1.Comparative proteome research of epithelial cells and stroma from invasive ductal carcionma patients

Aim To study the difference of proteins expression of epithelial cells and stroma of breast canc-er patients.Methods Formalin-fixed paraffin-embed-ded tissue samples from invasive ductal carcionma pa-tients were stained by improved HE staining methods. And the neoplastic epithelium and stroma were micro-dissected using laser capture microdissection and ana-lysed by mass spectrometry.Results Total 43 1 differ-ent proteins were detected by mass spectrometry and there were respectively 384 and 298 different kinds of proteins identified in epithelial cells and stroma.A-mong them,25 1 proteins were commonly expressed in two samples,while the contents of 69 and 60 proteins in stroma were respectively higher or lower than the ones in epithelial cells.The expression level and local-ization of proteins identified in epithelial cells and stro-ma were associated with their roles in development and progression of tumor cells.Conclusions The differen-tial expression proteins between epithelial cells and stroma of invasive ductal carcionma patients may func-tion as biomarkers for breast cancer screenning,diag-nosis and prognosis.
2.Inhibitory effect of mastoparan-like peptide from wasp (Vespa magnifica) venom on angiogenesis

Aim To investigate the anti-angiogenic effect of 12a, the mastoparan-like peptide from wasp ( Vespa magnifica ) venom. Methods The tube for-mation and proliferation of human umbilical vein endo-thelial cells ( HUVEC ) and chicken chorioallantoic membrane ( CAM) model were used to observe the an-ti-angiogenic effect of 12a in vitro and in vivo, respec-tively. Results In the CAM model, 0. 1 μg or 1 μg of 12 a could markedly inhibit angiogenesis induced by 0. 2 μg rh-bFGF with heavy loss of color, decreasing density and obscure frame of the vessels. The inhibi-tion rates of angiogenesis were 60 . 2 % for 0 . 1 μg and 90. 3 % for 1 μg, respectively. Accordingly, in HU-VEC culture experiment, the proliferation and angio-genesis of HUVEC treated by 1 mg·L-1 and 10 mg· L-1 of 12 a were decreased by 55. 4 %, 39. 3 % and 51. 6 %, 26. 7 %, respectively. Conclusion 12a has a significant anti-angiogenic effect in a concentra-tion-dependent manner.
3.Gene cloning and bioactive analysis of thymosin-β4 from skin of Nanorana yunnanensis

Aim To investigate the gene sequences and pro-angiogenic activities of thymosin-β4 from skin of Nanorana yunnanensis. Methods Two cDNA se-quences of thymosin-β4 from Nanorana yunnanensis designated as pTβ4-1 and pTβ4-2 were obtained by PCR. The tube formation and proliferation of human umbilical vein endothelial cells ( HUVEC ) and chick-en chorioallantoic membrane ( CAM) model were used to observe the pro-angiogenic effects of synthetic pep-tides according to the reduced amino acid sequences of pTβ4-1 and pTβ4-2 in vitro and in vivo, respectively. Results The cDNA sequences of Tβ4-1 and pTβ4-2 were composed of 662 bp and 673 bp, respectively, and their deduced amino acid sequences contained 44 residues which shared highly similarity with those from human , mouse , chicken , claw frog , zebra fish , turbot and Amolops loloensis. Furthermore, the mature amino acid sequences of pTβ4-1 and pTβ4-2 had highly con-served structural regions when compared with other thy-mosin-β4 previously reported. Synthetic peptides could significantly promote proliferation and tube formation of HUVEC and angiogenesis of CAM. Conclusion There are two thymosin-β4 peptides in skins of Nanorana yunnanensis which have pro-angiogenic ac-tivities.
4.Effect of Speech Therapy after Cleft Palate Repairing: 28 Cases Report

ObjectiveTo observe the effect of speech therapy on patients with dysarthria after cleft palate repairing. Methods28 patients 4 weeks after cleft palate repairing received speech training for 3~9 months. They were assessed with articulation of speech and function of velopharyngeal closure before and after training. ResultsThe nasal leakage rate reduced from (50.64±13.10)% to (23.36±13.14)% after training. 8 patients improved their speech articulation to normal, the efficient was 100%. ConclusionSpeech training may improve the speech articulation for the patients after cleft palate repairing.
5.Qualitative study of undergraduate nursing students’psychological feeling during the internship

