Basic & Clinical Medicine 2006;0(06):-
Endoplasmic reticulum stress,an critical component of cell stress,is a protective response for eukaryotic cells. The change of partial pressure of oxygen (for instance hypoxia) is a primary condition for cell stress, significantly impacting the biological phenotype and behavior of cells (including survival, migration and invasion et al). Through endoplasmic reticulum stress, intracellular concentration of unfolded protein is reduced, and agglutination inhibited. Recently, endoplasmic reticulum stress has been demonstrated to play an important role in pathogenesis of some diseases such as cancer, diabetes and inflammation, which are characterized by the micro-environment hypoxia in local tissue. Therefore, investigations on the mechanism of endoplasmic reticulum stress caused by hypoxia may promote the novel therapeutic strategy for tumors, cardiovascular diseases and diabetes.

3. Proteomics technique in screening of differential proteins for liver regeneration after partial hepatectomy in rats
Academic Journal of Second Military Medical University 2010;29(1):30-35
Objective: To screen for the differentially expressed proteins during rat liver regeneration after partial hepatectomy (PH) by proteomics technique. Methods: Healthy male SD rats were randomly divided into 2 groups: hepatectomy group and sham operated group. The hepatectomy model was produced by 70% PH (n=35) and the sham operated rats (n=5) underwent the same surgical protocol without hepatectomy. Rats were executed at 2, 12, 24, 36, 48, 72 and 168 h after partial hepatectomy (each time 5 rats) and the right lobes were harvested. The total protein was extracted and analyzed by two-dimensional gel electrophoresis and mass spectrometric analysis. The differential proteins were then analyzed by Western blotting. Results: The spots of differential protein began to rise at 2 h after PH and peaked at 36 h after PH. A total of 78 protein spots were identified and 35 significant protein spots were found by mass spectrometric analysis. The 35 protein sports fell into 5 types according to their dynamic changes: 3 up-regulated and 2 down-regulated (with different regulation time periods and amplitudes); and their functions involved oxidative stress response, acute reaction, lipid and energy metabolism, intracellular signaling transduction, cell proliferation, etc., with some having unknown functions. Western blotting analysis showed that the prohibitin protein began to increase 2 h after PH and decreased to the normal levei after 48 h. Conclusion: It is indicated that many proteins and signal transduction pathways participate in the liver regeneration after partial hepatectomy.

Chinese Journal of Experimental Ophthalmology 2012;30(4):326-330
BackgroundHypoxia and hyperglycemia are the common causes of retinal neovascularization. In these states,H+ accumulates because of the elevated glycolysis and failure of retinal circulation, thus the retinas readily acidified. ObjectiveThe present study was to explore whether retinal acidosis independently regulates the production of vascular endothelial growth factor (VEGF) and pigment epithelium-derived factor (PEDF) and whether the regulation is related to oxidative stress. Methods The retinas from 2-week-old male SD rats were cultured with explant method in DMEM modulated by NaHCO3,and culture retinas were randomly divided into pH 7.2,6.8 and 6.5 groups for 24 hours. In addition, after 24 hours of culture as above described, retinas were washed using PBS two times and then followed by again culture in DMEM with pH 7.2 for another 24 hours. Also, antioxidant was added in different pH values of DMEM for culture as above described. The retinal samples were prepared for histopathological examination. The expressions of VEGF and PEDF proteins and their mRNA in retina tissue were detected by Western blot and fluorescence quantitative polymerase chain reaction (PCR) respectively. Results The retina showed the clear structure and morphology in pH 7.2 group and pH 6.8 group, but retinal vacuoles change was seen in pH 6.5 group after culture for 24 hours. No significant difference was seen in the expressing level of VEGF mRNA in retina between normal group and pH 7.2 group( 112% ±11% vs 100% ±7% ) (P=0.55),but those in pH 6.8 group and pH 6.5 group were significant increased in comparison with pH 7.2 group( 196% ±43% vs 100% ±7% ;251% ±29%vs 100% ±7% )( P<0.05 ).The expressing level of PEDF mRNA in retina in normal group was similar to that of pH7.2 group(86% ±19% vs 100% ±33%) (P=0.64),but that in pH 6.5 group was significantly higher than pH 7.2 group( 230% ±66% vs 100% ±33% ) ( P<0.05 ).The resemble results were found in the expressions of VEGF and PEDF protein. After pH reversion, the expressing levels of VEGF mRNA were 100% ±13%,111% ±9%,113% ±9% in pH 7.2 group,pH 6.8 group and pH 6.5 group respectively without significant difference among them (F=2.51,P=0.16).The expressing levels of PEDF mRNA were 100% ±13%,110% ±9%,108% ±11%in different groups ( F =0.98,P =0.43 ).Under the presence of antioxidant, the expressing level of VEGF mRNA in pH 6.5 group increased in comparison with pH 7.2 group and pH 6.8 group ( P < 0.05 ).The expressing levels of PEDF mRNA were significant different among pH 7.2 group( 100±31 )%,pH 6.8 group( 282±45 )% and pH 6.5 group(480±117)% (F=20.73,P=0.00). Conclusions VEGF can be induced by retinal acidification alone, which may be regulated by oxidative stress.Under the retinal acidification, antioxidants promote the expression of PEDF,suggesting that oxidative stress inhibits the production of PEDF.

