1.A Series Studies on Mechanism of TGF-β/Smads in Peritoneal Fibrosis and Regulating Pathway

Peritoneal fibrosis is the most important complication in maintenance of peritoneal dialysis(PD),also the main reason of the changes in peritoneal structure and function,the important factor affecting the long-term survival and prognosis in PD patients.TGF-β is a multifunctional growth factor and also a key factor in organ and tissue fibrosis.We had carried out the following series studies focused on the mechanism and regulatory factors of TGF-β and its signaling proteins-Smads,in peritoneal fibrosis:①the role and mechanism of TGF-β in peritoneal fibrosis;②Over-expressed Smad7 in prevention and treatment of peritoneal fibrosis;③the role and mechanism of TGF-β/Smad in acute peritonitis;④crosstalk between TGF-β and other signaling pathways.The Results showed that,TGF-β1/Smads signaling pathway played a crucial role in the process of peritoneal fibrosis,and over-expression of inhibitory signaling protein Samd7 had potential clinical value in prevention and treatment of PD-related peritoneal fibrosis.
2.Discussion on Action Mechanism of Acupoint Sticking Therapy in Respiratory Diseases Treatment in Summer

Based on the traditional Chinese medicine ( TCM ) theory of correspondence between man and the universe and supporting yang in spring and summer, acupoint sticking therapy has a good clinical effect in respiratory diseases treatment in summer . Its mechanism may be involved in regulating immune function , in-hibiting inflammatory response , scavenging oxygen free radicals and inhibiting protease-antiprotease imbalance , which need to be further studied and identified .
3.Thinking on Key Technical Steps of Establishing on Curative Effect Evaluation Indexes for Traditional Chinese Medicine Syndromes under Disease-syndrome Combination Mode

This paper was aimed to establish the curative effect evaluation indexes scientifically , which is the key technical step for the study of traditional Chinese medicine ( TCM ) effect evaluation methods under the mode of disease-syndrome combination. However, there is still a lack of generally accepted methods. In this paper , several key technical steps in the research process for establishing the curative effect evaluation indexes were put forward , namely , the framework of scientific and reasonable formation on the evaluation indexes is the premise; target selection of evaluation indexes is the core link; the evaluation and verification of evaluation in-dexes are key points , which provide the methodology reference for similar research in this field .
4.Current and challenge in treatment of lupus nephritis

Lupus nephritis (LN) is a major cause of morbidity and mortality in patients with systemic lupus erythematosus.Although the use of aggressive immunosuppression has improved both patient and renal survival over the past several decades,the optimal treatment of LN remains challenging.The therapeutic strategy and progress for patients with lupus nephritis are based on the accumulative evidences of the medicine,and must be personalized according to the patient condition.This paper discusses established and newer treatment options for LN.We also emphasize the challenges in the day-to-day management of lupus nephritis in the clinical practice.
5.Current situation and thinking on development of TCM clinical treatment guidelines

According to the current situation and issues about the development of traditional Chinese medicine (TCM) clinical treatment guidelines,to put forward ideas on the guidelines development:founding up an efficient organization composed of multi-disciplinary staff; following the procedures of the guidelines formulation and ensuring process quality; paying attention to evidence collection,evaluation,integration and recommendation; improving grade of TCM clinical evidence,exploring research methods appropriated to the characteristics of TCM; assessing the guidelines in order to enhance applicability of the guidelines,and so on.According to the features and reality of TCM,to put emphasis on improvement of the grade of the TCM clinical evidence,exploration and establishment of evidence evaluation and classification methods appropriated to the characteristics of TCM; exploration of recommendation methods reflected the characteristics of TCM,other relevant issues also in discussed.
6.Pathogens of Biliary Infection:A Surveillance

OBJECTIVE To find out the changes of pathogens and their sensitivity to antibiotics in bile samples of patients with biliary tract infection of our hospital during last six years. METHODS The data of 359 strains of microbes found in 371 patients with positive bile culture from Jan 2001 to Dec 2006 and their sensitivity to antibiotics were statistically analyzed. RESULTS There were 84 and 188 positive samples respectively in 140 samples during the first half of this study (2001-2003)and 231 ones during the second half (2004-2006) as well as 105 and 254 strains cultured. Respectively, Gram-negative bacteria accounted for 60.0%, and 50.8% and Gram-positive cocci 34.3%, and 40.2% and fungi 5.7%, and 9.0%; Escherichia coli was the major one and accounted for 36.2%, and 31.1%, with the increasing incidence of the positive rates of its ESBLs (34.2% vs 60.8%, P
7.Protective effects of bone marrow mesenchymal stem cells against the renal tubular epithelial cell damage induced by cisplatin

AIM:To investigate the protective effects and mechanism of mesenchymal stem cells on renal tubule cells treated by cisplatin.METHODS:The mesenchymal stem cells(MSCs)were separated from mouse bone marrow.The necrosis and apoptosis rate of renal tubule cells induced by cisplatin were detected by flow cytometry.After co-culture with MSCs,the above changes of renal tubule cells were detected.RESULTS:In co-culture system,the apoptotic rates of renal tubule cell were decreased obviously and the cell number increased obviously.CONCLUSION:The MSCs protect the renal tubule cells from apoptosis induced by cisplatin.This effect may be related with the paracrine effects of MSCs.

9.Effect of parathyroid hormone on differentiation and resorption of osteoclasts in vitro

Objective To study the direct effect of parathyroid hormone(PTH) on osteoclasts (OCs) differentiation and bone resorption in vitro and to explore the relationship between PTH and expression of RANKL (ligand of receptor activator of nuclear factor kappa B)gene and OPG (osteoprotegerin) gene in osteoblasts (OBs) in order to provide some theoretic evidence for understanding the pathogenesis of high-transferred renal osteodyslrophy (ROD). Methods PTH was directly used to induce OCs differentiation from whole bone marrow of C3h mouse by using OCs formation assay and then inspected the bone resorption of these OCs in vitro by using pits assay. Multiple RT-PCR assay was used to examine the expression of RANKL-mRNA and OPG-mRNA in different PTH concentrations and different treating times by PTH. Results (1) PTH in vitro could induce OCs formation form whole bone marrow of wild-type C3h mice.In a certain concentration scope, as the concentration of PTH increased, the number of differentiated OCs and the degree of bone destruction of OCs increased. (2) Expression of RANKL-mRNA and OPG-mRNA was in dose-dependent and time-dependent manner within a certain range of concentration and time. Conclusion PTH in vitro mediates OCs differentiation and bone resorption probably through the up-regulated expression of RANKL-mRNA and OPG-mRNA.
