1.Adverse Drug Reactions Induced by Rabies Vaccine:Literature Analysis of 156 Cases

OBJECTIVE:To discuss the general pattern and characteristics of adverse drug reactions (ADR) induced by rabies vaccine. METHODS: A total of 156 ADR cases induced by rabies vaccine were retrieved from China Hospital Digital Library and China Pharmaceutical Abstract between Jan. 1995 and Nov. 2008 and analyzed statistically. RESULTS: The ADR induced by rabies vaccine were more often seen in females aged from 1 year to 40 years. 43.59% occurred within 30 minutes after medication;94.87% manifested as allergies,and allergic shock was even noted in serious cases. CONCLUSION: Great importance should be attached to the ADR induced by rabies vaccine to ensure medication safety.
2.Study on the immunogenicity of Schistosoma japonicum mitochondria related protein

Objective To identify the immunogenicity and the potential of using Schistosoma japonicum mitochondria related protein(rSj338) as a candidate vaccine antigen for Schistosomiasis japonica . Methods The expressed recombinant fusion protein(rSj338/26GST) was purified and recognized by S.japonicum heavily infected rabbit sera and rSj338/26GST immunized rabbit sera by Western blotting. The inclusion bodies,the revivified protein and the purified protein from S 12 gel filtration were injected twice into rabbits to produce anti rSj338/26GST antibody which titer was then measured by dot ELISA. Balb/c mice were immunized with the inclusion bodies and the purified protein from S 12 gel filtration and challenged with S. japonicum cercariae to identify the protective immunity produced by rSj338/26GST. Results The inclusion bodies, the revivified protein and the purified protein from S 12 gel filtration could stimulate the rabbits to produce high level of anti rSj338/26GST antibodies and could be recognized by S. japonicum heavily infected rabbit sera and rSj338/26GST immunized rabbit sera. AIM To investigate the effect of oral roxithromycin on serum concentrations at steady state and its pharmacokinetics of aminophylline in New Zealand rabbits. METHODS The experiment was divided into 2 stages: (Ⅰ) The subjects only received a four-day course of oral aminophylline until steady-state; (Ⅱ) aminophylline and roxithromycin were coadministrated from the d 5~10. Ebola virus-related papers covered in PubMed and CNKI were analyzed by co-occurrence and cluster analysis, respectively. The current situation in research of Ebola virus in China and foreign countries was com-pared. Some suggestions were proposed for research of Ebola virus in China, such as attaching importance to re-search of Ebola virus, enforcing research of its vaccine,and strengthening cooperation between domestic and foreign scholars.

Rheumatoid arthritis (RA) is a systemic autoimmune disease with unknown etiology.The main feature of RA is chronic,symmetrical and invasive arthritis.RA may present with severe joint deformity and functional loss,which has higher disability and fatality rates.At present,no effective measures can reverse bone damage associated with RA clinically.Therefore,early diagnosis of RA is critical to improve prognosis.In recent years,imaging increasingly plays an important role in the early diagnosis of RA.This article reviews recent advances in imaging methods for early diagnosis of RA.
6.Clinical Characteristics of Ischemic Colitis:An Analysis of 75 Patients

Background:The incidence of ischemic colitis(IC)is increasing in recent years with the aging of population,and the rate of delayed diagnosis or misdiagnosis is high because of the lacking of specific clinical characteristics. Aims:To study the clinical characteristics of IC so as to improve its early diagnosis and treatment. Methods:The clinical and endoscopic characteristics of 75 IC patients from Jan. 2009 to Dec. 2014 at Urumqi General Hospital were retrospectively analyzed. Results:IC usually occurred in elderly patients with age ≥60 years,females were more common. Most patients had underlying diseases,including hypertension,coronary heart disease,diabetes,hyperlipidemia and arrhythmia,etc. Colonoscopy and medications could also induce IC. The main clinical presentations were abdominal pain,hematochezia and diarrhea. The lesions were mostly located in left colon and of the transient type. Conclusions:IC should be suspected when elderly patients with underlying diseases,complaining abdominal pain,hematochezia and diarrhea,colonoscopy and pathologic study should be undertaken as early as possible. Prognosis is good when treated timely.
7.Expression and clinical significance of MICA in laryngeal carcinoma tissue and cells.

