1.Characters of angiopoietins and their functions in the regulation of neovascularization

Neovascularization plays an important role in embyonic development and many diseases. VEGFs and angiopoietins are two known growth factor families that are specific to vascular endothelium. The action of angiopoietins is associated not only with angiogenesis but also with postnatal vasulogenesis. Thus there is a good prospective use for angiopoietins or their antagonists to promote or inhibit angiogenesis in clinical therapy.
2.Effect of bFGF on expression of endothelial cell integrin ?_1 subunits

AIM: To study the effect of basic fibroblast growth factor(bFGF) to the expression of integrin ? 1 subunit of endothelial cells. METHOD: The expression of integrin ? 1 subunit of endothelial cells was determined before and after bFGF treatment with Western Blot and immage analysis method.RESULTS: The mean gray value of immunostain by immage analysis method is 166.11?9.86 in the experimental group and 175.32?5.12 in the control group, suggested that bFGF may upregulate expression of integrin ? 1 subunits of endothelial cell. CONCLUSION: bFGF may play an important role in angiogenesis by way of inducing the overexpression of endothelial cell integrin ? 1 subunits.
3.Progress in the research of mechanism underlying the inhibitory effect of thrombospondin-1 on tumor angiogenesis

Inhibition of tumor neovascularization (angiogenesis) has become one of the novel therapeutic strategies for tumor, and natural inhibitor of angiogenesis has been highlighted in this field. Thrombospondin-1 (TSP-1), as an important extracellular matrix component which can influence endothelial cell adhesion, migration, and proliferation, is reported to be a potent inhibitor of angiogenesis in vivo. It will be valuable to use its active peptides or small fragments from TSP-1 in antiangiogenesis therapy.
4.Correlative study among CT signs, prognosis and the expression of CD44v3 and CD44v6 in peripheral non-small-cell lung cancer

Objective To observe the relationship between CT signs, prognosis and the expression of CD44v3 and CD44v6 in peripheral non-small-cell lung cancer (NSCLC). Methods The expression of CD44v3 and CD44v6 was detected with immunohistochemistry (SP method) in 46 patients with peripheral NSCLC confirmed by surgery and pathology. The expression, CT signs and prognosis of NSCLC were analyzed respectively. Results The expression of CD44v3 and CD44v6 correlated with deep lobulation, spinous protuberant sign, short spiculation and lymph node enlargement of hila or mediastinum (P<0.05), but not with the tumor size, vacuole sign, vessel convergence sign and pleural indentation in CT images of peripheral NSCLC (P>0.05). The expression of CD44v3 and CD44v6 correlated with the three-year survival rate in peripheral NSCLC patients (P<0.05). Conclusion Deep lobulation, spinous protuberant sign, short spiculation and lymph node enlargement of hila or mediastinum may indicate strong potential invasion and metastasis in peripheral NSCLC. The positive expression of CD44v3 and CD44v6 may suggest shorter survival time and poorer prognosis in peripheral NSCLC patients.
5.A study on the mechanism of bFGF promoting endothelial cells to adhere to polyurethane material.

bFGF can promote endothelial cell to adhere to artificial polyurethane material. However the mechanism is still unclear. Here the effect of bFGF on endothelial cell expression of integrin beta 1 subunits was determined with Western Blot and image analysis method. The result reveal that the mean gray value of immunostain is 166.1 +/- 9.86 in the experimental group and 175.32 +/- 5.12 in the control group, suggest that bFGF may upregulate the endothelial cell expression of integrin beta 1 subunits. It is concluded that the bFGF may induce the overexpression of endothelial cell integrin beta 1 subunits to increase the cell adhesion onto polyurethane material. It is suggested that bFGF can be taken as an important assistant factor in accomplishment of endothelialization on artificial heart and blood vessel.

6.Correlation analysis of gonadal hormone and seminal plasma with semen liquefaction

Objective To analyze the gonadal hormone and seminal plasma of patients with abnormal semen liquefaction and investigate the influence mechanism in order to provide guidance for the diagnosis and treatment. Methods 152 men of childbearing age were divided into two groups according to the liquefaction time (cut?off point: 60 minutes). Routine semen parameters,gonadal hormone and seminal plasma were tested and compared between the above groups. T?test was applied to compare individual gland function (pH value,neutralα?glycosidase,fructopyranose,seminal plasma zinc and citric acid) and gonadal hormone (FSH,LH,PRL,T and E2). Logistic regression analysis was adopted to probe the influencing factors for abnormal semen liquefaction. Results Seminal pH value (7.47 ± 0.13 vs. 7.32 ± 0.18),citric acid(51.12 ± 12.95 vs. 83.11 ± 33.46)and FSH (4.40 ± 1.03 vs. 4.85 ± 1.50)levels were significant different between the two groups (P < 0.05),but the other indexes showed no significant difference. Correlation regression analysis showed that semen liquefaction capacity has correlative relationship with seminal plasma fructose (OR=2.644),citric acid (OR=0.922),serum T (OR=1.029) and E2,while no correlative relationship with other indexes. Conclusions Correlation between two glands (seminal vesicle and prostate) and balance in the two hormones (T and E2) influence the liquefaction time. Specific causes should be distinguished before diagnosis.
7.A study on the endothelialization mechanism of left ventricular pump.

