1.The effect of hyperuricemia on vascular complications of type 2 diabetes and the related-factors analysis

Objective To evaluate the effect of hyperuricemia on vascular complications of type 2 diabetes and analysis of the possible mechanism. Methods 586 cases with T2DM were divided into two groups: normal serum uric acid (n=397) and hyperuricemia (n=189).The morbidity of macrovascular and microvascular complications and the serum levels of CRP,TNF,IL-1,IL-2,IL-6,IL-8 and PAI-1 were detected. Results (1)Hyperuricemia group showed higher BMI, blood pressure, TG,TC,LDL,WHR ratio and lower serum HDL-C than the group with normal uric acid (all P<0.05).(2)Group with hyperuricemia showed higher morbidities of macrovascular and microvascular complications than the group with normal uric acid (all P<0.05).(3) Group with hyperuricemia had higher levels of CRP,TNF,IL-6,IL-8,PAI-1 (all P<0.05). Conclusions (1) Hyperuricemia is associated with obesity and dyslipidemia.(2) Hyperuricemia may cause atherosclerosis by inflammation of vascular endothelium, resulting macrovascular and microvascular complications.
2.Clinical Observation of Minocycline Hydrochloride Combined with Tinidazole in the Treatment of Chronic Periodontis

OBJECTIVE:To observe the efficacy and safety of minocycline hydrochloride combined with tinidazole in the treat-ment of chronic periodontitis. METHODS:100 patients with chronic periodontitis were randomly divided into observation group 1, observation group 2,observation group 3 and control group. Patients in control group were given full-mouth dental scalers and sub-gingival clean scraping,then 0.9% Sodium chloride injection and 3% hydrogen peroxide were used to alternately wash periodontal pocket bottom. Based on the treatment,observation group 1 was injected Minocycline hydrochloride ointment to periodontal pocket bottom until slight overflow after saliva isolation and desiccation,once a week;observation group 2 was given Tinidazole buccal tablet 1 tablet in the buccal middle gingival surface,twice a day,observation group 3 was given Minocycline hydrochloride oint-ment+Tinidazole buccal tablets(the usage and dosage was the same with above-mentioned 2 groups). The treatment course was 4 weeks. The clinical efficacy,PLI,PAL,PPD,MD,GI before and after treatment and incidence of adverse reactions were observed. RESULTS:The total effective rate in observation group 3>observation group 1 and observation group 2>control group(P<0.05), however,there was no significant difference between observation group 1 and observation group 2(P>0.05). 3.Rapid detected of trisomy 21 syndrome by gene diagnosis techniques

Objective Setting a rapid, simple and accurate gene diagnosis method for trisomy 21 syndrome. Methods 4 short tandem repearts (STRs) loci (D21S11, D21S1411, D21S1412, D21S1414) in and near the core region(21q22.1-21q22.2) on 21 chromosome of blooding samples from 8 normal people, 11 cases of trisomy 21 syndrome and 1 case of fetus umbilical core were analyzed and detected by polymerase chain reaction(PCR), polyacrylamide gel electrophories, and silver staining. Results 6/8? 7/8? 3/8?7/8 of normal people showed two bands with a DNA content ratio of 1∶1. 2/8?1/8?5/8?1/8 of normal people showed one band in 4 loci. 7/11?8/11?6/11?7/11 of all patients showed two bands with a DNA content ratio of 2∶1;3/11?2/11?2/11?3/11 showed three bands with a DNA content ratio of 1∶1∶1. 1/11?1/11?3/11?1/11 showed one band. The blooding sample of fetus showed two bands with a DNA content ratio of 2∶1 at 3 loci and one band at 1 locus. 11 patents and 1 fetus were identified as trisomy 21 syndrome by analysis combining with 4 STR loci. Conclusion The 4 selected STR loci have high polymorphism.4 polymorphotic STR is a valuable gene marker for diagnosis of trisomy 21. Trisomy 21 can be diagnosed rapidly and accurately within 24 hours by PCR.
4.Analysis of the curative effect of Yunnan Baiyao combined with Wujia Shenghua capsule on drug abortion

