1.Comparison analysis of cellular morphology and immune phenotypic in Uygur and Han patients with myelodysplastic syndrome

Objective To investigate the cell morphology and immune phenotypic characteristics in Uygur and Han patients with myelodysplastic syndrome (MDS) in Xinjiang Uygur Autonomous Region.Methods Bone marrow smears of 67 cases diagnosed as MDS were systematicly observed and recorded,flow cytometry (FCM) was used to test the immunophenotyping.Results There were abnormal hematopoiesis in different extent in granulocyte series,erythron series and megakaryocytic series of all patients.Occurence of pathological hematopoietic performance in Uygur patients with MDS was similar to that in Han,granulocyte series [52 cases (77.6 %)] >megakaryocytic series [44 cases (65.7 %)] > erythrocyte series [36 cases (53.7 %)],the differences in two groups were not significant (x2 values 1.02,0.30,0.02,respectively,all P > 0.05).The changes type of pathological hematopoietic performance in two groups were similar,single garden nuclear megakaryocyte [36 cases (53.7 %)],false Pelger nuclear anomaly granulocyte [36 cases (53.7 %)],erythroid gigantic young cell change [33 cases (49.3 %)],granulocyte cell particles reduced or absent [27 cases (40.3 %)] etc.The result of FCM showed that along with the progressive change of RA/RAS to RAEB/RAEB-t,the expression ratios of mature CD15 were decreased,while the expression ratios of early stage CD117 and CD34 were raised gradually (x2 values 6.23,12.06,8.95,7.37,8.95,8.08,respectively,all P < 0.05),the differences in two groups were not significant (x2 values 0.715,0.024,0.146,respectively,all P > 0.05).Meanwhile,Uygur patients with MDS has great expression of CD56 and Han had HLA-DR,there were significantly differences in two groups (x2 values 3.91 and 3.90,respectively,both P < 0.05).Conclusion Occurence of pathological hematopoietic performance in Uygur patients with MDS are similar to that in Han.Most MDSs has two or more levels of pathological hematopoietic performance.The differences of immunophenotype characteristics in two groups are partially significant and test of immunophenotype are helpful in the diagnosis,classification,prognosis of MDS.

[ Abstract] Ischemic brain injury is closely associated w ith oxidative stress, neuron loss, and inflammatory reaction. The thioredoxin system is an important antioxidant system in human body. It plays the important roles in the process of fighting against oxidative stress damage, inhibiting apoptosis and inflammation. In recent years, the roles of thioredoxin system in cerebral ischemia have attracted more and more attention. This article review s the roles of thioredoxin in cerebral ischemia.
3.Prognostic impact of atrial fibrillation in patients with chronic heart failure

Objective To discuss the prognostic impact of atrial fibrillation (AF) in patients with chronic heart failure(CHF).Methods From January,2005 through December,2006 we studied 461 patients with a discharge diagnosis of CHF.Patients were divided by baseline rhythm in sinus rhythm(SR) or AF groups.The main endpoints were all cause death within 3 years and readmission to the hospital for heart failure.Results AF group were more likely to be older and female and to have a history of valvular disease and have a higher left ventricular ejection fraction (LVEF).However,SR group were more likely to have a history of ischemic heart disease.During 3 year follow-up,the main endpoints was higher in AF group than in those with SR group (P <0.01).After adjusting for other covariables,AF and age were related to increased the risk of main endpoints during 3 years follow-up (RR =1.311,95% CI:1.002-1.715,P <0.05 ; RR =1.014,95% CI:1.004-1.024,P < 0.01).Conclusion AF and age were the main risk factors of increased adverse forward prognosis in patients with CHF.
4.Effects of Acupuncture on Oxidative Stress and Neuronal Damage in Prefrontal Cortex of Vascular Dementia Rats

Objective To observe the effects of acupuncture on oxidative stress and neuronal damage in the prefrontal cortex of chronic cerebral hypoperfusion (CCH) rats; To investigate the molecular mechanisms of acupuncture in cognitive impairment of vascular dementia (VD) rats. Methods The animal model of VD was replicated by permanent bilateral common carotid artery occlusion (2VO) to induce CCH in rats. After modeling, rats were randomized into model group, acupuncture group and non-acupoint group. Sham-operation group was set as control group. Rats of acupuncture group and non-acupoint group were treated with needling for 2 weeks at 3 days after 2VO. Nissl and DHE staining were respectively used to evaluate the numbers of neurons and ROS level. SOD activity was examined by xanthine oxidase technique. Results Compared with sham-operation group, the number of neurons and SOD activity decreased in the model group, while the ROS level increased. Compared with the model group, the number of neurons and SOD activity increased in the acupuncture group, while the ROS level decreased. There were no obvious changes in all indexes in non-acupoint group. Conclusion Acupuncture can inhibit oxidative stress and improve neuronal damage and loss in the prefrontal cortex of VD rats.
5.Effects of Acupuncture on Hippocampal Synaptic Transmission Signal Molecules in Rats with Vascular Dementia

