1.Analysis on the factors influencing the serum potassium concentration changes before and after massive blood transfusion and transfusion hyperkalemia occurred

Objective: To study the changes of blood transfusion and blood potassium concentration and the occurrence of hyperkalemia after transfusion factors influence. Methods: In our hospital from 2010 April to select the diagnosis and treatment of -2012 in April 132 cases of non crush injury patients, these patients were randomly divided into two groups, the control group without blood transfusion therapy group, the observation group for massive transfusion treatment group, changes of serum potassium were compared between the two groups before and after transfusion, and to analyze the related effect factors. Results:The serum potassium levels showed no significant difference between the two groups of patients before and after blood transfusion, operation process, but the postoperative within 12h, observation of blood potassium level group were significantly higher than that in control group, the difference is significant, with statistical significance (t=1.562, P<0.05), in addition to 12h after operation, blood potassium levels between the two groups no significant difference. Conclusion: The clinical blood transfusion is not the main cause of hyperkalemia in patients with, but there are still some patients in massive transfusion potassium increased, therefore, in the process of transfusion, or to close monitoring and observation of the serum potassium level, but also to pay attention to changes in renal function, has ruled out the elevation of serum potassium results in impaired renal function because, in order to reduce the incidence of complications.
2.Expression of CA199,CA724 and CEA in gastric cancer and clinical significance.

Objective To explore the express levels of CA199,CA724 and CEA in different gastric mucosa pathological situation.Methods The experimental group included 82 cases including 15 of chronic atrophic gastritis,15 of intesti- nal metapiasia,15 of dysplasia of the gastric mucosa,19 of gastric ulcer,18 of gastric cancer;15 of the chronic Super- ficial gastritis (CSG) were set as control group.To test express levels of CA199,CA724 and CEA in these cases by the immunohistochemistry technology.Results The positive rate of CA199,CA724 and CEA in gastric cancer was sig- nificantly higher than that in precancerous lesion groups and precancerous state groups (P0.05),but there was a graduate increasing of the positive rate of CA199,CA724 expression in chronic atrophic gastritis,intestinal metaplasia and dysplasia of the gastric mucosa. Conclusion That different expression of CA199,CA724 and CEA in gastric cancer development situation may be help- ful for the early diagnose of gastric cancer.
3.The effects of hypertonic-hyperoncotic solution on the lipopolysaccharide-stimulated ICAM-1 mRNA expression in human vascular endothelial cell

Objective To investigate the alteration in intercellular adhesion molecule (ICAM)-l mRNA expression on the surface of human vascular endothelial cells (HUVECs) induced by LPS stimulation and the effects of hypertonic-hyperoncotic solution (HHS) . Methods HUVECs were enzymatically isolated and cultured in RPMI 1640 culture medium at 37℃ for about 7 days when a monolayer of endothelial cells has grown. The primarily cultured HUVECs were mixed with LPS 100ng/ml and incubated at 37℃ for 30min, 1,4,8, 12and 24 h. 10%NaCl and 10% hydroxyethyl starch (Fresenius) were added to RPMI 1640culture medium. The HHS concentrations were 0.25% and 0.5% respectively. HUVECs were first incubated with either HHS (0.25% , 0.5% ) or isotonic solution for 10 min, 1h and 4h and then stimulated by LPS for 4h. The ICAM-1 mRNA was measured by reverse transcriptase-PCR. Results There was slight expression of ICAM-1 mRNA on the surface of HUVECs in normal condition. 4.The levels of TGF-β1 in hypertensive renal function impairment patients and its clinical value analysis

Objective:To detect the levels of TGF-β1 in hypertensive renal function impairment patients and its clinical value. Methods: Primary hypertension patients with no target organs damage were enrolled in control group, primary hypertension patients with renal function stageⅠand stageⅡwere enrolled in renal function stageⅠgroup and stageⅡgroup. Serum TGF-β1 levels and renal function indices of three groups were compared. Results:There were no significant difference of serum creatinine and blood urea nitrogen of three groups(F=23.523, P<0.05);endogenous creatinine clearance rate of renal functionⅠandⅡgroups were lower than that of control group, TGF-β1 levels of renal functionⅠandⅡ groups were higher than of control group; TGF-β1 levels were negatively correlated with creatinine clearance rate. Conclusion:TGF-β1 level is an ideal index to reflect renal function for its level significantly changes in renal damage stageⅠ,Ⅱand has good correlation with Cin.
5.Effects of integrative medicine treatment of ear herpes zoster

Objective To observe the clinical effect of western medicine combined with traditional reatme medicine in the treatment of ear herpes zoster.Methods 60 herpes zoster oticus patients were selected,randomly divided into observation group and control group,30 cases in each group.The control group received conventional western medicine treatment,while the observation group treated with integrated traditional Chinese and western medicine treatment.Two groups of clinical curative effects,and the average healing time of clinical symptoms were observed.Results The total effective rate in observation group was 93.3 %,which was 73.3 % in the control group,the difference was statistically significant(x2 =4.32,P <0.05),the clinical symptoms of two groups of cure time average of the differences were statistically significant (t =5.24,7.72,7.70,6.21,all P < 0.05).Conclusion The western medicine combined with traditional reatme medicine in the treatment of ear herpes zoster has better effect than that of western medicine.

Objective To observe the effect of PIF1 knockdown on cell growth and cell cycle arrest induced by ionizing radiation.Methods HeLa cell lines that consistently down-regulated PIF1 were prepared by the lentivirus granules interfering technology and confirmed by real-time PCR and Western blotting.The effect of down-regulation of PIF1 on cell growth and cell cycle arrest induced by ionizing radiation was evaluated by cell counting and flow cytometry.Results HeLa cell lines consistently down-regulating PIF1 were established.The growth of HeLa that down-regulated PIF1 was inhibited greatly after 4 Gy of γ-ray irradiation.There was little cell proliferation until the 5th day post 4 Gy γ-ray.Moreover, the S phase block and G2/M phase block of PIF1 knock-downed cell lines were significantly delayed after 8 Gy γ-ray irradiation.Conclusion Knockdown of PIF1 can significantly enhance the radiation sensitivity and delayes the S phase block and G 2 /M phase block induced by ionizing radiation.
7.Chloroplast genomic characterization and phylogenetic analysis of Castanopsis hystrix.

The structure and size of the chloroplast genome of Castanopsis hystrix was determined by Illumina HiSeq 2500 sequencing platform to understand the difference between C. hystrix and the chloroplast genome of the same genus, and the evolutionary position of C. hystrix in the genus, so as to facilitate species identification, genetic diversity analysis and resource conservation of the genus. Bioinformatics analysis was used to perform sequence assembly, annotation and characteristic analysis. R, Python, MISA, CodonW and MEGA 6 bioinformatics software were used to analyze the genome structure and number, codon bias, sequence repeats, simple sequence repeat (SSR) loci and phylogeny. The genome size of C. hystrix chloroplast was 153 754 bp, showing tetrad structure. A total of 130 genes were identified, including 85 coding genes, 37 tRNA genes and 8 rRNA genes. According to codon bias analysis, the average number of effective codons was 55.5, indicating that the codons were highly random and low in bias. Forty-five repeats and 111 SSR loci were detected by SSR and long repeat fragment analysis. Compared with the related species, chloroplast genome sequences were highly conserved, especially the protein coding sequences. Phylogenetic analysis showed that C. hystrix is closely related to the Hainanese cone. In summary, we obtained the basic information and phylogenetic position of the chloroplast genome of red cone, which will provide a preliminary basis for species identification, genetic diversity of natural populations and functional genomics research of C. hystrix.
