Objective:To identify potentially pleiotropic genes for lean body mass ( LBM ) and age at menarche ( AAM).Methods:The discovery sample consisted of 1 692 unrelated female subjects of European ancestry.The replication sample consisted of 801 unrelated female subjects of Han Chinese ancestry.A total of 909,622 single nucleotide polymorphisms (SNPs) were genotyped in both samples with the Affymetrix genome-wide genotyping array SNP 6.0.Bivariate genome-wide association analyses were then performed to the appendicular LBM and AAM.Results: Two SNP rs1860547 and rs11030746 identified by the bivariate GWAS were significant at the genome-wide significance (GWS) level;their P-values were <0.05 after replications.In the upstream of rs1860547, two genes KCNA1 and KCNA5 were found to be important for both LBM and AAM.In the downstream of rs11030746, one gene KCNA4 was found.Univariate GWAS also identified both SNPs to be significant at the GWS level; their P-values were <0.05 after replications.In the upstream of rs1860547 , two genes KCNA1 and KCNA5 are found to be important for LBM.In the downstream of rs11030746 , one gene KCNA4 was found.Conclusion:KCNA1, KCNA4 and KCNA5 are likely to be pleiotropic genes closely related to both LBM and AAM in European females.

Objective To investigate the association of the polymorphism of serotonin transporter gene linked polymorphic(5-HTTLPR) region with depression and its clinical symptoms in the Han Chinese.Methods PCR-based technique was used to genotype 5-HTTLPR in 64 patients with major depression and 67 unrelated controls,and the association between the polymorphism and clinical symptoms of depression was explored according to HAMD score.Results Fewer patients tended to be of L/L genotype(9.4%) than controls(19.4%),but there was no significant difference.Among women,fewer patients tended to be of L/L genotype(2.9%) than controls(13.9%),but there was no significant difference.The symptom analysis showed that depressive symptoms were significantly associated with 5-HTTLPR genotypes or alleles in the patients of L/L genotype,and that L allele had significantly lower depressive scores compared with those of other genotypes or alleles(P

Objective To investigate relationship between Ala 54 Thr polymorphism in the intestinal fatty acid-binding protein (I-FABP) and plasma lipids in old people.Methods We collected fasting blood samples of selected groups,then we extracted DNA from whole blood,and detected lipids from serum.The 54A/T I-FABP genotypes were analyzed in 72 Han Chinese elderly by polymerase chain reaction (PCR) and DNA restriction enzyming.Total cholesterol (TC),triglyceride (TG),high density lipoprotein (HDL-C) and low-density lipoprotein (LDL-C) were detected.Genotype subgroups were Thr54 (-) and Thr54 (+) groups (36 cases for each).Results Compared with Thr54 (-) genotype group,Thr54 (+) genotype group had higher levels of plasma TC[(4.50±0.73) mmol/L vs.(5.48±0.49)mmol/L],TG[(1.08±0.48) mmol/L vs.(2.02±0.53)mmol/L],LDL-C [(3.10±0.44) mmol/L vs.(3.50±0.66)mmol/L],and had lower level of HDL-C [(1.14±0.25) mmol/L vs.(0.96±0.23)mmol/L](t=-6.67,-7.84,-3.03,3.05,all P＜0.05).

Objective To study the influence of qiqishen granules on the cellular and humoral immune functions in model mice. Methods Six-week-old mice were divided into control group and qiqishen granule (high, medium and low dose) groups. The suspension of chicken red blood cells was injected into the mouse abdominal cavity. The influence of qiq-ishen granules on the phagocytic function of the macrophages in mouse abdominal cavity was observed. The sheep red blood cells (SRBC) were prepared. The blood corpuscle coagulation was observed, and the serum hemolysin was detected. The ac-tivity of the mouse natural killer (NK) cells were detected by the interaction between the target cell (YAC-1) and spleen cell (the response cell). The influence of qiqishen granules on the cellular immunity was detected by the lymphocyte transforming assay. The influence of qiqishen granules on organ/body weight ratio was measured by calculating the thymus/body and spleen/body ratio. Results Compared with the control,qiqishen granules significantly improved phagocytic function of the macrophages in mouse abdominal cavity. The humoral immune function was also improved in mice. The activity of NK cell was enhanced by qiqishen granules, which also enhanced the lymphocyte transforming induced by the concanavalin (Con) A. There was no significant influence in the organ/body weight ratio. Conclusion The qiqishen granules could increase the im-munomodulating effect, indicating it would have a good using prospect.