The cultural model of porcine aortic endothelial cell (EC) was set up. By combined use of basic fibroblast growth factor(bFGF) with extracellular matrix (ECM), the endothelialization on PU under static culture was accomplished and the attachment rate as well as the morphological changes of EC under flowing shear stress condition were observed when LVAP was working at normal blood circulation pressure (12 KPa). The result revealed that EC treated with bFGF grew very well on the PU material precoated with fibronectin and collagen IV. Under shear stress condition for 1 and 2 hours, the percentage of cell attachment attained 74.42% and 59.7%. The general morphological changes of the cell were not obvious under the scanning electron microscope, slight shrink and cell death or loss were found. This work was done to find out the condition and method of experiment for the accomplishment of endothelialization under the flowing shear stress in the cavity of LVAP.

8.Bioinformatics analysis of angiogenesis-related miRNAs in endothelial progenitor cell exosomes

Objective By taking usage of bioinformatics screening methods,this medical research aimed at exploring how the miRNAs in endothelial progenitor cell exosomes relate to the regulation of angiogenesis.Methods miRNAs in endothelial progenitor cells exosomes and angiogenesis-related miRNA was intersected from the existing database to obtain the candidates of miRNA molecules related to angiogenesis in endothelial progenitor exosomes.Results 160 and 50 miRNAs in endothelial progenitor cell candidates were obtained through experimental data analysis and literature searching respectively.600 candidates of angiogenesis-related miRNAs were obtained through literature searching;the top 20 with the highest frequency were selected out.Finally,9 miRNA candidates (miR-126,miR-21,miR-221,miR-92a,miR-199a,miR-210,miR-214,miR-155,miR-146a) that may be highly expressed in endothelial progenitor exosomes and associated with angiogenesis were obtained for the following research.Conclusions Based on data analysis and literature searching,bioinformatics could screen out the target miRNAs for follow-up studies easily and reliably,it is worthy to be widely applied and popularized.
9.Quality Marker (Q-marker) of Tinosporae Radix Associated with Efficacy of "Relieving Sore Throat"

ObjectiveTo study the potential quality marker (Q-marker) of Tinosporae Radix associated with efficacy of "relieving sore throat" based on ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS), multivariate statistical analysis (MSA), and network pharmacology. MethodUPLC-Q-TOF-MS was used to identify the main chemical components in 18 batches of Tinosporae Radix. On this basis, principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA) were employed to screen out the main marker components that caused differences between groups. Moreover, network pharmacology technology was applied to predict the potential "sore throat-relieving" components, and the molecular docking between the common components resulting from MSA and network pharmacology and the core targets was carried out to verify the marker components. ResultA total of 17 compounds, including alkaloids, diterpenoid lactones, and sterols, were identified by UPLC-Q-TOF-MS. Five main differential components were found by MSA: Columbamine, jatrorrhizine, palmatine, menisperine, and columbin. Network pharmacology analysis yielded six compounds: tetrahydropalmatine, palmatine, menisperine, fibleucin, neoechinulin A, and columbin which were selected as potential "sore throat-relieving" components of Tinosporae Radix. They may relieve sore throat by acting on interleukin-6, epidermal growth factor receptor, prostaglandin G/H synthase 2, matrix metalloproteinase-9, proto-oncogene tyrosine-protein kinase Src and other targets, and regulating Hepatitis B, influenza A, human T-cell virus infection, human cytomegalovirus infection, coronavirus disease-2019, and other signaling pathways. The common active components in Tinosporae Radix resulting from MSA and network pharmacology analysis were palmatine, menisperine, and columbin, which had high binding affinity with six core targets and can be used as the Q-marker components of Tinosporae Radix in "relieving sore throat". ConclusionThis study predicts the "sore throat-relieving" Q-marker of Tinosporae Radix, which lays a basis for developing the quality standard of Tinosporae Radix based on the efficacy and improving the quality evaluation system of the medicinal.
