AIM: To elucidate whether the mechanism that L-arginine can relieve hypoxia pulmonary hypertension involves inhibition of the activity of protein kinase C(PKC). METHODS: Twenty-one male Wistar rats were randomly divided into NS control, hypoxia and L-arginine(500 mg?kg -1 ?d -1 , ip) treatment groups. After two-weeks treatment, mean pulmonary artery pressure (mPAP), mean systematic artery pressure (mSAP) and the ratio of the weight of right ventricle to that of left ventricle plus septum were measured, then two pulmonary arterial rings were isolated to be exposed to PDBu(a specific activator of PKC ) and observed: (1) The maximal response (P 1) to 500 nmol/L PDBu, the time required to achieve a half-maximal response to 500 nmol/L PDBu (t 1/2 ), the time during which the maximal response to 500 nmol/L PDBu maintained (T) and the isometric responses at different times (2, 4, 8, 12, 20, 40, 60, 80 min). The isometric response was represented as the percentage of the maximal response (P 0) of the same arterial ring to 5?mol/L phenylephrine hydrochloride (P 0%). (2) Dose-response curve in response to PDBu (10-11 000 nmol/L) and the dose producing a half-maximal response in the curve (EC 50 ). RESULTS: mPAP, RV/(LV+S), P 1, T and the isometric responses at 2, 4, 8, 20 min of NS control and L-arginine treatment groups were lower than those of hypoxic group respectively (P

AIM: To explore the effects of levcromakalim(Lev) on pulmonary arterial endothelial cells (PAEC) and smooth muscle cells (PASMC) exposed to hypoxia and the mechanisms involved. METHODS: The effects of Lev on i, and expression of PKC ?, eNOS, iNOS and PDGF-B mRNA and protein levels were observed. The nitrite (NO- 2) and entothelin-1(ET-1) concentrations in supernatant in cultured PAEC and PASMC were measured. The proliferation and apoptosis of PASMC was also detected. RESULTS: When PASMC were exposed to hypoxia, Lev reduced concentration of ET-1 in cultured cell supernatant, lowed the expression of PKC ?, iNOS and PDGF-B both at mRNA and protein levels, decreased i concentration, increased proliferation and promoted the apoptosis in PASMC. However, in the presence of Lev, the i concentration was not changed in the hypoxic PAEC. The NO- 2 concentration in cultured cell supernant and expression of eNOS at mRNA and protein levels in hypoxic PASMC and PAEC were also unchanged. The downregulated ET-1 activity in PASMC and PAEC and proliferation in PASMC involved in the inhibition of PKC ? signaling pathway. CONCLUSIONS: Lev reduce some disadvatage effect of hypoxia on PASMC and PAEC. The mechanism of Lev action may partly involve in the downregulation of PKC ? signaling functions.

Objective To investigate transplantation of autologous peripheral blood stem cells(PBSC)for therapeutic treatment of ischemic arterial disease of the lower extremity.Methods Mobilized PBSC was directly injected into the ischemic hindlimb muscles in rabbit models.Two and four weeks after the transplantation of autologous PBSC selective angiography was carried out,and the capillary density was determined histochemically in the specimens using CD31 mAb.Results In angiography,collateral arteries were significantly increased in ischemic hindlimbs.The capillary density was significantly higher in treatment group.Conclusion Transplantation of autologous PBSC is a simple and effective therapy for the ischemia hindlimb.

Objective The aim of this study was to evaluate the in vitro activity of Ciprofloxacin(CIP)or Amikacin(AK)combined with four clinically-used antipseudomonas ?-lactams against multidrug-resistant Pseudomonas aeruginosa strains.Methods Determining the MICs of six antibiotics to 35 clinically isolated Pseudomonas aeruginosa strains which have different resistance patterns by 2-fold broth dilution method;determining the in vitro activity of Ciprofloxacin or Amikacin combined with Ceftazidime(CAZ),Imipenem(IMP),Meropenem(MEM)and Piperacillin/Tazobactam(TZP)by broth checkerboard method.Results The combinations of Ciprofloxacin or Amikacin with four ?-lactams have mainly synergistic or additive effect,the synergistic rate of AK-group(AK+CAZ:71.43%;AK+IMP:42.86%;AK+MEM:54.29%;AK+TZP:85.71%)was higher than that of CIP-group(CIP+CAZ:20%;CIP+IMP:25.71%;CIP+MEM:14.29%;CIP+TZP:31.43%).The combination of CIP+TZP has the highest synergistic rate in CIP-group;the combination of AK+TZP has the highest synergistic rate in AK-group.None of the combinations had an antagonistic effect.Conclusion the combination of ciprofloxacin or amikacin with ?-lactams have good in vitro activity to multidrug-resistant Pseudomonas aeruginosa strains.

Objective To investigate and analyze the state of injury and treatment for the patients with maxillofacial injuries in Wenchuan earthquake, and search for more effective methods for treating maxillofacial injury. Methods One hundred and sixty-five patients wounded in Wenchuan earthquake were admitted to authors' hospital, and they received specialized diagnosis and treatment. In this paper, the emphasis was put on those patients with severe ear-nose-throat and maxillofacial injuries. Based on the analysis of the state of injuries, the patients were treated with debridement and suture, reshaping and suture, reduction and fixation of fracture, or local or systematic anti-infection therapy. All the treated patients were followed up 2 weeks after treatment. Results Thirty-three of the 165 admitted patients suffered from various degrees of maxillofacial injury (20.0%), including 10 cases of ear injuries (6.1%), 8 cases of nasal trauma (4.8%) and 2 cases of lip laceration (1.2%). All the wounds healed by primary intention after the treatment with no disfiguration. 8 cases of nasal sinus and zygomatic fractures (4.8%) and 5 cases of mandibular fracture (3.0%) recovered nicely after the treatment. No trauma was found in throat, neck, trachea and esophagus. 6 patients (3.6%) were complicated with infection after injury (onset of infection outside of hospital). There was no death. Conclusion Patients with maxillofacial injury should receive early and appropriate treatment. Besides restoration of function, it is also important to restore the physiognomy

