3.P53 overexpression in different subcellular location in non small cell lung cancer
Chinese Journal of Pathophysiology 1986;0(04):-
AIM: Nuclear P53 localization was considered to be proof of mutation, cytoplasmic staining has also been signalled. In this study, special attention has paid to protein subcelluler localization. A different functional meaning may attributed to tumor biological characteristic and evolution. METHODS: Immunohistochemical approach and flow cytometry were used to explored the relationship between P53 overexpression pattern (nuclear and cytoplas- mic) and G1/S checkpoint function of P53 protein in cell cycle. RESULTS: Two patterns of P53 overexpression are related to disease stage significantly. S - phase fraction (SPF) was higher and G0 - G1 phase fraction was lower in P53 nuclear pattern than cytoplasmic (SPF, 42.42? 5 .45/14. 12? 4 .42, G0 - G1: 53. 04 ? 6. 14/75. 56 ? 4. 46, respectively P
4.Diabetic demyelinating neuropathy:a report of five cases
Journal of Chongqing Medical University 1986;0(02):-
Objective:To report the clinical characteristics and diagnoses of diabetic demyelinating neuropathy. Methods:Five cases were systematically studied based on the clinical observations and data from aided examination. Results:All the five patients with diabetes mellitus were more than 50 years old.The clinical manifestations included predominantly motor symmetric weakness, proximal and distal, associated with areflexia and a relapsing or a chronic progressive course. The five cases were indistinguishable from idiopathic chronic inflammatory demyelinating polyneuropathy(I-CIDP) in clinical picture and electrophysiology. They responded to glucocorticosteroid treatment, plasma exchange and intravenous immunoglobulin. Conclusion:Diabetic demyelinating polyneuropathy is one of several types of diabetic peripheral neuropathies.
5.Study of biological properties of bone marrow stromal cells in vitro in wistar rats
Journal of Chongqing Medical University 2007;0(09):-
Objective:To observe the biological properties of bone marrow stromal cells(MSCs) in vitro in Wistar rats for inducing MSCs to differentiate into neurons and transplantating MSCs to treat cerebral ischemic stroke. Methods:The Wistar rats for 4 weeks old were sacrified, the bone marrow separated and the MSCs cultured, and the biological properties of the cells observed by phasecontrast microscope.Cell surface markers were evaluated by indirect immunofluorescence cycochemistry staining in primary and different generation cells.Results:95.18% and 94.36% of the third generation cells showed positivity to CD54 and CD44(MSCs surface marker). The percentage of hematopoietic lineage marker CD14 decreased obviously from 23.19% in primal culture of the cells to 2.37% in the third generation. Conclusion:The third generation cells can be used as the seed-cells for study of cells induce and differentiation and the resoure of cells transplantation.

Objective:To study the identification and cultivation of neural stem cells in neonatal rats in vitro. Methods:Cell culture in serum-free medium, indirect immunofluorescence cytochemistry were used. Results:The neural stem cells isolated in newborn rats had the potential of forming clones and serial passage, expressing neuroepithelial stem cell protein(nestin)and differentiating into neurons and astrocytes and oligodendrocytes. Conclusion:Neural stem cells maintain their undifferentiated features and have the capacity of self-renewal,proliferation and pluripotentiality.
7.Reassessment of defibrase in treatment of cerebral infarction
Journal of Chongqing Medical University 1986;0(04):-
Objective:To assess the safety and efficacy of defibrase in the treatment of acute cerebral infarction by a randomized,double-blind,and placebo-controlled clinical trial.Methods:65 cases with acute infarction were randomly allocated to receive either an initial intravenous infusion of defibrase 10U or placebo 250ml of normal saline within 24 hours of stroke onset.Subsequent infusions of defibrase 5U or placebo were given on the third and the fifth day respectively. The end points included Clinically Neurological Deficits Scale of Stroke,Barthel Index, fatality rate of stroke in 3 months and 12 months, adverse reaction and the level of plasma fibrinogen(FIB).Results:(1)The level of plasma FIB in defibrase group was remarkably declined after treatment whereas the bleeding event and other adverse reaction were not increased as compared with the control group.(2)There were no statistically significant differences at Clinically Neurological Deficits Scale of Stroke at 2 weeks,Barthel Index score and fatality rate at 3 months between two groups.(3)The recurring rate of stroke in control group was higher than that in defibrase group at 12 months(P
8.Construction and identification of vectors expressing short hairpin RNA targeting mouse STAT4 and STAT6 gene
Journal of Chongqing Medical University 1987;0(01):-
Objective: To construct short-hairpin RNA(shRNA)expressing vectors aimed at mouse signal transducers and activators of transcription-4/6(STAT4/STAT6). Methods: Target sequences in the middle of STAT4 and STAT6 gene were selected, and then the complementary 65 mer oligonucleotides of each sequence were synthesized. After annealed, the inserts were ligated into the linearized pGenesil-3 plasmid. The plasmids were transformed into DH5? bacteria to amplify and then purified. GAPDH plasmid was constructed as a positive control and targeting none gene was served as a negative control. These purified plasmids were identified by gel electrophoresis after being cut with restriction enzyme and sequencing. Results: The results of gel electrophoresis and sequencing showed that the plasmids were identical with the positive control, and the sequences were identical with what we have inserted in. Conclusion: The vectors which can express siRNA hairpin aimed at STAT4/STAT6 gene have been constructed successfully and it lays the groundwork for the further study.
