Objective To study the effect of ligustrazini (LGT) and propofol (PRO) on hepatocellular energy metabolism in the reperfusion injury after hepatic ischemia in rabbits and its mechanism. Methods The rabbits were randomly divided into four groups (n=10 in each), hepatic ischemia-reperfusion (HIR) group, HIR+LGT group (B), HIR+PRO group (C) and HIR+ LGT+PRO group (D). The contents of adenosine triphosphate (ATP), adenosine diphosphate (ADP), adenosine monophosphate (AMP), total adenylic acid number (TAN), energy charge (EC), malondialdehyde (MDA), superoxide dismutase (SOD) and nitric oxide products (NO2-/NO3-) in the liver tissue were measured at 45 minutes after reperfusion. AIM: To observe the relationship between the reversal effect of 4 alkaloids of TCM on expression of P170 being obtained multi-drug resistance to mouse S180 tumor cell induced by chemotherapy protocol of PFC and the adjustment of the cell apoptosis.And to discuss the difference among alkaloids of TCM on reversion of being obtained multi-drug resistance in clinic.To improve the curative effect thereby. METHODS: By the methods of less dosage of Chemotherapy PFC,to set up the mouse models of multi-drug resistance of S180 tumor cell.At the same time to give the mouse matrine,terandrine,oxymatrine and berberine hydrooh loride for 2 weeks and then to observe the P170,TOPOⅡ,Fas and apoposis by flow cytometry. RESULTS: Matrine and terandrine could obviously reduce the expression of P170,the activation of TOPOⅡ induced by Chemotherapy and increase the ratio of the express of Fas and the apopsis of drug resistance to tumor cell.And berberine hydrochloride could obviously reduce the expression of TOPOⅡ. CONCLUSION: The adjustment to the apoposis of tumor cell by matrine and terandrine is related to the reversion of being obtained multi-drug resistance to tumor cell' expression,besides the different degree
3.Effect of L-arginine on function and structure of mitochondria in ischemia-reperfusion myocardial cells in rabbits

AIM: To study the effect of L-arginine (L-Arg) on function and structure of mitochondria in ischemia-reperfusion (MRI) myocardial cells. METHODS: Thirty rabbits were randomly divided into three groups (n=10 in each), control group, MIR group and MIR+L-Arg group. The mitochondrial respiratory function, Ca~(2+) concentration ([Ca~(2+)]m), malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were determined. Meanwhile, the contents of adenosine triphosphate (ATP), adenosine diphosphate (ADP), adenosine monophosphate (AMP), total adenylic acid number (TAN) and energy charge (EC) in the myocardial tissue were respectively measured. Moreover, the ultrastructure changes in myocardial mitochondria were observed during MIR. RESULTS: The mitochondrial respiratory control rate (RCR), velocity 3 (V_3), SOD, surface density (Sv) and specific surface (?) in MIR+L-Arg group were higher than those in MIR group, velocity 4 (V_4), [Ca~(2+)]m, MDA, volume density (Vv), horizental diameter (Hd) were lower than those in MIR group. ATP, ADP, TAN and EC levels of myocardial tissue were higher than those in MIR group. There was no significant difference between MIR+L-Arg and control group in V_3, V_4, SOD, MDA, Vv, Sv, ?, Nv, Vd, AMP and TNA. CONCLUSION: It is suggested that L-Arg improves the function and structure of mitochondria in myocardial cells in the reperfusion injury after myocardial ischemia by decreasing oxygen free radical level and Ca~(2+) overload in the mitochondria. 4.Investigation on Demodex infection status and influencing factors in medical students in Wuhu City