Purpose To investigate the value of 11C-CFT PET/CT dopamine transporter (DAT) imaging in differential diagnosis of multiple system atrophy (MSA) and Parkinson's disease (PD).Materials and Methods The 11C-CFT PET/CT images of clinically confirmed MSA patients (21 cases),PD patients (24 cases) and healthy adults (10 cases as normal control) were analyzed retrospectively.The volume of interest (VOI) were drawn manually,and the DAT binding indexes and asymmetry indexes of different regions of striatum,including caudate and putamen,were calculated.The differences of DAT binding indexes and asymmetry indexes among the above three groups were analyzed.Results Compared with the normal control group,the striatal DAT binding indexes of MSA group or PD group were significantly reduced (P＜0.05).There were no significant differences in DAT binding indexes between the MSA group and the PD group (P＞0.05).Compared with the normal control group,the DAT binding asymmetry indexes were significantly increased in the PD group (P＜0.05),but the indexes showed no significant differences in MSA group (P＞0.05).The DAT asymmetry indexes of caudate and putamen in the PD group were higher than those in the MSA group (P＜0.05).Conclusion 11C-CFT PET/CT imaging can detect the degeneration of dopaminergic neurons in striatum.The number of striatal dopamine transporters declines in both MSA and PD patients,but the asymmetry of striatal dapamine transporter in PD patients is higher than that in MSA patients.11C-CFT PET/CT can differentiate MSA and PD.

Objective To evaluate the relieving effects of vanillin sinffing on depression-like behaviors in depressed rats and to explore the possible underlying mechanism.Methods Depression animal model established by chronic unpredictable medium intensity stress combined with isolation and destroy the olfactory bulb.The depressed rats were divided randomly into vanillin inhalation group,fluoxetine hydrochloride group,depression model group,olfactory bulbectomy with the vanillin inhalation treatment group and sham-operated group.Nervous behavioral changes had been observed at different time after the administration of 5 weeks.The concentration of brain derived neurotrophic factor(BDNF) in the brain homogenate and the positive expression of BDNF in hippocampus had been also measured.Results Two weeks after the intervention,the immobility time of vanillin group((12.78 ±7.50) s) was lower than that of the model group((57.33±32.16) s) (P＜0.05).The consumption of saccharose in vanillin group((52.88±25.18)g) was higher than that of model group((37.40±19.33) g) (P＜0.05).BDNF of the brain homogenate in vanillin group (0.54±0.13) was significantly increased compared with model group (0.36± 0.06) (P＜0.01).When compared with the OBX group (0.40±0.06),similar result was obtained.Immunohistochemistry and the average density of image analysis revealed that the expression of BNDF of hippocampal CA3 in vanillin group (0.40±0.03)was significantly increased compared with model group (0.25±0.04) and OBX group (0.28±0.03) (P＜0.01).Conclusion Vanillin inhalation significantly relieves depression-like behaviors in depression rats.The possible mechanism may increase hippocampal neurogenesis by raising brain derived neurotrophic factor in brain.

Purpose To investigate the expression of CD147,MMP-9 and VEGF and their relationships in lung adenocarcinoma.Methods The expression of CD147,MMP-9 and VEGF proteins was examined in 90 lung adenocarcinoma tissues by SP immunohistochemical method.The positive unit (PU) of CD147,MMP-9 and VEGF was assessed quantitatively using Imagepro Plus image analysis software.Results Positive intensity of CD147,MMP-9 and VEGF in lung adenocarcinoma was significantly higher than that in non-neoplastic internal controls and benign control tissues (P ＜ 0.001).The positive rates of CD147,MMP-9 and VEGF were 88.89％ (80/90),84.44％ (76/90),and 80％ (72/90),respectively.The expression of CD147,MMP-9 and VEGF was positively correlated with the degree of histologic differentiation,lymph node metastasis status and TNM stage (P ＜ 0.001).CD147 was correlated positively with MMP-9 and VEGF expression (rp =0.992,rp =0.984,P ＜ 0.001).Conclusion CD147,MMP-9 and VEGF may play an important role in the pathogenesis and progression of lung adenocarcinoma;CD147 may induce tumor invasion and metastasis by stimulating production of MMP-9 and VEGF,which is expected to be a potential promising target for immunotherapy.

Aim To investigate the effects of cholecys-tokinin octapeptide(CCK-8)on the withdrawal symp-toms of morphinistic rats by the observation of praxiology,and change of expression of proto-oncogene c-jun in caudate nucleus(Cd) after CCK-8 injected by immunohistochemistry.Methods Animal praxiology evaluation and immunohistochemistry were used.The influence of CCK-8(10 mg?L-1,1 ?l) on the withdrawal symptoms and the expression of c-jun of morphinistic rats was observed.Results Morphinistic rats had obvious praxiological changes in the withdrawal symptoms compared with normal saline rats;the scores of withdrawal symptoms decreased most obviously at withdrawal 40 h after Cd injection of CCK-8;the expression of c-jun protein in Cd decreased in morphinistic rats,but increased after Cd injection of CCK-8.Conclusions Models of morphinomania rats were successfully built.The Cd injection of CCK-8 could decrease the withdrawal symptoms of morphinomania rats.The CCK-8 could increase the expression of c-jun protein in Cd.These findings suggested that CCK-8 could regulate the production of withdrawal symptoms and the expression of c-jun protein in Cd of morphinomania rats.
