1.High concentration IL - 2 induces the expression of IL - 2 receptor chian on resting PBL

Using ~(125)I-labeled anti-IL-2R_? McAb, we investigated the effect of rIL-2 R_? expression . In addition to increasing the IL-2 R_? expression the actived-lymphoblasts, we found that the higher concentration of IL-2 also directly induced the IL-2 R_? expression on the resting PBL, but the peak was at the 5th day. After treated by a higher concentration of IL-2, there was a positive interrelation of time courses between the incorporation of ~3H-TdR and/or ~3H-UR and IL-2 R_? level on resting PBL. This result indicated that it was by means of inducing IL-2 R_? expression that the higher concentration IL-2 caused the proliferation of the resting PBL.
2.Clinical analysis of Level of glutathione redox-state in patients with acute coronary syndrome

Objective To study the changes and clinical significance of glutathione redox-state in patients with coronary heart disease (CHD). Methods Based on both with or without atherogenic risk factors and the result of coronary arteriography, 118 patients were calssified into three groups: control group( n =30) ,risk group(n =28) and CHD group (n =60). Venous blood sample was obtained, then the plasma reduced glutathione ( GSH) and oxidized glutathione (GSSC) were measured by using glutathione reductase recycling method. Redox potential(EhCSH/GSSG) was calculated using Nernst equation. According to clinical situation,CHD group was divided into two subgroups:un-stable angina pectoris( UAP) group(n = 32) and acute myocardial infarction (AMI) group (n =28). Results From control group to risk group,then to CHD group,GSH gradually decreased,and there was significant difference between CHD group and control group (P < 0.05 ) , but GSSG and EhGSH/GSSG gradually increased, there was significant difference between CHD group and risk group and control group (P < 0. 05). There was no significant difference be-tween the UAP subgroup and AMI subgroup in GSH,GSSG and EhGSH/GSSG(P >0.05). Conclusions Body's an-tioxidant capacity decreases and the oxidative stress exists in artherosclerosis, and furthermore there is a certain degree of relevance between oxidative stress and hyperlipoidemia,all these show that improving this situation may decelerate or prevent the formation and progress of artherosclerosis.
3.Comparison of the curative effect between two methods of helicobacter pylori treatment for peptic ulcer evaluation

Objective To compare the clinical curative effect between sequential therapy and triple therapy for helicobacter pylori positive peptic ulcer.Methods 92 patients with peptic dcer were randomly divided into the observation group and the control group,46 cases in each group.The observation group was treated with sequential therapy,control group with triple therapy.The two groups were observed and compared in helicobacter pylori eradication rate,ulcer healing and relief of symptoms.Results After 4 weeks treatment,the observation group Hp clearance rate was 93.5% (43/46),ulcer healing rate 91.3% (42/46),the control group were 73.9% (34/46),78.3%(36/46),two groups had significant differences(x2 =2.714,3.028,all P < 0.05) ;observation group after treatment of symptomatic integral average (0.94 ± 0.21),and the control group (0.92 ± 0.17) divided the difference was not statistically significant (t =0.483,P > 0.05).Conclusion The sequential therapy on helicobacter pylori positive peptic ulcer curative effect is remarkable,and can be more effective in the eradication of helicobacter pylori infection,promote ulcer healing.
4.Purification and characterization of soluble interleukin 2 receptor

By using 5 G 1-sepharose 4 B immune affinity column, we recovered approximately 160 ?g purified and soluble IL-2 receptor (sIL-2 R ) from 500ml PHA-stimulated human PBL supernatant. Only one band was showed in SDS-PAGE, its molecular weight was about 44 Kd. The purified sIL-2 R, which blocked the binding of anti-IL-2 R monoclonal antibody with IL- 2 R~+ cell, inhibited (at the higher doses) but enhanced (at the lower doses) the growth of IL-2 dependent cells.

Objective To investigate the intrapulmonary expression of tumor necrosis factor-? (TNF-?) gene during acute pancreatitis, as well as correlation between TNF-? and lung injury seen in many patients undergoing acute necrotizing pancreatitis (ANP). Methods Ninety SD rats were divided into three groups randomly. Acute edematous pancreatitis(AEP) and ANP were induced by retrograde infusion of sodium taurocholate (0.5% and 5%) into biliopancreatic duct respectively. Animals in the last group only underwent sham-operation. All the animals were sacrificed at certain time, plasma level of amylase and TNF-? was assessed. Intrapulmonary expression of TNF-? mRNA and TNF-? protein was detected by semi-quantitative RT-PCR and immunohisto- chemistry. Cell apoptosis was assessed by TUNEL method. 10.Effectiveness of early goal-directed fluid therapy with fresh frozen plasma for severe acute pancre-atitis

Objective To evaluate the effect of early goal-directed fluid therapy with fresh frozen plasma in severe acute pancreatitis(SAP). Methods From January 2010 to June 2014,79 SAP patients were enrolled according to the continuous sampling method. All the patients were randomly divided into a control group who accept the traditional fluid therapy(group A),an experimental group 1 who accept early goal-directed fluid therapy(group B),and an experimental group 2 who accept the early goal-directed fluid therapy with fresh frozen plasma(group C). There were no significant differences of general conditions a-mong groups. The differences of ICU admission,mortality and occurrence rate of abdominal compartment syndrome(ACS)and MODS were compared among groups. Results Compared with group A,group B and C have a shorter length of ICU admission,a lower mortality and a lower occurrence rate of ACS and MODS (P < 0. 05). Compared with group B,group C have a shorter length of ICU admission,a lower mortality and a lower occurrence rate of ACS and MODS(P < 0. 05). Conclusion The method of early goal-direct-ed fluid therapy with fresh frozen plasma will contribute to shorten the length of ICU admission and reduce mortality and occurrence rate of ACS and MODS for patients with SAP.