Objective To explore the effect of abortion, effect of Yunnan Baiyao Combined with Wujia Shenghua capsule in patients with medical abortion vaginal bleeding time.MethodsSelect 98 cases from March 2014 to May 2016 by drug abortion in second hospital of shandong university, were randomly divided into combined group and control group with 49 cases in each group, two groups were given routine drug treatment, combined group was given Yunnan Baiyao combined with Wujia Shenghua capsule, comparing the two groups of patients with abortion, vaginal bleeding time.ResultsCombined group and control group differences in the complete abortion rate was not statistically significant, combinde group of pregnant sac discharge time is shorter than the control group (P<0.05);combined group with the duration of vaginal bleeding, vaginal bleeding and vaginal bleeding in 7~14 days>14 days the percentage of patients were lower than those in control group (P<0.05);combinde group of vaginal bleeding (16.33%), the amount of menstrual and menstrual amount (59.18%), > menstruation (24.49%) and the control group (12.24%, 38.78%, 46.94%) with significant difference (P<0.05);combined group of menses time, menstrual duration compared with the control group, the difference was not statistically meaning.ConclusionYunnan Baiyao combined with Wujia Shenghua capsule can accelerate the gestational sac discharge, shorten the time of vaginal bleeding, reduce the amount of vaginal bleeding.

6.Study on Formulation Optimization and Drug-release Mechanism of Cilnidipine Sustained-release Tablet

OBJECTIVE:To optimize the formulation of Cilnidipine sustained-release tablet,and study its drug-release mecha-nism. METHODS:Solvent method was adopted to prepare the cilnidipine solid dispersion,then Cilnidipine sustained-release tablet was prepared by using hypromellose K4M(HPMC K4M)as release material. Using comprehensive scores of cumulative release de-gree in 2,6,12 h as indexes,single factor method and Box-Behnken response surface method were used to screen the amounts of HPMC K4M and ethyl cellulose (EC),lactose-microcrystalline cellulose (MCC) ratio in formulation of Cilnidipine sustained-re-lease tablet,and verification test was conducted. The drug-release mechanism of Cilnidipine sustained-release tablet was investigat-ed by model fitting way. RESULTS:The optimal formulation was as follow as 25% of cilnidipine solid dispersion,30% of HPMC K4M,10% of EC,lactose-MCC ratio of 1:1(m/m). The adhesive was 5% PVPP ethanol solution and the lubricant was 0.5%magnesium stearate. The cumulative release degrees of prepared sustained-release tablet in 2,6,12 h were(21.4±3.3)%,(62.9± 2.8)%,(85.4±0.5)%(n=3),relative error of which to predicted value 25%,60%,90%were 14.4%,4.8%and 5.1%. Release curve showed the highest fitting degree with the first-order release model,conforming to non-Fick diffusion. CONCLUSIONS:Cil-nidipine sustained-release tablet with sustained-release effect is successfully prepared by optimized formulation.
7.Clinical study of reattribution-cognitive-pharmacy model in the treatment for irritable bowel syndrome

Objective To investigate clinical effect of reattribution-cognitive-pharmacy model (RCPM) in the treatment for irritable bowel syndrome(IBS). Methods 125 subjects with diarrhea predominant irritable bowel syndrome (IBS-D) were divided into two groups randomly. 62 patients in group A were treated with 10 ~ 20 mg of paroxetine without any other medication or psychological interview and 63 patients in group B received RCPM with interviewing once a week for 6 sessions and took 10 ~ 20 mg of paroxetine in the same way as group A after a week. The effect was evaluated at the end of 4 weeks and 12 weeks by a questionnaire. Results At the end of 4 weeks,29 patients in group A reported a reduction in abdominal pain,and 28 reported a reduction in stool frequency ,and 12 patients stopped taking paroxetine because of worrying about those side effect . In group B 48 reported a reduction in abdominal pain ,and 42 reported a reduction in stool frequency ,and 3 patients stopped taking paroxetine. At the end of 12 weeks,36 patients in group A reported a reduction in abdominal pain ,and 30 reported a reduction in stool frequency,and 14 patients stopped taking paroxetine because of worry about those side effect. In group B,54 cases reported a reduction in abdominal pain,and 45 reported a reduction in stool frequency,and 5 patients stopped taking paroxetine because of no obvious improvement. Conclusion RCPM can alleviate the abdominal pain and bowl movement frequency of IBS-D,and it seems better than paroxetine treatment alone. RCPM can improve compliance of paroxetine in patients with IBS-D.
8.Association between polymorphism of NUDT15 gene and hepatotoxicity induced by 6-MP in children with acute lymphoblastic leukemia.

