1.The role of Fas/FasL expression caused by H.pylori in gastric adenocarcinoma.

Objective To observe the effects of different H.pylori strains (cagA + and cagA -) infection on Fas/FasL expression and the pathogenesis of gastirc carcinoma. Methods Gastric biopsy specimens were taken from 80 patients, and these patients were divided into 9 groups according to the H E staining and H.pylori infection. H.pylori (+) groups were subdivided into cagA + and cagA - groups. RUT, PCR and histology were used to detect H.pylori and PCR for cagA detection. Immunohistochemical staining was used to detect Fas/FasL expression. Results The infection rate of H.pylori was 60.0%, and rate of cagA(+) is 90.47%. H.pylori (+) specimens express Fas/FasL more higher than H.poylori (-) ones ( P 0.05). Conclusions Our data suggest that H.pylori infection can induce the expression of Fas/FasL in gastric epithelial cells. Gastric adenocarcinoma express high amount of FasL, which may plays an important role in immune escape of stomach cancer.
2.A study of the relationship between intestinal subepithelial myofibroblasts and ulcerative colitis

Objective Intestinal subepithelial myofibroblasts(ISEMFs) exist throughout the gastrointestinal tract. Due to their ability to secrete cytokines, prostaglandins, growth factors, and extracellular matrix proteins, they are thought to play critical roles in inflammation, repair, fibrosis and neoplasia. The goal of this study was to investigate the relationship between ISEMFs and ulcerative colitis by establishing the method of ISEMFs detection from the biopsy specimens. Methods ISEMFs in colon biopsy specimens (15 from active ulcerative colitis patients and 20 from controls) were detected by immunohistochemistry with smooth muscle actin monoclonal antibody. Results ISEMFs existed in a subepithelial location of colon, were located in the basement of epithelial, and were oval or scaphoid in appearance. 3.Prediction of fetal macrosomia:evaluation of the combined application of ultrasound and clinical measurement

Objective To investigate the relativity for weight of macrosomia predicted by ultrasound and clinical measurements . Methods 1 037 subjects who were delivered in May to November 2012 ,in Chongqing Health Center for Women and Children ,were collected and analyzed satistically .The parameters for the comparisons included maternal abdominal circumference (MAC) ,fundal height(FH) ,and fetal diameters measured by ultrasonic imaging such as biparietal diameter (BPD) ,head circumference(HC) ,femur length(FL) ,and abdominal circumference(AC) .The coincidence was calculated with real weights and compared with clinical predic-tions .Results When FH+ MAC 134 cm ,the accuracy of preliminary screening for macrosomia is the highest ,the area under the ROC curve is 0 .895 .Combining with the abdominal circumference(AC) by prenatal ultrasound ,the highest prediction rate of mac-rosomia is 75 .2% .when Fetal AC≥37 cm ,macrosomia incidence rate took 91 .7% .Conclusion Pregnant women with FH+MAC<134 cm ,whose incidence of Macrosomia is low .While if the FH+MAC≥134 cm ,combining with the prenatal ultrasound whose fetal AC>36 cm ,especially ≥37 cm ,the possibility of macrosomia is very high .
4.Clinical Study on Trigger Points Combined with Acupoints by Acupuncture for Subjective ;Tinnitus

Objective To assess the therapeutic efficacy of trigger points (TrPs) combined with acupoints by acupuncture for subjective tinnitus (ST);To discuss the correlation of TrPs acupuncture inactivation and the releasing of symptom. Methods Sixty patients with ST were randomly divided into trial group and control group, 30 cases in each group. Those in trial group were acupunctured by TrPs combined with acupoints, and those in control group received regular acupuncture treatment. The scores of tinnitus severity assessment were available for effect evaluation after 3 treatment course. The size and tenderness degree of TrPs contraction knots were checked, and TrPs local thermal infrared spectrum was compared by infrared camera. Results The post-treatment tinnitus severity scores had significant difference compared with pre-therapy in each group (P<0.01). The total effective percentage of trial group with 93.33%(28/30) was obviously higher than control group with 86.67%(26/30) (P<0.05). The post-treatment and pre-therapy contraction knots size, tenderness and temperature difference of TrPs within trial groups had a very significant difference (P<0.01), and difference of tenderness and temperature of patients’ TrPs had a positive correlation with the difference of tinnitus severity scores before and after treatment (P<0.01). The contraction knots area of TrPs had not significant correlation with the difference of tinnitus severity scores (P>0.05). Conclusion TrPs combined with acupoints by acupuncture for ST patients are better than conventional acupoints in improving clinical symptoms. The deactivation of TrPs is closely related to tinnitus symptoms, therefore, temperature and tenderness of TrPs can be employed to assist in the diagnosis, or evaluated the tinnitus severity and therapeutic efficacy.
5.Research progress of the central mechanism of acupuncture-moxibustion for simple obesity

