1.Clinical features of OSAHS closely associated with supine position

OBJECTIVE To study the clinical features of OSAHS patients closely associated with supine position. METHODS The clinical data of 105 patients with OSAHS diagnosed by overnight polysomnography(PSG)were retrospectively studied. The patients were divided in two groups: the positional patients(PP)group, those patients who had a apnea-hypopnea index(AHI) that was at least two times higher than the lateral AHI, and the non-positional patients(NPP)group, those patients in whom the AHI in the supine position was two times less than that in the lateral position, and the two groups were compared with age, BMI and the severity of OSAHS. RESULTS The difference of age and BMI were not significant between the two groups[(40.4?10.5)year versus(37.0?10.6) year,(28.2?3.8)kg/m2 versus(29.1?3.0)kg/m2]. There was significantly different in AHI between two groups[(36.8?25.4)/h versus (51.2?29.0)/h ]. AHIsupine and AHInot-supine was significantly different in mild and moderate OSAHS patients [(23.9?13.8) /h versus(12.4?10.1)/h ]. CONCLUSION Mild and moderate OSAHS patients are mainly PP. Positional therapy should play an important role in treatment of mild and moderate patients.
2.Determination of drug release in vitro of atractylenolide Ⅰ liposome by RP-HPLC

Objective To establish a RP-HPLC method for the determination of drug release in vitro of atractylenolide Ⅰ liposomes. Methods The release behavior of the drug from liposomes was studied by the third method for dissolution. ZORBAX C18 column (4.6 mm × 250 mm, 5 μm) was used with a mobile phase of Methanol-Acetonitrile-0.2% from liposomes in vitro fitted the log-normal distribution equation and had a property of sustained release. Conclusion The method is simple, fast and selective. It is suitable for the determination of release profile in vitro of atractylenolide Ⅰ liposomes.
3.The ReIationship of Mean PIateIet VoIume and Sudden SensorineuraI Hearing Loss

Objective Our aim was to investigate the association between sudden sensorineural hearing loss (SSHL)and mean platelet volume (MPV)values,in order to evaluate the etiology of SSHL.Methods This study included 40 patients with SSHL in our ward between Jan.2012 and Jan.2014 and 30 age- and sex-matched healthy subjects were included as controls.The correlation among the levels of the number of platelets,mean plate-let volume(MPV),platelet distribution width (PDW)was evaluated in the two groups.ResuIts The platelet count was significantly lower in the SSHL than the control group(212.0±47.9 versus 237.8±46.2 ×109/L).MPV and PDW were significantly higher in SSHL(10.9±0.8 versus 10.3±0.8 fL;12.9%±2.0% versus 12.0%±1.3 %). The SSHL patients were divided two subgroups:the mild-moderate subgroup (hearing loss 25~70 dB)and the se-vere subgroup(hearing loss≥70 dB).The MPV values were insignificantly different between the two subgroups(11. 0±1.1 fL versus 10.8 ±1.0 fL).Compared to the control group,the two subgroups had both significant differ-ences in MPV.ConcIusion High MPV values are a risk factor for thrombotic vascular diseases.Considering the in-creased MPV levels,SSHL appears to be characterized by ischaemic or thrombotic events.
4.Effect of erigeron on intercellular adhesion molecule-1 and its mRNA expression during cerebral ischemia and reperfusion in rats

AIM: To investigate the effect of erigeron on intercellular adhesion molecule-1 (ICAM-1) and mRNA expression during cerebral ischemia/reperfusion. METHODS: The rat models of middle cerebral artery (MCA) focal cerebral ischemic reperfusion were established with the suture method in the study. The ICAM-1 mRNA and protein expression were measured by RT-PCR and immunohistochemistry techniques, respectively. RESULTS: By down-regulating the expression of ICAM-1 protein and mRNA and alleviating inflammation in cerebral ischemic region, erigeron exerted a protective effect in cerebral ischemia and reperfusion. CONCLUSION: The results suggest that erigeron protects the brain against cerebral ischemia and reperfusion injury via inhibiting ICAM-1 expression. [
5.Analysis the relationship of mean platelet volume and nasal obstructive disease.