Objective To explore undergraduate nursing students’psychological feelings during the internship, and provide a theoretical basis for the development of clinical nursing teaching measures. Methods The phenomenological research methods in qualitative study were adopted in this paper and 15 undergraduate nursing students were done with unstructured interview. In order to extract the theme, the obtained information were analyzed and organized with phenomenological analysis. Results Using the generic analytical method we got four topics about psychological feeling that influenced undergraduate nursing students. The low professional identity, mental depression, limited capacity cannot meet the needs of patients and the teacher was too busy to care of undergraduate nursing students that influence the practice enthusiasm. Conclusion The care managers and teachers should take various measures to improve the quality of nursing students and their theoretical and technical skills, develop a reasonable plan to improve the mental health status of undergraduate nursing students and the quality of clinical nursing practice can be improved.
6.Diagnosis and treatment of laterally spreading tumor (LST) through endoscopy

Objective The laterally spreading tumor (LST) of the large intestine have received a great deal of study in Japan, but no report of such cases was found in China. We here of report 46 cases of LST (47 lesions, because one of the patients was found to have two lesions) diagnosed in our hospital in last two years. The morphological features under endoscopy, pit pattern, pathology and treatment under endoscopy were studied. Methods The 47 lesions of LST were found in a total of 5 735 patients who underwent colonoscopic examination in the last two years (from 01-12-2000 to 30-11-2002). The site, size and morphological features of LST were recorded, and the pit pattern was classified with magnification. 42 patients (totally 42 lesions) received colonoscopic surgery, and the rest refused to accept endoscopic therapy, therefore only a biopsy was done for pathological study. In those patients who accepted colonoscopic surgery, 14 patients were treated by mucosal resection (EMR) after injection of saline solution, 24 were treated by piecemeal mucosal resection (EPMR) after injection of saline solution, in 3 patients the lesion was removed by high frequency loop resection, and Cap-EMR was performed using distal hyaline hat attachment fitted to the end of endoscope. The resected or biopsy specimens were studied pathologically. Results 47 lesions of LST were found in a total of 5863 patients who underwent colonoscopic examination. The distrihution of LST lesions were as follows. 22 lesions were situated in the rectum, 10 lesions in the sigmoid colon, 7 lesions in the descending colon, 4 lesions in the transverse colon, 2 lesions in the ascending colon, and 2 lesions were located in the cecum. Macroscopic type: 25 lesions were homogeneous granular type, 12 lesions were mixed nodular granular type, 8 lesions were flat elevated type, and 2 lesions were pseudo-depressed type. The pit pattern as revealed under magnifying endoscopy indicated that 2 lesions were of type II pit, 8 lesion of type II+III L pit, 9 lesions of type III L pit, and 28 lesions of type IV. The pathological diagnosis revealed that 28 lesions were villous adenomas in which 7 lesions showed early malignant degeneration (6 intramucosal cancer and 1 submucosal cancer). 11 lesions were tubular adenomas, in which 1 was intramucosal cancer, 2 lesions were hyperplastic polyp, and 6 lesions were mixed hyperplastic polyps (serrated adenomas). Conclusion The LST manifest some special features, and their incidence may be higher in China than in Japan. EMR or EPMR may be considered as a good method for treatment for LST.
7.In vitro study on the reguIation of Sheng-mai San on dysbacteriosis of intesti-naI fIora

To study the regulation effect of Sheng-mai San (SMS)on intestinal dysbacteriosis,ceftriaxone sodium was used to induce intestinal dysbacteriosis in vitro,the bacteria were cultured in the selective medium and enu-merated by plat counting method,and the short chain fatty acid were also analyzed .The results showed that SMS (16.5,11.2,5.5 mg/mL)could significantly inhibit the proliferation of potentially harmful bacteria such as Enterococci,Enterobacteriaceae and promote the proliferation of Lactobacillus and Clostridum.Compared with the model group,the proliferation of Enterococci and Enterobacteriaceae was inhibited by 24.9% and 19.1%,while the proliferation of Lactobacillus and Clostridum was increased by 22.3% and 25.8% respectively in the middle-concentration of SMS group (11.2 mg/mL)at 24 h.Meanwhile,SMS in different concentration significantly increase the accumulation of acetic acid and propionic acid generated by gut microbes which could significantly inhibit the proliferation of harmful bacteria.And SMS showed prebiotic effects on intestinal microflora imbalance induced by antibiotics in vitro.
8.The Role of HIV Replicative Fitness in Perinatal Transmission of HIV