OBJECTIVETo evaluate the accuracy and safety of pedicle screw insertion with the aid of novel patient-specific drill-guide templates in scoliosis cases.
METHODSTen patients with scoliosis were selected to participate in the research (the observation group) from December 2013 to December 2014. The data was obtained from CT scanning, and put into the computer to perform reconstruction of spine, simulation of pedicle screw insertion, and design of patient-specific drill-guide templates with software. The templates were made with rapid prototyping technique. After sterilization, the templates were used to aid the pedicle screw insertion intraoperatively. The blood loss, operation duration, change of creatinine level pre- and post-operation, and complications related to pedicle screw insertion were recorded. The location of pedicle screws were graded so as to evaluate the accuracy. A comparative study was then performed with the data of ten scoliosis cases operated with free-hand method during the same period (control group). There were 5 cases of idiopathic scoliosis and 5 cases of congenital scoliosis in the observation group, including 3 males and 7 females. Their average age was 11.9 years old (ranged, 4 to 18 years old), and the average Cobb angle of main curve was 54.9° (ranged, 42.1° to 78.4°). There were also 5 cases of idiopathic scoliosis and 5 cases of congenital scoliosis in the control group, including 2 males and 8 females. Their average age was 12.6 years old (ranged, 6 to 17 years old), and the average Cobb angle of main curve was 56.6° (ranged, 38.2° to 93.4°).
RESULTSA total of 167 pedicle screws were inserted intraoperatively, with 138 screws (82.6%) in grade I, 26 screws (15.0%) in grade II, 4 screws in grade III (2.4%), but no screws in grade IV according to the CT image. There were 29 (17.4%) screws perforated, and 163 (97.6%) screws could be accepted. In the control group, a total of 165 pedicle screws were inserted intraoperatively, with 98 screws (59.4%) in grade I, 39 screws (23.6%) in grade II, 21 screws in grade III (12.7%), and 7 screws in grade IV (4.2%). There were 67 (40.6%) screws perforated, and 137 (83.0%) screws could be accepted. The grade distribution of screw position, ratio of perforated and accepted screws were significantly different between the two groups respectively (Z=-5.013, P=0.000; χ2=9.347, P=0.002; χ2=20.242, P=0.000). The correction rate of Cobb angle were (74.1±10.0)% vs (69.7±17.6)%; blood loss were (455±447) ml vs (415±389) ml; operation duration were (163.5±53.7) min vs (164.0±48.7) min; and the changes of creatinine level pre- and post-operatively were (-5.3±3.2) μmol/L vs (-3.4±3.1) μmol/L; all above data had no significant differences respectively (t=0.696, P=0.496; t=0.214, P=0.833; t=0.022, P=0.983; t=1.375, P=0.192). There were no complications related to pedicle screw insertion in each group.
CONCLUSIONThe novel patient-specific drill guide template can be used to assist the insertion of pedicle screws in scoliosis cases with much higher accuracy than that of freehand method and fair safety.
