The OD490 of MWH group, Galectin-3 group, combined group and control group were (0.285±0.018), (0.297±0.041), (0.351±0.016), and (0.233±0.005) respectively, which indicates that the combined group could increase the cell proliferation significantly(P<0.05).Cultured for 24 hours, the cell migration rate was(42.02±7.62), (45.82±3.96), (68.53±11.22),and (34.21±3.99), suggesting that combined group have the largest cell migration(P<0.05).Conclusion The association of low molecular heparin and Galectin-3 could improve the cell migration and cell proliferation of vascular endothelial cell from esenchymal stemcells significantly.

Objective To investigate the relationship between HCV genotype and its level and the response to interferon (IFN).Methods HCV genotypes were detected by type-specific primes modified PCR assay.HCV-RNA levels were determined by branched DNA(bDNA) singnal PCR amplification fluorescence assay. The patients with chronic hepatitis C virus infection treated with IFN ? were divided into different groups according to genotypes or levels of HCV.Result Patients with pre-treatment HCV levels10 9 eq?L -1 showed partial responses (28 6%) or no responsces (28 6%).Stasistical analysis showed significant difference (P

Objective To construct a qseC-deleted mutant strain of E.coli by Red recombination and to study the effect of qseC gene on biofilm formation in the mutants. Methods The chloramphenicolresistant gene flanked by homologues of target genes was amplified by PCR and electro-transformed into E.coli MC1000.When induced by L-arabinose,the plasmid pKD46 could express three recombinant proteins of λ-prophage,which led to the replacement of target gene(qseC) with chloramphenicol-resistant gene. Then the chloramphenicol-resistant gene was eliminated by FLP-promoted recombination events. The biofilm formation of wild-type and mutant strain was detected by crystal violet staining. Results The qseC-deleted mutant of E.coli was confirmed by various PCR and DNA sequencing.Gene qseC was completely deleted. There was no significant difference in growth ability between the qseC mutant strain and the wild-type strain MC1000.The biofilm formation of wild-type and mutant strain was quantified by crystal violet staining. The absorbance determined with a plate reader at 570 nm was 1.00±0.15 and 0.47±0.10 respectively. Conclusion The qseC-deleted mutant of E.coli was constructed successfully. And the qseC gene plays an important role in regulation of biofilm formation in E.coli.

Objective To investigate the effect of a step-wise diversified teaching mode on cardiac electrophysiology (EP) education. Methods 64 young doctors (male:36, female:28) who studied in EP sub-specialty were enrolled. The atrioventricular nodal reentrant tachycardia (AVNRT) and atrioventricular reentrant tachycardia (AVRT) were selected as teaching content. All the doctors were randomly divided into traditional teaching group (TT) and step-wise diversified teaching group (DT) according to cross-over design protocol. In TT group, only teaching and answering the questions by teachers were given to students, for the DT group, in different stages of teaching, multiple teach-ing strategy were combined to complete teaching. T test and Chi square test were used to compare the effect of two teaching methods. Results There was no difference of basic features and time of self-studying between the two groups. In examinations, the students in DT group got higher score than TT group[DT:(92.3±9.8) and (93.1±7.8),TT:(88.3±8.6) and (87.1±10.0),P<0.05], and had better perfor-mance of learning interest, initiative, ability of finding and resolving problems, teamwork spirit and so on. Conclusion The step-wise diversified teaching mode contributes improving the quality of instruction of cardiac EP, moreover, arousing learning interest and initiative, enhancing ability of find-ing and resolving problems, teamwork spirit and so on, which is worthy of wide-popularizing in cardiac EP training.

Objective To explore the differences in disease location, pathological feature, disease severity, extraintestinal manifestations and drug treatment between inflammatory bowel disease (IBD) patients with comorbid autoimmune disease (AD) and simple IBD patients. Methods From January 2009 to December 2014,the clinical data of 54 IBD patients with comorbid AD and at the same period 74 simple hospitalized IBD patients were retrospectively analyzed.According to IBD type and whether combined with AD,patients were divided into Crohn's disease (CD)+AD group (n=16),CD group (n=26),ulcerative colitis (UC)+AD group (n=38) and UC group (n=48).Chi square test was performed to compare the differences in disease severity, location, extraintestinal manifestations and drug treatment between IBD patients with and without AD.Results There was no statistically significant difference in location among four groups (all P>0.05).The most common concomitant AD of IBD was rheumatoid arthritis (20.4%,11/54) and ankylosing spondylitis (13.0%,7/54).The proportion of mild active patients of CD+ AD group was lower than that of CD group (2/16 vs 53.8% (14/26),x2 =7.180,P=0.007),while the proportion of severe active patients was significantly higher that of CD group (6/16 vs 0,x2 =8.519,P=0.004).There was no statistically significant difference in moderate active patients between the two groups (P=0.808).Main type of patients of UC+ AD group (76.3 %,29/38) and UC group (68.8 %,33/48) were moderate active patients. There was no statistically significant difference in disease stage and location (all P>0.05).The incidence of extraintestinal manifestations of IBD+AD group (55.6 %,30/ 54) was significantly higher than that of IBD group (9.5 %,7/74,x2 =32.279,P<0.01),and the main manifestation was arthritis (37.0% (20/54) vs 5.4% (4/74),x2=20.504,P<0.01).The rate of glucocorticoid and immunosuppressant application in IBD+AD group was higher than that of IBD group (40.7% (22/54) vs 17.6%(13/74),x2 =8.438,P=0.004;20.4%(11/54) vs 0,x2=14.000,P< 0.01).Conclusions The condition of patients with IBD and comorbid AD is more severe, and the incidence of extraintestinal manifestations is higher.Early treated with glucocorticoid and immunosuppressant could effectively achieve remission.
