2.Clinical analysis of Level of glutathione redox-state in patients with acute coronary syndrome

Objective To study the changes and clinical significance of glutathione redox-state in patients with coronary heart disease (CHD). Methods Based on both with or without atherogenic risk factors and the result of coronary arteriography, 118 patients were calssified into three groups: control group( n =30) ,risk group(n =28) and CHD group (n =60). Venous blood sample was obtained, then the plasma reduced glutathione ( GSH) and oxidized glutathione (GSSC) were measured by using glutathione reductase recycling method. Redox potential(EhCSH/GSSG) was calculated using Nernst equation. According to clinical situation,CHD group was divided into two subgroups:un-stable angina pectoris( UAP) group(n = 32) and acute myocardial infarction (AMI) group (n =28). Results From control group to risk group,then to CHD group,GSH gradually decreased,and there was significant difference between CHD group and control group (P < 0.05 ) , but GSSG and EhGSH/GSSG gradually increased, there was significant difference between CHD group and risk group and control group (P < 0. 05). There was no significant difference be-tween the UAP subgroup and AMI subgroup in GSH,GSSG and EhGSH/GSSG(P >0.05). Conclusions Body's an-tioxidant capacity decreases and the oxidative stress exists in artherosclerosis, and furthermore there is a certain degree of relevance between oxidative stress and hyperlipoidemia,all these show that improving this situation may decelerate or prevent the formation and progress of artherosclerosis.
3.Comparison of the curative effect between two methods of helicobacter pylori treatment for peptic ulcer evaluation

Objective To compare the clinical curative effect between sequential therapy and triple therapy for helicobacter pylori positive peptic ulcer.Methods 92 patients with peptic dcer were randomly divided into the observation group and the control group,46 cases in each group.The observation group was treated with sequential therapy,control group with triple therapy.The two groups were observed and compared in helicobacter pylori eradication rate,ulcer healing and relief of symptoms.Results After 4 weeks treatment,the observation group Hp clearance rate was 93.5% (43/46),ulcer healing rate 91.3% (42/46),the control group were 73.9% (34/46),78.3%(36/46),two groups had significant differences(x2 =2.714,3.028,all P < 0.05) ;observation group after treatment of symptomatic integral average (0.94 ± 0.21),and the control group (0.92 ± 0.17) divided the difference was not statistically significant (t =0.483,P > 0.05).Conclusion The sequential therapy on helicobacter pylori positive peptic ulcer curative effect is remarkable,and can be more effective in the eradication of helicobacter pylori infection,promote ulcer healing.
4.High concentration IL - 2 induces the expression of IL - 2 receptor chian on resting PBL

Using ~(125)I-labeled anti-IL-2R_? McAb, we investigated the effect of rIL-2 R_? expression . In addition to increasing the IL-2 R_? expression the actived-lymphoblasts, we found that the higher concentration of IL-2 also directly induced the IL-2 R_? expression on the resting PBL, but the peak was at the 5th day. After treated by a higher concentration of IL-2, there was a positive interrelation of time courses between the incorporation of ~3H-TdR and/or ~3H-UR and IL-2 R_? level on resting PBL. This result indicated that it was by means of inducing IL-2 R_? expression that the higher concentration IL-2 caused the proliferation of the resting PBL.
5.Purification and characterization of soluble interleukin 2 receptor

By using 5 G 1-sepharose 4 B immune affinity column, we recovered approximately 160 ?g purified and soluble IL-2 receptor (sIL-2 R ) from 500ml PHA-stimulated human PBL supernatant. Only one band was showed in SDS-PAGE, its molecular weight was about 44 Kd. The purified sIL-2 R, which blocked the binding of anti-IL-2 R monoclonal antibody with IL- 2 R~+ cell, inhibited (at the higher doses) but enhanced (at the lower doses) the growth of IL-2 dependent cells.
6.STUDY OF BEAT TO BEAT SURFACE RECORDING OF HIS BUNDLE ACTIVITY WITHOUT AVERAGING IN MAN