OBJECTIVE:
High mean platelet volume (MPV) values are a risk factor for cardiovascular diseases. Our aim was to investigate the association between nasal obstructive disease and MPV value.
METHOD:
This study included 40 patients with nasal obstructive disease and 30 age-and sex-matched healthy subjects as controls. The levels of the number of platelets, MPV, platelet distribution width (PDW) were evaluated and compared in the two groups. Then the MPV was compared in the patients before and after operation in the study group.
RESULT:
Platelet count is significantly lower in the study group than the control group [(215.0 ± 47.5) x 10(9)/L versus (237.8 ± 46.2) x 10(9)/L]; MPV and PDW is significantly higher in the study group [(10.7 ± 0.7) fL versus (10.3 ± 0.8) fL]; [(12.9 ± 2.0) % versus (12.0 ± 1.3) %]. Comparing to preoperation, MPV values of the study group is significantly decreased after nasal obstructive operations [(10.7 ± 0.7) fL versus (10.4 ± 0.6) fL].
CONCLUSION
MPV levels are elevated in patients with nasal obstructive disease, and can be decreased by nasal obstructive operation.

6.Analysis the relationship of mean platelet volume and obstructive sleep apnea and hypopnea syndrome.

OBJECTIVE:
Uur aim was to investigate the association between OSAHS and mean platelet volume (MPV) value.
METHOD:
This study included 70 cases with OSAHS in our ward between Jan. 2012 and Jan. 2014, and the OSAHS patients were divided two groups: mild- to moderate group and severe group; 30 age-and sex-matched healthy subjects was in control group. The correlation among the levels of the number of platelets (PLT), MPV, platelet distribution width (PDW) were evaluated in the two groups.
RESULT:
PLT count was significantly lower in the severe group than the control group [(202. 8 ± 68. 9] × 10(9)/L, (235. 9 ± 65. 2) × 10(9)/L]; MPV and PDW were significantly higher in the severe group [(10. 9 ± 0. 9), (10. 4±0. 8) fL; (12. 9 ± 1. 9) %, (12. 0 ± 1. 4) %]. There was not significantly difference of MPV between the mild to moderate group and the control group [(10. 7 ± 0. 7), (10. 4 ± 0. 8)fL].
CONCLUSION
MPV levels are elevated in patients with the severe OSAHS.

7.Expression and clinical significance of programmed cell death ligand 1in liver tissues of patients with chronic hepatitis B virus infection

Objective To investigate expressions of programmed cell death ligand 1 (PD-L1) in hepatic tissues at the different stages of hepatitis B virus ( HBV) infection, and clarify its role in the mechanism of chronic hepatitis B virus infection. Methods The expressions of PD-L1 were detected by immunohistochemistry and computer image quantitative analysis in the hepatic tissues of 65 chronic HBV infected patients and 5 healthy controls. The correlations between PD-L1 expression and inflammatory grading in the hepatic tissues, total bilirubin (TBil), alanine aminotransferase (ALT), aspartate aminotransferase (AST), serum HBV DNA level were analyzed. Results The PD-L1 expressions in hepatic tissues of HBV infection with G0 - G4 inflammatory grades were 3. 07 % ±0.93%, 8.01%±1.49%, 11.60%±2.60%, 18.41%±2.21% and 26. 04% ±3. 41%, respectively,which were all significantly stronger than that in controls (0. 64%±0. 28%). PD-L1 expression was a positively correlated with inflammation grading of hepatitis tissues, TBil, ALT and AST level in serum (r=0. 917, 0. 787, 0. 483, 0. 628; all P<0. 05), and negatively correlated with serum HBV DNA load (r=-0. 620, P<0. 05). Conclusion The upregulated PD-L1 expression may be probably involved in the chronicity of HBV infection.
8.Factors Influencing Facial Profile Evaluation