Objective To explore the relationship between FABP2 54 gene polymorphism and serum lipid levels in different ethnic patients with coronary heart disease.Methods We collected fasting blood from participants,then extracted DNA from whole blood,and detected lipids from serum.We performed 54A/T FABP2 genotyping for participants in the case and the control groups by polymerase chain reaction (PCR) and DNA restriction enzyme digestion techniques.Results ( 1 ) In Hans population with coronary heart disease,the allelic frequency of point in 54T gene was 0.542,and 54A gene was 0.458 ; in urban Mongolian with coronary heart disease,the allelic frequency of point in 54T gene was 0.708,and that of 54A gene was 0.292.The allelic frequency of point in 54T and 54A genes were 0.284 and 0.716 respectively in the Han population control group and 0.353 and 0.647 respectively in the normal urban Mongolian group.Compared with the control groups,mutant 54T allele frequency in the case groups of Han and urban Mongolian was significantly higher ( Han:x2 =14.967,P ＜ 0.05 ; Mongolian:x2 =28.083,P ＜ 0.05 ).Compared with the Han Chinese population with coronary heart disease,the mutant 54T allele frequency in urban Mongolian with coronary heart disease was significantly (x2 =7.111,P ＜ 0.05 ).There was no significant difference in mutant 54T allele frequency between the Mongolian control group and the Han control group ( x2 =3.392,P ＞ 0.05 ).( 2 ) Patients with coronary heart disease who carry Thr54 ( + ) had a significantly higher level of fasting plasma TG [ CC Thr54 (-):( 1.89 ± 0.57 ) mmol/L,CC Thr54 ( + ):( 3.92 ± 1.63 ) mmol/L; CM Thr54 (-):(2.23 ± 0.13 ) mmol/L,CM Thr54 ( + ):(4.03 ± 1.14) mmol/L; Hans:P =0.001,Mongolian:P =0.035 ],and LDL-C [ CC Thr54 (-):( 3.09 ± 0.92) mmol/L,CC Thr54 ( + ):(4.05 ± 1.14 ) mmol/L ; CM Thr54 (-):( 4.26 ± 0.08 ) mmol/L,CM Thr54 ( + ):( 5.10 ± 0.56 ) mmol/L; Hans:P =0.025,Mongolian:P =0.045 ] than those with Thr54 (-).Conclusion (1) Threre is FABP2 gene polymorphism in urban Mongolian and Han population.Gene mutation frequency is in accordance with Hardy-Weinberg law of genetic equilibrium,suggesting the sample group representative.(2) FABP2 polymorphism may have a certain contribution to dyslipidemia in patients with coronary heart disease.FABP2 gene polymorphism may be associated with coronary heart disease.Individuals of Mongolian with FABP2 Thr54( + ) has a higher risk of coronary heart disease.

Objective To evaluate the normalized correction effect of the ROX reference fluorescence on the real-time quantitative RT-PCR assessment for the Nrf2 mRNA.Methods The Taq-man probe was utilized in the real-time quantitative RT-PCR method to assess the Nrf2 mRNA; The reaction specificity of the Nrf2 PCR products obtained from the rat cDNA was assessed using gel electrophoresis and sequencing identification.The PCR calibrators with different concentrations varied at 10 gradients(from 2.0 × 109 to 2.0 × 100) were prepared for real-time quantitative RT-PCR assessment,and then the influence of the ROX reference fluorescence correction on the linear range of the PCR standard curve was investigated experimentally.The recombinant plasmids with different concentration levels(e.g,high,medium or low concentration) were assessed by the real-time RT-PCR and 20 replicate measurements were performed for all the samples in both intra- and inter-assays.The repeatability differences of both the inter- and intra-assays were analyzed systemically with/without the ROX reference fluorescence correction.Results The primers and probes required for Nrf2 mRNA real-time quantitative RT-PCR assessments were constructed,and the reaction system and conditions were also established.The length of the amplified fragment agreed well with the expected length(122 bp),and the sequencing analysis showed the amplified fragment was verified by the sequencing identification.After applying the ROX reference fluorescence correction,the linear range ofthe PCR standard curve was measured as 2.0 × 109 -2.0 × 100 copies/μl(R2 ＞ 0.99),which was 100times wider than that measured without ROX reference fluorescence correction(2.0 × 109 - 2.0 × 102copies/μl,R2 ＞ 0.99).Without the ROX reference fluorescence correction,the intra-assay CVs of the Ct value were measured as 4.1%,2.7% and 2.1% for the recombinant plasmids with high-,medium-,and low-level concentration,respectively.The inter-assay CVs of the Ct value were 4.3% 、3.0% 、2.4%.By contrast,with the ROX reference fluorescence correction,the intra-assay CVs of the Ct value became 0.7%,0.5%,0.4% and the inter-assay CVs of the Ct value turned to 1.0%,0.8% and 0.7%.The discrete degree of the Ct value with the correction was lower than that without the correction.The intra- and interassay Ct values of the high- and medium-level samples exhibited statistic significance for the measurements with/without the correction.In the high-level group,the t values of the intra- and inter-assays were 4.843 and 2.566,with P＜0.05.In the medium-level group,the t values of the intra- and inter-assays were 4.293 and 4.423,with P＜0.05.However,no statistic significance was observed the low-level group(the t values of the intra- and inter-assays were 0.753 and 1.279,with P ＞.0.05).Conclusions The utilization of the ROX reference fluorescence in the real-time quantitative PCR assessment for Nrf2 mRNA could widen the linear range of the standard PCR calibration curve and improve the repeatability of the assessment,which should be quite helpful for accurately assessing the expression level of the Nrf2 mRNA.