Obesity often results when regular caloric intake exceeds the energy expenditure. Hypothalamus is the center that regulates feeding and energy balance inside the body, especially the lateral hypothalamic area (LHA) and ventromedial nucleus (VMN). Acupuncture-moxibustion has achieved significant progress in treating simple obesity, and the relevant studies have revealed the central action mechanism underlying the treatment. By reviewing the literatures associated with the mechanism studies on acupuncture-moxibustion for simple obesity, this article is going to provide clinical and scientific guidance for the treatment of simple obesity with acupuncture-moxibustion.
6.Clinical and neuroimaging features of frontotemporal dementia with parkinsonism linked to chromosome 17

Objective To explore the clinical and neuroimaging features of a frontotemporal dementia with parkinsonism linked to chromosome 17 ( FTDP-17 ) pedigree caused by mutation of microtubule-associated protein tau ( MAPT) gene.Methods The proband and one patient from a FTDP-17 pedigree were assessed through standardized clinical evaluation , neuropsychology assessment , video-electroencephalogrom ,MRI, genetic sequencing , as well as 18 F fludeoxyglucose ( FDG) SPECT for brain metabolism and 11 C 2β-carbomethoxy-3β-( 4-fluoro ) tropane ( CFT ) PET for dopamine transporter ( DAT ) distribution, respectively.Results A FTDP pedigree with 15 patients (6 still alive) was recruited to this study.The proband and one affected patient were genotyped and confirmed as MAPT c .1788T>G mutation. Parkinsonism was the first symptom for both two patients . Personality, speech changes and dementia accompanied with brain atrophy were developed at the later stage in one patient .The 18 F FDG SPECT studies illustrated asymmetric hypometabolism of the temporal , frontal lobes and basal ganglia in two patients . Regarding to the 11 C CFT PET, one affected patient showed asymmetric decreased uptake of tracer in basal ganglia regions.Conclusions FTDP-17 can display a confusingly broad clinical phenotype , with the parkinsonism as the first symptom . Brain glucose metabolism and DAT distribution could be potential biomarkers in early diagnosis of FTDP-17.
7.Simultaneous Determination of 10 Flavonoids in Astragalus membranaceus by HPLC

OBJECTIVE:To establish a method for simultaneous determination of 10 flavonoids in Astragalus membranaceus. METHODS:HPLC method was adopted. The determination was performed on Agilent SB-C18 column with mobile phase consisted of acetonitrile-0.3% formic acid (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was set at 254 nm, and the column temperature was 35 ℃. The sample size was 10 μL. RESULTS:The linear ranges of calycosin-7-O-glucoside,iso-quercitrin,genistin,ononin,calycosin,quercetin,genistein,kaempferol,isorhamnetin and formononetion were 0.03029-1.5145μg (r=0.9994),0.01500-0.7500 μg(r=0.9995),0.00739-0.3695 μg(r=0.9991),0.12011-6.0055 μg(r=0.9998),0.03836-1.918 μg (r=0.9999),0.02989-1.4945 μg(r=0.9995),0.00704-0.352 μg(r=0.9994),0.01683-0.8415 μg(r=0.9995),0.00454-0.227μg(r=0.9999),0.01336-0.668 μg(r=0.9999),respectively. RSDs of precision,stability and reproducibility tests were all lower than 2.0% . The recoveries were 99.55% -100.45%(RSD=0.36% ,n=6) ,99.34% -101.00%(RSD=0.59% ,n=6) , 98.05%-100.36%(RSD=1.27%,n=6),99.73%-100.13%(RSD=0.18%,n=6),99.70%-100.30%(RSD=0.22%,n=6), 99.67%-103.27%(RSD=1.37%,n=6),98.13%-104.41%(RSD=2.37%,n=6),96.35%-100.06%(RSD=1.46%,n=6), 99.47%-101.13%(RSD=0.60%,n=6),99.70%-100.06%(RSD=0.15%,n=6),respectively. CONCLUSIONS:This method is convenient,sensitive,stable and reproducible,can be used for simultaneous determination of 10 flavonoids in A. membranaceus.
8.Mutation analysis of the PNPLA1 gene in a family with autosomal recessive congenital ichthyosis