Objective To observe the effects of acupuncture on synaptic transmission signal molecules in rats with vascular dementia (VD), such as PKC, CaMKⅡ and NR2B, and discuss the molecular mechanism of acupuncture treatment for VD.Methods The multi-infarct dementia model was established by injection of emboli into the internal carotid artery. Experimental rats were randomly divided into normal group, model group, acupuncture group and non-acupoint group. For acupuncture group, acupuncture needles were penetrated into bilateral Zusanli. Non-acupoint group was given acupuncture treatment at the bilateral hypochondrium (10 mm above iliac crest). The rats in normal group and model group were performed to the same amount of capture stimulation as the acupuncture and non-acupoint groups. After treatment, the hippocampal PKC activity was detected by ELISA. Western blot was used to detect CaMKⅡ expression, and the protein expression of NR2B in CA1, CA3 and DG zones was assayed by immunohistochemical staining.Results Compared with normal group, PKC activity and NR2B expression in the hippocampus significantly decreased in the model group (P<0.01). After the acupuncture treatment, PKC activity increased significantly (P<0.05), and the protein expression of NR2B showed a trend to increase. There was no obvious difference in CaMKⅡ expression among all groups.Conclusion Acupuncture at Zusanli can enhance the activity of hippocampal PKC, a synaptic transmission signal molecule, which maybe one of the important molecular targets for the treatment of VD.

Cellular prion protein (PrP(C)) is ubiquitously expressed in the cytomembrane of a considerable number of eukaryotic cells. Although several studies have investigated the functions of PrP(C) in cell proliferation, cell apoptosis, and tumorigenesis of mammals, the correlated functions of chicken PrP(C) (chPrP(C)) remain unknown. In this study, stable chPrP(C)-downregulated Marek's disease (MD) virus-transformed avian T cells (MSB1-SiRNA-3) were established by introducing short interfering RNA (SiRNA) targeting chicken prion protein genes. We found that downregulation of chPrP(C) inhibits proliferation, invasion, and migration, and induces G1 cell cycle phase arrest and apoptosis of MSB1-SiRNA-3 cells compared with Marek's disease virus-transformed avian T cells (MSB1) and negative control cells. To the best of our knowledge, the present study provides the first evidence supporting the positive correlation between the expression level of chPrP(C) and the proliferation, migration, and invasion ability of MSB1 cells, but appears to protect MSB1 cells from apoptosis, which suggests it functions in the formation and development of MD tumors. This evidence may contribute to future research into the specific molecular mechanisms of chPrP(C) in the formation and development of MD tumors.

7.Znx Ni1 -x S@ DOX nanostructures for combined photothermal-chemotherapy of cancer

Objective :
To explore the application of ZnxNi1-xS@ DOX nanostructures in combined photothermal- chemotherapy treatment of tumors,and to provide new ideas for the study of tumor treatment methods.
Methods:
The morphology and elementary composition of ZnxNi1-xS nanostructures were characterized by scanning emission e- lectron microscopy and other methods.The photothermal properties of ZnxNi1-xS nanostructures were explored by infrared thermal camera.The release of DOX under different temperature and pH was investigated by fluorescence spectrophotometer.The biosafety of ZnxNi1-xS nanostructures and the in vitro antitumor properties of ZnxNi1-xS@ DOX nanostructures were investigated by MTT assay.
Results :
The as-prepared ZnxNi1-xS nanostructure had good dispersion and uniform size.The ZnxNi1-xS nanostructures had good photothermal effect.The drug release behavior showed temperature and pH responsiveness.The MTT assay showed that ZnxNi1-xS nanostructures were nontoxic to normal cells.ZnxNi1-xS@ DOX nanostructures could significantly inhibit the growth of tumor cells under near-infra- red illumination.
Conclusion
The as-prepared ZnxNi1-xS nanostructure has good biocompatibility,on the basis of which the ZnxNi1-xS@ DOX nanostructures achieve good antitumor performance,which can be used for combined photothermal-chemotherapy treatment of tumors.
8.Influential factors and mechanism of non-specific effect in acupuncture.

Even though a large amount of researches showing the clinical effectiveness of acupuncture, there are the researches indicating the absent difference between acupuncture and sham-acupuncture (placebo), which provides the doubts on the specificity of acupuncture. It may be considered that acupuncture intervention is complica-ted and may produce unique non-specific effect, which results in the failure to evaluate effectively the acupuncture efficacy. The influential factors of the non-specific effect of acupuncture include the interaction between physician and patient, expectancy, physician authority, acupuncture treatment experiences, etc. It may be also relevant with the anticipated biological effect, Hawthorne effect, Pavlovian conditioned reflex, etc.