Objective To investigate the application of open beat to beat techniques recording His bundle activity without digital averaging. Methods Recording was performed with very low noise, high gain amplifiers and by analog filtering. 3 brigade amplifiers and 16 bite A/D were connected to a microcomputer by USB. The signal was processed by A pattern recognition plus digital filtering software. The subjects were in a supine position during the recording. Results A prominent waveform was observed between the atrial and ventricular complexes and corresponded to His bundle activity in 46 volunteers. Recordings in 8 patients were compared with records obtained by intracardiac electrode, and a good conformation was seen. Conclusion The noninvasive method of beat to beat surface recording of His bundle activity has broad clinical applicability and high utility for the patients with latent A-V block and eletrophysiologic study.
7.New breakthrough in beat to beat surface recording of his bundle potential

Objective To evaluate a noninvasive method which has been applied on human subjects to observe beat to beat activity of the His bundle potential. Methods Recording was performed with very low noise, high gain amplifiers and by analog filtering. 3-brigade amplifiers and 16-bite A/D were connected to a microcomputer by USB. The signal was processed by a combined pattern recognition and digital filtering software. Results The results of surface monitoring were compared with intracardiac method in 30 patients. The results of non-invasive method were consistent with that interventional method. A prominent waveform was observed between the atrial and ventricular complexes (A, H, V waves), and corresponded to His bundle activity in all patients. Conclusion The noninvasive method of beat to beat surface recording of His bundle activity has broad clinical applicability and high utility for the patients with latent A-V block and eletrophysiologic study.
8.A nested PCR assay to diagnose disseminated trichosporonosis in murine model

Objective To explore the feasibility of the nested PCR technique to detect the Trichosporon asahii DNA in the blood and liver samples from the murine model of disseminated trichosporonosis.Methods On day 3,7,14 after intravenous inoculation of Trichosporon asahii suspension,blood and liver samples of 30 BALB/c mice were collected.Another ten normal mice serve as control.DNA was amplified by nested PCR,with a pair of primer.Fungal culture was done as control.Results On day 3,7,14 after inoculation,the positive rate of the serum fungal cultivation were 0,20%,0;the positive rate of the serum after nested PCR assay was 80%,90%, 88.9%;the positive rate of liver fungal cultivation were 50%,40%,22.2%;the positive rate of liver after nested PCR assay was 90%,90%,88.9%.There were statistical differences between the positive rate by nested PCR assay and fungal cultivation to check the serum and liver samples on day 3,7,14 after inoculation.The survival mice to the 14~(th) day were too less to make statistical analysis.Conclusion The results suggest that T.asahii DNA assay by nested PCR is more specific,more sensitive,and faster than fungal cultivation.The nested PCR assay could be a potential assay for the diagnosis of disseminated trichosporonosis.
9.A study on intrapulmonary expression of tumor necrosis factor-? gene and cell apoptosis in rats with acute pancreatitis

Objective To investigate the intrapulmonary expression of tumor necrosis factor-? (TNF-?) gene during acute pancreatitis, as well as correlation between TNF-? and lung injury seen in many patients undergoing acute necrotizing pancreatitis (ANP). Methods Ninety SD rats were divided into three groups randomly. Acute edematous pancreatitis(AEP) and ANP were induced by retrograde infusion of sodium taurocholate (0.5% and 5%) into biliopancreatic duct respectively. Animals in the last group only underwent sham-operation. All the animals were sacrificed at certain time, plasma level of amylase and TNF-? was assessed. Intrapulmonary expression of TNF-? mRNA and TNF-? protein was detected by semi-quantitative RT-PCR and immunohisto- chemistry. Cell apoptosis was assessed by TUNEL method. 10.Diagnosis and treatment of bladder outlet obstruction caused by bladder neck fibrosis (report of 30 cases)

Objective To present experience on the diagnosis and treatment of bladder outlet obstruction (BOO) caused by bladder neck fibrosis. Methods 30 cases of BOO caused by bladder neck fibrosis were studied. All the patients were treated by transurethral partial resection or incision of the cervix vesicae combined with alpha 1 adrenoceptor antagonists such as tamsulosin and terazosin or parazosin. Results Urination has been improved in all.The patients have been followed up for 3 to 30 months.On repeated uroflowmetry,the 30 patients showed the urine flow rates have been more than 15ml/s. IPSS scores was 5.3?1.7(25.4?4.2 preoperative) and quality of life scores was 1.4?0.6(4.1?0.8 preoperative).Fibroblast proliferation was noted in all the 30 and obvious chronic inflammation in 18. Conclusions Clinical symptoms, pressure rate measurement and cystoscopy are the reliable diagnostic methods.Transurethral partial resection or incision of cervix vesicae combined with alpha 1 adrenoceptor antagonists is effective.