Objective To investigate the Demodex infection status and influencing factors in medical students in Wuhu City. Methods The mite specimens were collected by the cellophane tape method,and the influencing factors were investigated through the questionnaires. Results A total of 316 medical students were surveyed,117 students were infected with Demodex mite,and the total infection rate was 37.03%. There was no significant difference between the male and female in the infection rate( χ2 = 0.00,P > 0.05). Of the 117 infected cases,the infection rate of simple Demodex folliculorum was 51.28%,the infec-tion rate of simple Demodex brevis was 24.79%,and the rate of mixed infections was 23.93%. The rate of the mild infection (83.76%)was significantly higher than that of the moderate infection(12.82 %)and that of the severe infection(3.42%). The infection rate in the forehead was the highest(29.41%). Among the students with the oily skin,the infection rate of Demodex mite was related to sex. The infection rate of simple Demodex brevis in the male(3.70%)was lower than that in the female (16.42%)( χ2 = 6.92,P < 0.05),while the rate of the mixed infections in the male(17.28%)was higher than that in the fe-male(4.48%)( χ2 = 5.91,P < 0.05). The infection rate of facial skin with symptoms(51.80%)was higher than that of normal facial skin(33.05%)( χ2 = 9.14,P < 0.01). There was no significant difference between the infection rate of Demodex mite and the living habits and other factors. Conclusions There is some infection of Demodex in the medical students. The infection rate of facial skin with symptoms is higher than that of the normal facial skin. So we should strengthen the prevention and treat-ment of Demodex infection in medical students.
5.Expression of PTEN in Myocardial Tissue in Coronary Heart Disease

Objective To observe the expression of phosphatase and tensin hom ology deleted on chrom o-som e ten (PTEN) in m yocardial tissue in patients w ith coronary heart disease, and explore the relevance betw een the expression of PTEN and the occurrence and developm ent of coronary heart disease. Methods A total of 16 death cases w ith pathological diagnosis of coronary heart disease w ere collected as experi-m ental group, and 19 cases w ithout m yocardial lesions w ere selected as control group. The expression of PTENprotein and its m RNA w ere detected by im m unohistochem istry and real-tim e fluorescence quanti-tative PC R respectively. The correlation betw een the expression of PTEN and the pathogenesis of coronary heart disease w as analyzed. Results The expression of PTENprotein in myocardium in cases w ith coro-nary heart disease w as significantly low er com pared w ith the control group (P<0.05). There w as no sta-tistical difference of the expression of PTEN m RNA betw een experim ental and control group (P>0.05). Conclusion PTEN m ay be involved in the occurrence and developm ent of coronary heart disease.
6.Inhibition of cell proliferation and arrest of cell cycle progression by blocking Cl- channels in nasopharyngeal carcinoma cells

AIM: The roles of Cl-channels in regulatory volume decrease (RVD), cell proliferation and cell cycle progression in nasopharyngeal carcinoma cells (CNE-2Z) were investigated. METHODS: Image analysis of living cells was used to detect the volume changes following exposure to hypotonic solutions. Cell viability was determined by the trypan blue assay. MTT method was applied to detected cell proliferation. The effect of the blocker on the cell cycle distribution was monitored by the flow cytometry. RESULTS: 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB) inhibited RVD and cell proliferation in a dose-dependent manner. NPPB at the concentration of 100 ?mol/L arrested cells in G 1 phase (G 1 population increased from 54% to 71% at 48 h after treatments), but did not significantly alter cell viability. CONCLUSION: Block of chloride channels suppressed cell proliferation by arresting cells in G 1 phase. The results suggest that activation of Cl-channels and RVD is necessary for facilitating cells to proceed to the S phase from G 1 phase and maintaining cell proliferation.
7.Roles of chloride channels in the migration of nasopharyngeal carcinoma cells at different stages of the cell cycle

S. The migration of CNE-2Z cells was inhibited by chloride channel blockers (ATP, NPPB and tamoxifen), but the inhibitory effect of the blockers varied with cells at different stages. CONCLUSIONS: The migratory ability is associated with the cell cycle in CNE-2Z cells. Chloride channels play an important role in cell migration of CNE-2Z cells.
