1.A study on biological characteristics of normal adult human nasal septal chondrocytes cultured in vitro

Objective:To study the characteristics of normal adult human nasal chondrocytes cultured in vit-ro. Method :The adult human nasal septal chondrocytes were cultured in vitro. The changes of cellular morpholo-gy from primary to subculture were investigated ;to observe the proliferation of chondrocytes by the growth curveand the survive rate of chondrocytes;and to determine the biologic synthesis of the chondrocytes by toluidineblue,alkaline phosphatase staining and type Ⅱ collagen immunohistochemistry reaction. The survive rate of thenasal septal chondrocytes was observed after being frozen (-196C). Result: The morphology of the primarychondrocytes maintained the spherical shape ,and some became polygonal shape after 4 days. Most of the chondro-cytes transformed into the shape of the fibroblast cell since the fifth generation. The number of the chondrocytes(2th generation)after being cultured in vitro for 5 days was approximately 4 fold of that of the plating. The phe-notype of the chondrocytes was maintained in the first 4 generation,the survive rate of the chondrocytes suscitat-ing from being frozen (-196C ) was 93 %. Conclusion:The normal adult nasal septal chondrocytes cultured inmonolayer in vitro maintained the specific chondrocytes phenotype in the first 4 generation,the growth was nor-mal ,and could survive in the condition of deep-frozen.
2.Effects of Calpain inhibitor Ⅰ on glucocorticoid receptor expression and its transcript activation ability

Objective To investigate the effects of Calpain inhibitor Ⅰ on glucocorticoid receptor and its transcript activation ability. Methods Raw-264.7 cells were treated respectively with Calpain inhibitor Ⅰ and dexamethasone or both for 24 h. The changes of glucocorticoid receptor were observed. COS-7 cells were co-transfected with PRsh-GR? and pMAMneo-CAT vectors, and then the effects of Calpain inhibitor Ⅰ on glucocorticoid receptor and its transcript activation ability were detected. Results The glucocorticoid receptors was decreased after Raw-264.7 cells were treated with dexamethasone for 24 h. Calpain inhibitor Ⅰ could inhibit this effect to some extent. Co-transfection experiment revealed that Calpain inhibitor Ⅰ could also promote glucocorticoid receptor transcript activation ability. Conclusion Calpain inhibitor Ⅰ can inhibit the down-regulation of dexamethasone on glucocorticoid receptor, but promote glucocorticoid receptor transcript activation ability.
3.Characteristics of and surgical strategies for distal tibiofibular syndesmosis separation based on ankle axial CT scan

Objective To explore the clinical characteristics of and surgical strategies for distal tibiofibular syndesmosis separation based on ankle axial CT scan.Methods From January 2009 to January 2016,63 patients with injury to the distal tibiofibular syndesmosis were treated.Their injuries were characterized according to the anatomic characteristics on their ankle axial CT scan images as pronation-extorsion type (28 cases),supination-extorsion type (11 cases) and abduction type (24 cases).Specific strategies of reduction and fixation depended on the specific characteristics of distal tibiofibular syndesmosis separation.After 12 months postoperatively,the reduction quality was assessed by Burwell-Charnley's radiological evaluation system and the function of ankle joint was assessed using American Orthopaedic Foot and Ankle Society (AOFAS) ankle-hindfoot score system.Results The patients were followed up for 6 to 48 months (average,19.3 months).Anatomical reduction was achieved in 19 cases,good reduction in 7 and fair reduction in 2 in the pronation-extorsion type,yielding an excellent to good rate of 92.8%;anatomical reduction was achieved in 6 cases,good reduction in 4 and fair reduction in one in the supination-extorsion type,yielding an excellent to good rate of 90.9%;anatomical reduction was achieved in 17 cases,good reduction in 6 and fair reduction in one in the abduction type,yielding an excellent to good rate of 95.8%.By the AOFAS system,the pronation-extorsion type scored 88.6 points,the supination-extorsion type 89.4 points and the abduction type 86.6 points.Conclusion In the treatment of distal tibiofibular syndesmosis separation,reduction and fixation strategies should depend on analysis of the characteristics on the ankle axial CT scan,so as to achieve positive outcomes.