Perinatal transmission of Human immunodeficiency virus(HIV),also called mother-to-child transmission(MTCT),accounts for 90% of infections in infants worldwide and occurs in 30%-45% of children born to untreated HIV-1 infected mothers.Among HIV-1 infected mothers,some viruses are transmitted from mothers to their infants while others are not.The relationship between virologic properties and the pathogenesis caused by HIV-1 remains unclear.Previous studies have demonstrated that one obvious source of selective pressure in the perinatal transmission of HIV-1 is maternal neutralizing antibodies.Recent studies have shown that viruses which are successfully transmitted to the child have growth advantages over those not transmitted,when those two viruses are grown together.Furthermore,the higher fitness is determined by the gp120 protein of the virus envelope.This suggests that the selective transmission of viruses with higher fitness occurred exclusively,regardless of transmission routes.There are many factors contributing to the selective transmission and HIV replicative fitness is an important one that should not be neglected.This review summarizes current knowledge of the role of HIV replicative fitness in HIV MTCT transmission and the determinants of viral fitness upon MTCT.
9.Diagnostic significance of dendritic cells for chronic obstructive pulmonary dis-ease

Objective:To detect the levels of dendritic cells(DCs)in peripheral blood of patients with acute exacerbations of chronic obstructive pulmonary disease(AECOPD)and explore the relationship between the content of DCs with AECOPD.Methods:The levels of DCs subsets( mDCs and pDCs) in peripheral blood with thirty-four cases of AECOPD and fifteen cases of healthy persons were measured by flow cytometry with four-color fluorescent analysis;and the pulmonary function tests in every subjects were evaluated by spirometry.Results:Compared with the control group,the levels of mDCs and pDCs inAE COPD groups with GOLDⅠ,GOLDⅡ, GOLDⅢand ClassⅣwere significantly higher( P<0.05) ,and with the severity of lung function in AECOPD patients,the levels were also increased( P<0.05).Conclusion:The levels of mDCs and pDCs inAE COPD patients were higher and significantly correlated with the severity of the disease,suggesting that DCs may be involved in the inflammation reaction process of COPD.
10.Transforming growth factor β1-induced epithellal-mesenchymal transition of rat peritoneal mesothelial cells via RhoA-Rock signaling pathway

Objective To investigate the role of RhoA-Rock signaling pathway in the process of rat peritoneal mesothelial cells (RPMCs) epithelial-mesenchymal transition (EMT) induced by transforming growth factor β1 (TGF-β1). Methods Primary RPMCs were cultured in vitro. After synchronization for 24 hours, RPMCs were randomly assigned to 4 groups: group A (control), group B (TGF-β1, 10 μg/L), group C (10 μg/L TGF-β1+10 μmol/L Y-27632, an inhibitor of Rock, pretreated for 2 hours with Y-27632 before TGF-β1 stimulation), group D (Y-27632 alone, 10 μmol/L). Growth arrested and synchronized RPMCs were stimulated by 10 μg/L TGF-β1 for different time. The mRNA and protein expression levels of E-eadherin, α-SMA and collagen Ⅰ were measured by RT-PCR and Western blotting respectively. The protein expression level of vimentin was measured by Western blotting. Active RhoA was extracted by Plasma Membrane Protein Extraction Kit, then it was assessed by Western blotting. Results (1) TGF-β1 stimulation elicited a robust increase in RhoA activity in time-dependent manner, which was (2.57±0.52) folds compared with control group (P<0.05) after 10 min stimulation. RhoA activity peaked at 1 hour, which was (4.35±0.41) folds compared with control group (P<0.05). (2) TGF-β1 up-regulated mRNA and/or protein expression of α-SMA, vimentin and collagen Ⅰ , and down-regnlated mRNA and protein expression of E-cadherin in RPMCs. (3) The Rock inhibitor Y-27632 effectively revered TGF-β1-induced expression of α-SMA, collagen Ⅰ and vimentin. The mRNA levels of α-SMA and collagen Ⅰ decreased by 53.8% and 55.7%, and the protein levels of α-SMA, vimentin and collagen Ⅰ decreased by 42.6%, 60.1% and 58.1% compared with TGF-β1-stimulated groups (P< 0.05). But Y-27632 had no effect on the level of E-cadherin. Conclusions RhoA-Bock signaling pathway may mediate EMT induced by TGF-β1 in rat peritoneal mesothelial cells. RhoA-Rock pathway may be the potential therapeutic target in the progress of peritoneal fibrosis.