OBJECTIVE:
To investigate the association between single nucleotide polymorphism of NUDT15 gene (SNP rs116855232) and hepatotoxicity in children with acute lymphocytic leukemia (ALL).
METHODS:
A total of 135 children with ALL in Shandong Province were recruited in this study, and patients were divided into two groups based on the presence of liver injury. Genotypes of each patient were detected using PCR and Sanger sequencing. Clinical data and the average dose of 6-mercaptopurine (6-MP) were collected and analyzed by SPSS 19.0 software.
RESULTS:

Objective To study the relationship and prognosis between E-cadherin gene expression and lymphatic hyperplastic reaction in gastric cancer. Methods The degree of lymphocytosis and draining lymph node from 86 cases of gastric cancer were observed and the expression of E-cadherin gene in gastric cancer were detected by SP method of immunohistochemistry (IHC). Results Lymphocyte infiltration degree around gastric cancers was positively related to the reactive hyperplasia of the lymphnodes and was inversely related to lymphatic metastasis. The expression of E-cadherin has relationship with the infiltration degree of stomach carcinoma. To compare with T1/T2 and T3/T4 phases, the result is significantly different (P <0.01).Meanwhile, the expression of E-cadherin was positively correlated with tumor-infiltrated lymphocytes and reactive hyperplasia of the lymph nodes, was negatively correlated with lymphatic metastasis in drainage area.Conclusion The over-expression of E-cadherin gene is significantly related to lymphoproliferation and lymph node metastasis. The abnormal expression of E-cadherin can be used as an index to determine prognosis of gastric carcinoma.
10.Expression of cysteine rich 61 and vascular endothelial growth factor in patients with multiple myeloma and their relationship with staging and therapeutic response

Objective To investigate the expression of cystein rich 61 (Cyr61) and vascular endothelial growth factor (VEGF) in different stages of multiple myeloma (MM) patients,evaluate their relationship with angiogenesis and prognosis,and to determine the relationship between Cyr61 and VEGF.Methods Expression of Cyr61 and VEGF in BMMNC from 31 patients with different stages of MM and 10 cases of control were detected by RT-PCR.Results Expression of Cyr61 and VEGF in patients with MM (0.3652±1.5423,0.4815±1.3423) were significantly elevated in comparison to control (0.1862±0.7542,0.2012±1.2331) (P < 0.05,P < 0.01).Expression of Cyr61 and VEGF in stage Ⅲ (0.4632±0.1634,0.5356± 0.2342) was significantly higher than those in stage Ⅰ and stage Ⅱ (t =2.423,2.524,P < 0.05),but there was no difference between stage Ⅰ and stage Ⅱ (0.2513±0.1365,0.3064±0.2124; 0.3084±0.2254,0.3653±0.1265) (t =1.782,1.824,P > 0.05).The levels of Cyr61 and VEGF were significantly decreased after chemotherapy compared to before chemotherapy (P < 0.01).Expression of Cyr61 and VEGF were positively correlated (r =0.8941,P < 0.01).Conclusion Cyr61 and VEGF may play roles in the angiogenesis and pathophysiology of MM.It can be used to guide treatment and estimate prognosis by monitoring Cyr61 and VEGF.