Objective To identify a causative gene of autosomal recessive congenital ichthyosis (ARCI) in a Chinese family,and to analyze the genotype-phenotype correlation.Methods Peripheral blood samples were collected from the proband,his elder brother and parents,and genomic DNA was extracted from these blood samples.Genome-wide exome sequencing was conducted to determine the mutation site in the proband,and then allele-specific oligonucleotide primers were designed based on the mutation site.PCR was performed to detect the mutation site to further identify the causative gene of ARCI in the family.Results A new homozygous missense mutation was identified in exon 4 in 1 allele of the PNPLA1 gene in the proband,which led to a codon change from cytosine (C) to thymine (T) at position 700 (c.700C > T) and resulted in the substitution of proline by serine (p.pro234ser).The same mutation was also detected in the proband's brother,and his parents were the mutation carriers.No mutations were found in unrelated healthy Chinese individuals.Conclusion The missense mutation in the PNPLA1 gene (p.pro234ser) is associated with clinical symptoms of the patient with ARCI.
9.Theoretic analysis of Professor Shao Jing-ming's 'Three-acupoint and Five-needle Method' for asthma

'Three-acupoint and Five-needle Method' is a summary of Prof. Shao Jing-ming's clinical experience in the treatment of asthma for years. It is used to alleviate asthma during seizure and to improve the pulmonary functions, strengthen the body constitution and prevent recurrence in the remission stage. This article introduces the theoretic source and basic principle, and concrete operating method, i.e. puncturing bilateral Feishu (BL 13), Dazhui (GV 14) and bilateral Fengmen (BL 12), and adding acupoints based upon pattern identification, in combination of moxibustion and cupping therapy for promoting the clinical application of this method.
10.Systematic Review of the Efficacy and Safety of Rosiglitazone and Metformin in the Treatment of Type 2 Dia-betes

OBJECTIVE:To systematically review the efficacy and safety of rosiglitazone and metformin in the treatment of type 2 diabetes,and provide evidence-based reference for the clinical treatment. METHODS:Retrieved from PubMed,Medline, EMBase,Cochrane Library,CJFD,VIP and Wanfang database,randomized controlled trials (RCT) about rosiglitazone (test group)and metformin(control group)in the treatment of type 2 diabetes. After data extract and quality evaluation,Meta-analysis was performed by using Rev Man 5.0 statistics software. RESULTS:A total of 5 RCT were included involving 636 patients. Re-sults of Meta-analysis showed the incidence of diarrhea [RR=0.23,95%CI(0.07,0.81),P=0.000] in test groupwere significantly lower than control group,the HbA1c level [MD=0.22,95%CI(0.07,0.38),P=0.004],the incidence of edema [RR=0.20,95%CI (0.05,0.57),P=0.011] in test group was significantly higher than control group,and there was no significant difference in the fast-ing blood glucose level[MD=0.27,95%CI(-0.26,0.79),P=0.32],the incidence of nausea/vomiting [RR=0.94,95%CI(0.06, 0.89),P=0.692]between 2 groups. CONCLUSIONS:Metformin is more effective than rosiglitazone in the treatment of type 2 dia-betes,however,the incidence of diarrhea should be noticed. Due to the limit of methodological quality and sample size,it remains to be further verified with more rigorously designed and long-term follow-up of large-scale RCT.