Objective To characterize differences within facial profile esthetic evaluations among three groups of evaluators and the influences of sagittal or vertical skeletal patterns on facial profile esthetics. Methods A total of 114 silhouettes,from the pre?or post?treatment facial profiles of 57 patients,were scored by three groups of evaluators(professionals,patients'parents,and laypersons)to characterize differences within facial es?thetics evaluation among evaluators. The highest?scoring 30 silhouettes and the lowest?scoring 30 silhouettes were selected as the high score group and the low score group,respectively. Sixteen indices of the relative lateral cephalometric films were measured. Data were analyzed by SPSS 21.0. Results Facial profiles following orthodontic treatment scored higher than pre?treatment(P<0.05). There were significant differences between professionals and laypersons in facial profile scoring. Different sagittal skeletal patterns had no effect on esthetic evaluation ,but lower facial height significantly influenced whether a patient received a high or low esthetic score(P<0.05). Conclusion Orthodontic treatment can improve the fa?cial profile significantly,and lower facial height is the most sensitive index for facial profile evaluation in our study. However,all three evaluator groups have different esthetic standards that don't overlap. Notably,the expectations of laymen are higher than those of orthodontists.
9.Effect of methotrexate packaged by tumor derived microparticles combined with radiotherapy on proliferation of cancer stem cells in lung adenocarcinoma in vitro

Objective To explore the effect of methotrexate packaged by tumor derived microparticles (T-MP MTX) combined with radiotherapy on lung cancer stem cell (CSC) in vitro. Methods T-MP MTX was prepared from non-small cell lung cancer A549 cells. Proliferative changes of A549 cells, bronchial epithelial cells H460 and 16HBE cells treated by T-MP MTX were assayed by MTT method. Cell cycles of A549 cells in blank group and T-MP MYX group were examined by fluorescence activated cell sorting (FACS). The effect of T-MP MTX combined with radiotherapy on CSCs was assessed by tumor sphere formation experiment and animal experiment. The expressions of stemness relative genes (such as β-catenin, Nanog, SOX-2 and KLF4) were measured by Western blot. Results T-MP MTX dose-dependently inhibited the cell growth in A549 cells, but didn't in H460 cells and 16HBE cells. The S cycle ratio of A549 cells in blank group and T-MP MYX group measured by FACS were (15.83±3.14)%and (47.47±6.69)%, respectively. S cycle ratio of T-MP MYX group was notably higher compared with that of blank group (t=7.411, P=0.002). Further study revealed that the number of tumor sphere in blank group, control group, 2 Gy group, 4 Gy group and 6 Gy group was (268.9±22.4), (172.4±18.7), (48.3±5.1), (16.3±3.5) and (5.1±3.1), respectively. The number of tumor sphere in other groups was decreased compared with that in blank group (F=228.291, P=0.000). The numbers of tumor sphere in 2 Gy group, 4 Gy group and 6 Gy groups was also reduced compared with that in control group. Importantly, the number of tumor sphere in these groups were decreased dramatically as the dose of radiotherapy increased (F=95.142, P=0.000). The results of tumor sphere volume were similar with the number of tumor sphere. Western blot experiment showed that T-MP MTX treatment in A549 cells decreased the expression of stemness relative genes (β-catenin, Nanog, SOX-2 and KLF4), and its role was reinforced when radiotherapy was combined. Animal experiment implied that activity of luciferase in T-MP MTX group was decreased compared with that in blank group (P=0.000), and the activity of luciferase in T-MP MTX plus 2 Gy group was reduced significantly (t=6.887, P=0.002). Conclusions T-MP MTX has a potential to sensibilize radiotherapy, and it will synergistically inhibit the proliferation of CSCs when combined with radiotherapy. Moreover, its mechanism may be related with T-MP MTX activating CSCs from hypometabolism state and blocking process of cell cycle.
10.Extraction Conditions Optimization and Content Determination of Gallic Acid in Wudang Pineneedle Tea by Orthogonal Test

Objective:To optimize the extraction conditions of gallic acid inWudang pineneedle tea and establish the assay meth-od. Methods:The content of gallic acid was determined by RP-HPLC-DAD. Orthogonal test was used to optimize the extraction condi-tions of gallic acid in Wudang pineneedle tea on the base of single factor tests. The optimal extraction conditions were as follows:adding 50-fold amount of water, and ultrasonic extracting for 60 min under 200W ultrasound power. Results:The linear range of gallic acid was 5. 10-51. 00 mg ·L-1(r=0. 9993), and the average recovery was 98. 7% (RSD=1. 93%, n=6). The average content of gallic acid in the samples was 0. 35 mg·g-1 . Conclusion: The optimal extraction conditions are simple and feasible, and the assay method is stable and reliable, which can be used for the extraction and content determination of gallic acid in Wudang pineneedle tea.
