Objective To investigate the effects of hyperbaric oxygen (HBO) on the expression of caveolin2 and matrix-metalloproteinase-9 (MMP-9) in brain tissues and the blood brain barrier (BBB) after cerebral ischemia and reperfusion (I/R) in rats. Methods Three hundred and seventy male Wistar rats were randomly assigned into sham,ischemia-reperfusion,HBO,and I/R + HBO groups. After creating cerebral I/R models,oxygen at 0.25 MPa was administered 5 times,and 2% Evans blue (EB) was injected into the tail veins 1 h before the rats were sacrificed.The permeability of the BBB,the expression of caveolin-2 and MMP-9,and EB content were determined by Western blotting,immunohistochemistry,and spectrophotometery,respectively. Results In the I/R group,the EB content increased steadily to a peak at 4 hours.EB content in the IR + HBO group was significantly lower than in the I/R group.Caveolin-2 and MMP-9 were significantly augmented by I/R injury at the 24th,48th and 72nd hours.Compared to the I/R group,HBO intervention decreased their expression levels. Conclusion HBO intervention can reverse the increase of caveolin-2 and MMP-9 caused by I/R injury,which suggests a mechanism for protective effects of HBO on the permeability of the BBB in I/R injury.
3.Effects of hyperbaric oxygen on the expression of MMP-9 and MMP-2 mRNA in the brain tissues and on the permeability of the blood brain barrier after cerebral ischemia and reperfusion

Objective To investigate the effects of hyperbaric oxygen(HBO)on the expression of MMP-9 and MMP-2 mRNA in the brain tissues after cerebral ischemia and reperfusion,and the permeability of blood brain barrier(BBB).Methods Using cerebral ischemia-reperfusion models with conscious mice,0.25 MPa(atmosphaera absolutus,ATA)HBO was applied 5 times during the reperfusion period,and 2%Evan's blue(EB)was injected into the tail vein 1 hour before the animals were sacrificed.The expression of MMP-9 and MMP-2 mRNA and EB content were determined by RT-PCR and spectrophotometry.Results The expression of MMP-9 and MMP-2 mRNA aswell as EB content significantly increased in the ischemia-reperfusion group as compared with a sham surgery group.The expression of MMP-9 and MMP-2 mRNA,and EB levels in the HBO group were similar to those in the sham surgery group.The expression of MMP-9 and MMP-2 mRNA and EB levels in the group given HBO plus reperfusion group were significantly decreased as compared with those in the group receiving reperfusion alone. Conclusion HBO can significandy reduce the expression of MMP-9 and MMP-2 mRNA and the permeability of the BBB.
4.Effects of hyperbaric oxygenation on the activity of antioxidation enzymes and the permeability of blood-brain barrier in mice with cerebral ischemia-reperfusion

Objective To explore the effect of hyperbaric oxygenation (HBO) on generation of free radicals and the permeability of blood-brain barrier(BBB) during cerebral ischemia-reperfusion(CIR). Methods Three hundred and twenty Kunming mice were randomly divided into four groups: a sham operation group, a HBO group, a CIR group, and a HBO + CIR group, with 80 mice in each group. Conscious mice were used to establish the CIR model, and 0.25MPa (ATA) HBO were applied 5 times after operations. The activities of SOD, CAT, GSH-PX and the concentration of MDA, EB in the cerebral hippocampal tissues in each group were measured with colorimetry. AIM: To investigate the effect of hyperbaric oxygen (HBO) on gelatinase,nitric oxide synthase and the permeability of brain blood barrier(BBB) in ischemia-reperfusion(I/R) mice. METHODS: Using cerebral I/R models, during the reperfusion period, 0.25 MPa (ATA) HBO were applied 5 times. matrix metalloproteinase(MMP)-2,9, nitric oxide synthase(NOS) and evans blue (EB) in brain were measured. RESULTS: ①HBO had significanty effect on MMP-9, but had little effect on MMP-2. ② HBO decreased the activity of NOS.③ The maxium amount of EB in IR group was at 4 hours after reperfusion and gradually decreased at 11 h, 23 h,48 h, 72 h. CONCLUSION: HBO may decrease the activities of MMP-9,NOS and the permeability of BBB in cerebral ischemia-reperfusion mice.
6.CT Diagnosis of Maxilla Sinus Malignant Tumor

Objective To analyze the role of CT scan in the diagnosis of maxilla sinus malignant tumor. Methods Data from CT scan in 17 patients were retrospectively analyzed. All cases were confirmed histologically. Results All cases manifested soft tissue mass in the sinus cavity and sinus wall bone destruction.The tumor could infiltrate out of maxilla sinus through destructive bone or nerve. Conclusion CT scan is an effective examination in diagnosis and evaluation of staging.
7.Association between WNK4 polymorphism and essential hypertension in Kazak population in China

