1.Establishment of PAF receptor radioligand binding assay in mouse peritoneal macrophages

AIM To establish radioligand binding assay of PAF (platelet activating factor) receptors in mouse peritoneal macrophages and observe the characteristics of PAF receptors. METHODS PAF receptor radioligand binding was studied in intact adherent mouse peritoneal macrophages by -PAF. The radioactivity was counted with an LS6500 scintillation system. RESULTS The PAF receptor binding was shown to be saturable with an equilibrium K D of 3.2 nmol?L -1 and a B max of 100.2 fmol?1?10 6 cells -1. The competitive analysis showed that such specific binding could be inhibited by BN52021. CONCLUSION Utilizing the adherent character of macrophages, the binding ligands could be separated from non-binding ligands without negative pressure filtration, then cells could reserve fine activity, and PAF receptors could be near to physiological properties for screening of PAF antagonist.
2.Status Analysis and Countermeasure Research for Patent Application Quality of Traditional Chinese Medicine

Traditional Chinese medicine (TCM) is a treasure of five-thousand-year civilization of China, with systemic theory and reliable clinical efficacy. As the origin of TCM, China should have a large number of independent intellectual property rights of TCM beyond any doubt. However, the quality of TCM patent application as a whole is far from optimistic currently. Targeting the differences in number and quality of TCM patent application in China, this article analyzed current situation of TCM patent application, discussed the reasons and offered countermeasures, and proposed that the work focus of intellectual property in China should shift from quantity to quality. It also argued that targeted countermeasures should be chosen according to the causing reasons, and overall promotion of the TCM intellectual property competiveness should be achieved through multi-aspect and multi-department cooperation.
3.Effect of ginkgolide B on the platelet-activating factor induced changes of chemotaxis and cytoskeleton of macrophages

Aim To study the inhibitory effect of ginkgolide B (BN52021) on the PAF induced changes of chemotaxis of murine peritoneal macrophages and the related polymerization of F-actin.Methods Chemotaxis assays were performed using a modified 48-well Boyden chamber. Actin polymerization of murine peritoneal macrophages was analyzed by flow cytometry using a specific fluorescent stain. Results Peritoneal macrophages significantly migrated toward platelet-activating factor(PAF) through a micropore filter; however, in the presence of PAF receptor antagonist BN52021 (0. 01the actin polymerization of murine peritoneal macrophages induced by PAF in the presence of Ca2+ , but not in Ca2+ -free medium. Conclusion The results suggested that preventing polymerization of F-actin may be a pathway by BN52021 to inhibit the chemotaxis of macrophages, and this effect seems to be Ca2+dependent. The data further indicated that inhibition of PAF induced macrophage chemotaxis is an important mechanism underlying the anti-inflammatory action of BN52021.
4.Research on influencing of CYP2C18 gene polymorphism on warfarin dose after valve replacement surgery

Objective To investigate the CYP2C18,VKORC1 and CYP2C9 genotype distribution situation and the correlation between their polymorphism with warfarin stable dose.Methods A total of 176 Yungui plateau Han patients with continuously warfarin anticoagulation therapy after valve replacement in this hospital from January 2011 to January 2014 served as the research subjects.The venous blood was collected for detecting genotypes at various loci.The genotype and allele frequency distribution were analyzed,and the correlation between the gene polymorphism with warfarin stable dose was analyzed.Results The genotypes and allele frequencies at various loci conformed to the Hardy-Winberg genetic balance (P>0.05).The polymorphisms of CYP2C18 (rs7896133),VKORC1 (rs9923231),CYP2C9(rs1057910) and CYP2C9(rs4086116) gene loci were correlated with warfarin stable dose (P>0.05).Conclusion The gene polymorphism of CYP2C18(rs7896133),VKORC1 (rs9923231),CYP2C9(rs1057910) and CYP2C9(rs4086116) in Yungui plateau H an patients may be the influence factors which contribute to warfarin stable dose personalized difference.
