1.Interventional Therapy of Ischemic Necrosis of Femoral Head

Objective To study the curative effect of ischemic necrosis of femoral head by interventional thrombolysis.Methods By applying Seldinger's technique,the catheter was guided via arteria femoralis with super-selection into circumflexa femoris medialis,lataralis,and acetabuli to performed the interventional thrombolysis in 50 cases(60sides)with necrosis of femoral head.Results All patients were followed-up for 6~48 months,combined with angiography,the clinical symptoms and the change of bone.The curative effect was evaluated.The ratio of excellent and better effect was 91.4%,the ratio of the improvement of angiography was 87%.the angiography showed that after treatment,the supply artery of femoral head was increased in number;the stained area of femoral head was extended,the capillaries were incereased and the reflux of vein was remarkably improved.The ratio of the improvement of clinical symptom was 96.6%.90% of the necrotic area of femoral head showed hyperplasia,sclerosis,and reduction of cystic changes.Conclusion The interventional therapy by leading to target vascular with thrombolysis in treating necrosis of femoral head has remarkable therapeutic effect.
2.Efficacy observation of botulinum toxin type A in elderly patients with primary intractable trigeminal neuralgia

Objective To investigate the therapeutic effects of botulinum toxin type A (BTX-A) in elderly patients with primary intractable trigeminal neuralgia.Methods 27 elderly patients with primary intractable trigeminal neuralgia were treated with BTX-A local multiple point injection.The efficacy was assessed by visual analog scores (VAS) before and 1 week,2 weeks,1 month,3 months and 6 months after the treatment.Results VAS scores was (9.2±1.1),(5.8±3.0),(3.6± 2.3),(2.3±2.3),(3.2±2.9) and (4.6±3.2) before and 1 week,2 weeks,1 month,3 months and 6 months after BTX-A treatment respectively.VAS score was gradually decreased,reached the lowest at 1 month after BTX-A injection,and then was gradually increased.There were significant differences in VAS scores between between pre-and post-treatment (P<0.05).The efficiency was 37.0%,85.2%,92.6%,70.4% and 59.3% at 1 week,2 weeks,1 month,3 months and 6 months after the treatment respectively.There were significant differences in efficacy between different time points after the treatment (all P<0.05).3 patients had the transient numbness of mouth askew and incomplete eyelid closure and recovered spontaneously after 4-8 weeks.No severe adverse effects were found in the other 27 patients.Conclusions BTX-A is safe and effective in the treatment of primary intractable trigeminal neuralgia in elderly patients.
3.Analysis of the factors affecting the accuracy of detection for single base alterations by oligonucleotide microarray.

The oligonucleotide microarray, a high-throughput polymorphism detection technology, holds great promise for the characterization of complex genetic variance. To achieve greater sensitivity and specificity for it to be an effective platform technology we present results and discuss some of the factors influencing signal intensities and single-mismatch discrimination in array-based mutation/SNP detection. Probes with a series of concentrations were spotted onto the slide in order to find the optimal concentration with the identifiable satisfying signals and the stable ratios between matched and mismatched probes. It was found that under our experimental conditions, when the initial probe concentration is higher than the maximum immobilization capability of the slide (7.5 micrometer), the hybridization signal will be saturated and the ratio between matched and mismatched probes will be more stable than at a lower probe concentration. Considering the cost of probes and the systematic stability, a constant spotting concentration of 10 micrometer was selected. The stability of different types of mismatched oligo-DNA duplexes on the glass surface was also confirmed. The results show that the order of stability of mismatched oligo-DNA duplexes on a glass surface is in general agreement with previous reports conducted using liquid and polyacrylamide gel pads. This suggests that the influence of the mismatched base pair on the stability of the duplex in a solid hybridization system is similar to that in the solution hybridization environment.