Methods In the process of schistosomiasis screening by IHA,the written records,electronic records,and se-rum sample bank were combined to make comprehensive archives. Results The S. japonicum antibody IHA test archives can pre-serve the schistosomiasis screening data in the long term and even can trace the source of experiments,and the operation was sim-ple. Conclusion The archives of S. japonicum antibody IHA tests are simple and useful,and worth of popularization.

Objective To grasp bilirubin in the quality control serum and calibration items in the same batch the variation law of the designed to ensure that the test quality and the use of reasonable quality control serum and calibration items ,control testing cost .Methods laboratories use The double level quality control serum and calibration items after dilution is divided into five groups ,with automatic biochemical analyzer test for 12 weeks and recorded results of TBIL ,DBIL .use SPSS 17 .0 to calculate the mean ,standard deviation ,coefficient of variation ,Normal distribution analysis and make the results trend line chart to observe any changes .Results RANDOX normal concentration quality control serum TBIL and DBIL values variation RCV% 26 .0% ,48 .2% , more than CLIA bilirubin projects allow 1/3 of the total error value(6 .67% ) .Normal distribution test ,P value was 0 .006 ,0 .012 , less than 0 .05 ,do not obey the normal distribution ,test results line chart is on the decline .LEADMAN high concentration quality control serum and bilirubin calibration items TBIL and DBIL variation RCV% <6 .67% ,normal distribution test all P>0 .05 ,obey the normal distribution ,test results no trend line chart .Conclusion Through the experiment observation to the quality control ser‐um and calibration items bilirubin in the variation law of 12 weeks .Experimenter can be used according to the actual situation to ad‐just ,not only to ensure the quality of the bilirubin test ,and to get the most out of the quality control serum and calibration items use efficiency ,reduce the test cost .

Objective To understand the changes of positive rates of IHA detections of outpatients in schistosomiasis clinic. Results The data of IHA detections of outpatients in schistosomiasis clinic in Hubei Provincial Center for Disease Control and Prevention were collected and analyzed statistically from 2005 to 2014. Results A total of 7 113 outpatients were detected by IHA test,and 547 of them were positives with a positive rate of 7.69%. The positive rate of IHA test was on an upward slope be?fore 2008,and the rate reached 14.85% in 2008,which was significantly higher than that in 2005(5.81%)( χ2 = 47.40,P<0.01),then it was on a declined stage after 2008,and the positive rate decreased to 3.76 in 2014,which was significantly lower than that in 2008( χ2 = 12.29,P<0.01). The positive rate of outpatients in the 10~<30 years age group was higher than those in other age groups(all P < 0.012 5),and the male positives were more than the female ones. Conclusions The schisto?somiasis endemic situation has been significantly decreased in Hubei Province. The male and people in 10~<30 age group are the high risk groups,so the targeted health education should be strengthened.

Objetives To investigate the outcomes of sirolumus-eluting stent(CypherTM,Cordis/Johnson&Johnson,Warren,NJ,USA)and paclitaxel-eluting stent(TaxusTM,Boston Scientific,Natick,MA.USA)in the treatment of coronary heart disease after a four.year clinical follow-up.Methods 237 consecutive patients were enrolled in this study and treated with Cypher(136 patients)or Taxus(101 patients)from January to October 2003.111e rates of stenosis.stent thrombosis according to ARC definition and major adverse cardiac events(MACE a composite of cardiac death,myocardial infarction and target vessel revascularization)were analysed.Results There was no significant difference on secondary restenosis.target lesion revascularization(TLR)and MACE between Cypher and Taxus groups at six.month angiographic follow-up,but late luminal loss was higher in Taxus group[(0.15±0.43)mm vs(0.42±0.34)mm,P=0.022].At four-year follow-up,TVR-free survival rate was 88.97% in Cypher group versus 82.28% in Taxus group(P=0.158).MACE.free survival rate was 83.8% in Cypher group and 79.2% in Taxus group(P=0.056).The incidence of stent tllrombosis was no difference between the two groups(1.47% vs 1.98%).There was also no difference among early(0 vs 0.99%),late(0.73%vs0.99%)and very late stent thrombosis(0.73%vs 0)between the 2 groups.Conlusions There were nodifference between Cypher and Taxus in the treatment of coronary artery disease:Both Cypher and Taxus have good clinical outcomes except that Taxus had highcr late loss.