OBJECTIVE:
To investigate the mRNA and protein expression of MICA in laryngeal squamous cell carcinoma tissue and the Hep-2 cells.
METHOD:
Reverse transcriptase-polymerase chain reaction (RT-PCR) and Western-blot were used to detect the expression of MICA mRNA and protein levels in the Hep-2 cells and laryngeal cancer tissues.
RESULT:
The MICA mRNA showed higher expression in Hep-2 cells by RT-PCR. Compared with the control, the mRNA expression of MICA was significantly enhanced in laryngeal cancer tissues (t = 11.878, P < 0.01). The intensity of MICA expression is not related to the clinical stage of cancer. MICA protein demonstrated higher level expression by Western blot. The intensity of MICA protein expression was decreased with increased clinical stage in laryngeal cancer tissues.
CONCLUSION
The MICA mRNA showed stronger expression in Hep-2 cells and laryngeal cancer tissues. The intensity of its expression is not related to clinical stage of cancer. The MICA protein expression was strong in Hep-2 cells. The intensity of MICA protein expression was decreased with increased clinical stage in laryngeal cancer tissues. MICA may play an important role in laryngeal carcinoma process.

Objective: To observe the targeting therapy of the hdsFv-RC-RNase recombinant single chain immunotoxin on the xenograft of the human hepatocellular carcinoma in nude mice and to explore its clinical potentiality. Methods: The prokaryotic expression vector TIG-hdsFv-RC-RNase was transformed into E.coli BL21(DE3)plys to largely express recombinant single chain immunotoxin hdsFv-RC-RNase against hepatocellular carcinoma induced by IPTG. The expressed product was purified via Ni-NTA affinity chromatography under native conditions and mildly refolded. The ELISA was used to analyze its immunological activity of antigen-binding capability. The xenogrft model of the human hepatocellular carcinoma in nude mice was established and the targeting therapy of hdsFv-RC-RNase was evaluated. Results:After induced by IPTG, a new protein band with M_r 41 000 was found in the supernatant of the bacteria and expressed in a soluble form. The expressed product was purified to homogeneity via Ni-NTA affinity chromatography under native conditions. 9.Effects of interleukin-17 on human laryngeal carcinoma Hep-2 cells

Objective To investigate the effects of interleukin-17 (IL-17) on the cell proliferation, apoptosis and migration of human laryngeal carcinoma Hep-2 cells.Methods IL-17 was transiently transfected into Hep-2 cells, and at the same time empty vector group (pEGFP-N1) and normal control group were set up.The efficiency of transfection was evaluated by fluorescence microscope, and the mRNA and protein expressions of IL-17 were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting.The proliferation of cells was detected by methyl thiazolyl tetrazolium (MTT) method, and the apoptosis was detected by flow cytometry.The migration ability was detected by wound-healing assay and Transwell assay.ResultsHep-2 cells transfected with empty vector pEGFP-N1 and IL-17 showed green fluorescence under the fluorescence microscope.Hep-2 cells expressed IL-17 at both mRNA and protein levels after transfection with IL-17.Compared with the normal control group, the proliferation of IL-17 transfected Hep-2 cells was significantly inhibited after 48 h transfection (0.34±0.03 vs.0.46±0.04, P=0.006).The apoptotic rate of IL-17 transfected cells was higher than that of normal control group (26.80%±0.80% vs.2.90%±0.31%, P=0.000).According to the wound-healing assay, compared with the normal control group, the scratch width of IL-17 transfected cells was significantly greater (1.59±0.01 vs.1.36±0.01, P=0.000).Transwell migration experiment showed that the migration of IL-17 transfected cells was significantly lower than that of the normal control group (26.33±2.08 vs.49.33±1.53, P=0.000).Conclusion IL-17 can inhibit the proliferation of human laryngeal carcinoma Hep-2 cells, reduce their migration ability and enhance their apoptosis ability.Therefore, IL-17 may inhibit the occurrence and development of laryngeal carcinoma through a variety of mechanisms.
10.Expression and clinical significance of FoxA 2 in human non-small cell lung cancer

Objective To investigate the expression and clinical significance of FoxA 2(hepatocyte nuclear factor 3β)in human non-small cell lung cancer(NSCLC)tissues and adjacent normal lung tissue.Methods The expressions of FoxA 2 protein in 80 cases with NSCLC tissues and adjacent normal lung tissue were tested by immunohistochemistry,the expressions of mRNA was detected by RT-PCR.Results The expressions of FoxA 2 both on protein and mRNA in NSCLC lung tissue were significantly less than those of adjacent normal lung tissue(protein expression comparison t value was 187.51,186.43,178.24 respectively;mRNA expression comparison t value was 236.70,260.13,126.27,respectively,P < 0.05).However there was no difference on histopathological variability.F value equal to 1.86 and 1.69,P > 0.05).In low differentiated group,the group of Ⅲ stage,with lymph node metastasis,the expression of FoxA 2 protein and mRNA was less than that in the group of high differentiated,the group of Ⅰ-Ⅱ stage,the group of non lymph node metastasis(pretein expression t values were 4.31,3.92,341,mRNA expression t values were 5.44,3.28,5.07,P<0.05).Conclusion FoxA 2 might inhibit progression of human NSCLC.It may participate in the process of tumor tissue differentiation and lymph node metastasis in NSCLC.