Objective To identify the genetic variants of WNK4(with no K=lysine kinase)gene in Kazak population in Xinjiang province,to determine whether the WNK4 gene intron 10 polymorphism iS associated with essential hypertension(EH)and to investigate the distribution of genotype and allele frequencies of WNK4 gene. Methods One hundred and ninety-one patients with EH and 173 normal blood pressure controls were included in the study to assess the contribution of polymorphism of WNK4.Direct DNA sequencing was performed to identify the single nucleotide polymorphism(SNP)in 16 SUbjects with EH and 16 subjects of normotension(NT).Then the polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method was used for the detection of WNK4 genotype. Results One SNP located in the 10 intron of WNK4 (1156666 base of chromosome 17)was found in the Kazak population.The genotypes of the variants were found to be in Hardy-Weinberg equilibrium.GG,AG,AA genotypes were 88.0%,11.0 oA,1.0%in the EH group and 91.9%,8.1%,0%in the NT group,respectively.The frequencies of genotype and allele in EH group were not significantly different from NT group in Kazak population.Conclusions It suggests that the intron 10 polymorphism of WNK4 gene might be not associated with hypertension in Kazak population.
8.Changes and clinical significance of peripheral blood CD34+ cells in patients with acute cerebral infarction

Objective To determine the level of peripheral blood CD34 positive (CD34+) cells in patients with acute cerebral infarction (ACI),and to explore its clinical significance.Methods The level of peripheral blood CD34+ cells was determined by flow cytometry within 72 hours of onset of patients with acute cerebral infarction (infarct group,n=45),cerebrovascular risk factors in patients without cerebral infarction (high risk group,n=27) and healthy subjects (control group,n=20).The neural function defect score,infarction lesion volume and carotid artery intima-media thickness (IMT) were determined in patients with infarction group.Results The percentages of peripheral blood CD34 cells in infarction group (0.034 ±0.012)% and the high risk group of patients (0.047±0.009)% were lower than that of control group(0.063±0.009)%,and were lower in infarction group than in high-risk groups (all P<0.05).The percentages of peripheral blood CD34+cells were significantly decreased compared with control group (P<0.05) in infarction patients with mild [(0.047±0.009)%],moderate [(0.036±0.009)%],severe [(0.022±0.007)%] infarction nervous function defect score.Wherein,the percentages were lower in severe group than in the moderate group,moderate group was lower than in mild group (all P<0.05).The percentages of peripheral blood CD34 cells in infarction patients with small,moderate,large infaret lesion volume were lower than in control group (P<0.05),wherein,were lower in large group than in moderate group,lower in moderate group than in treatment group (all P<0.05).Infarction patients were confirmed with carotid atherosclerosis (CAS) by carotid ultrasound.The extent of lesion were divided into carotid artery intimal thickening group [(0.043±0.010)%],carotid artery plaque group [(0.036±0.010)%],and carotid artery stenosis group [(0.023±0.009)%].The levels of peripheral blood CD34+ cells in three groups of patients were decreased compared with control group.The levels were lower in carotid artery stenosis group than in carotid artery plaque group,lower in carotid artery plaque group than in carotid artery intimal thickening group (all P<0.05).Conclusions The level of peripheral blood CD34+ cells in acute cerebral ischemia is reduced,it can become a sensitive and early indicator of cerebral ischemia,and its level is related to neurologic impairment,infarction size and the degree of carotid artery atherosclerosis.
9.Technique Analysis of the DSA Value and Effectiveness in Hepatic Arteriography

Objective To analyze the correlation factors affecting DSA picture quality and to investigate the DSA value and effectiveness in hepatic arteriography. Methods 36 cases with liver tumor were analyzed through femoral artery intubatton with Seldinger and had hepatic arteriography with RH catheter. Results In the 192 images of 36 cases,the percentage of first-rate image was 92.7%,the second-rate image (9/192) was 5.3%,the third-rate image (5/192) was 2%. Conclusion All of the second-rate images and the third-rate images which were handled with selective image postprocessing can improve picture quality and can reach diagnosis request.

Total aralosides of Aralia elata (Miq) Seem (TASAES) from Chinese traditional herb Longya Aralia chinensis L was found to improve cardiac function. The present study was to determine the protective effects of TASAES on diabetic cardiomyopathy, and the possible mechanisms. Therefore, a single dose of streptozotocin was used to induce diabetes in Wister rats. Diabetic rats were immediately treated with low, medium and high doses of TASAES at 4.9, 9.8 mg/kg and 19.6 mg/kg body weight by gavage, respectively, for eight weeks. Cardiac function was evaluated by in situ hemodynamic measurements, and patch clamp for the L-type Ca2+ channel current (ICa2+-L) and transient outward K+ channel current (Ito). Histopathological changes were observed under light and electron microscope. The expression of pro-fibrotic factor, connective tissue growth factor (CTGF) was monitored using immunohistochemistry staining. Compared with diabetic group, medium and high doses, but not low dose, of TASAES showed a significant protection against diabetes-induced cardiac dysfunction, shown by increased absolute value of left ventricular systolic pressure (LVSP) and maximum rates of pressure development (+/-dp/dt(max)), and enhanced amplitude of ICa2+-L (P < 0.05). Histological staining indicated a significant inhibition of diabetes-caused pathological changes and up-regulation of CTGF expression (P < 0.05). The results suggest that TASAES prevents diabetes-induced cardiac dysfunction and pathological damage through up-regulating ICa2+-L in cardiac cells and decreasing CTGF expression.
