After testing the resistance of Streptomycin to the strain NS-41-80 which is the Meilingmycin producer- Streptomyces nanchangensis , a lot of streptomycin-resistant ( str ) mutants were screened after the spores regenerated on the lethal media when they were treated with 4 different dosage of super-mutagent EMS. We have abtained the high-yield strain 80-5. 11-221 from these str mutants which only produces Meilingmycin and no Nanchangmycin in the rotation-flask experiments. The productivity of 80-5. 11-221 is 1521?g/mL 77. 9% higher than CK's 855?g/mL. After the shake flask fermentation experiment of 80-5. 11-221 for six generations, the productivity of F 2 and F 3 is stable and the productivity of F 4?F 5?F 6 decreases hastily with the increment of generations. It was showed that the chemical-resistance mutation of the strains had closely relation with yield mutation and the EMS dosage of the yield mutation was higher than the resistance-chemical mutation's through the statistical analysis between the dosage of mutation and the chemical-resistance mutational frequency?the variate capacity of the mutants' yield. A model of the resistance-chemical mutational labelling rational selection of the strain producing Meilingmycin was established.

Microbiology 1992;0(05):-
After testing the resistance of streptomycin to NS-41-80 Streptomyces Nanchangensis NS-41-80, the spores treated with 4 different dosage of super-mutagent EMS were regenerated on the lethal media, thus 3,000 streptomycin-resistant ( str ) str mutants were obtained. Then 202 spore mutants were screened through the pre-screening experiments and their yields are beyond 20% higher than the original strains respectively. In the re-screening experiments 48, 7 and 1 mutants were obtained whose yields are 100%, 200% and 300% higher than the original strains respectively and take the precentage of 23.76% and 1.60%, 3.46% and 0.23% and 0.5% and 0.03% of the number of the pre-screened strains and the rescreened strains respectively. Then 5 mutants were fermented whose yields are above 240% higher and the original strain through the continuous 3 batches fermentation, the yields are 57%~96.4% more than the original strains. Among them the yield of 80-5.3-165 is upwards of 6,000?g/mL and its norm is 5,855?g/mL. And a model of the streptomycin-resistance mutational screening of the strain producing Nanchangmycin was established.

The study was purposed to explore the effect and mechanisms of decitabine and/or Trichostatin A (TSA) on SKM-1 cells in vitro. The effect of decitabine and/or TSA on proliferation of SKM-1cells was analyzed with trypan blue exclusion; the differentiation of SKM-1 cells was detected by nitro-blue tetrazolium (NBT) reduction and flow cytometry; the apoptosis of cells was measured by Annexin V-FITC; the mRNA expression of Fas, survivin and P15(INK4B) in cells treated with decitabine and/or TSA was evaluated by RT-PCR. The results showed that decitabine and/or TSA were capable of inhibiting SKM-1 cell growth and promoting cell differentiation; they stimulated the expression of CD14 and CD11b and inhibited HLA-DR expression; meanwhile and decitabine or/and TSA could induce cell apoptosis, up-regulate mRNA expression of Fas and P15(INK4B), and down-regulate survivin mRNA expression. It is concluded that decitabine can induce apoptosis/differentiation of SKM-1 cells, whose mechanisms may related to the expression of Fas, survivin and P15(INK4B). Decitabine has the synergistic effect with TSA.

Chinese Journal of Experimental Traditional Medical Formulae 2019;25(9):25-30
Objective: To explore the effect of modified Si Junzitang (MSJZT) drug serum on the expression of apoptosis-related molecules of gastric cancer cell SGC-7901 and further its anti-tumor mechanism.Method: A total of 40 SD rats were randomly divided into four groups:low-dose,middle-dose,high-dose MSJZT (0.213,0.426,0.853 g·kg-1) groups and normal group (n=10).The treatment groups were administrated through gastric perfusion, and the normal group was given the equivalent volume of normal saline for 10 days.1.5 h after the last treatment, chloral hydrate peritoneal anesthesia was performed, blood was collected from heart, and different doses of serum were separated to prepare drug-containing serum of low-dose,middle-dose,high-dose MSJZT groups,in order to incubate SGC-7901 gastric cancer cell.Early and late apoptosis rates were detected with flow cytometry.Afterwards, the tumor suppressor gene p53,c-nucleoprotein gene (c-Myc),cysteine-aspartic acid protease-3(Caspase-3),B-cell lymphoma-2(Bcl-2) mRNA expressions were confirmed by fluorescence quantitative polymerase chain reaction (Real-time PCR).The protein expressions of p53,c-Myc,Caspase-3,Bcl-2 were detected by immunofluorescence.Result: Compared with the normal group, the high-dose MSJZT group could obviously increase the apoptosis rate to 22.58%(P<0.01).The results of Real-time PCR showed that the middle-dose MSJZT group significantly promoted the mRNA expression of Caspase-3(P<0.01),the medium-dose group significantly increased the mRNA expressions of p53 and c-Myc (P<0.01),and the high-dose group significantly inhibited the mRNA expression of Bcl-2(P<0.01).The immunofluorescence results showed that the high-dose MSJZT group could significantly increase the protein expressions of c-Myc,Caspase-3 and p53(P<0.01),while the protein expression of Bcl-2 was significantly reduced (P<0.05).Conclusion: MSJZT drug serum could exert an anti-tumor effect by inhibiting the expression of the anti-apoptotic protein Bcl-2,and promoting the expressions of pro-apoptotic-related molecules p53,c-Myc,Caspase-3.
