Objective To explore the diagnostic value of plasma fetal DNA level in preeclampsia.Methods Thirty cases of pregnant women with preeclampsia(at 33 weeks and 3 days) and 30 cases of normal pregnant women(at 34 weeks and 3 days) were selected.All the pregnant women carried a male fetus by B-ultrasound,and were sampled at gestational 20 weeks,third trimester and at 1 hour,3 hours,6 hours after delivery.SRY levels in maternal blood were quantitated by polymerase chain reaction(QF-PCR).The endotheliotoxin(ET) level was measured with RIA.Results (1) Mean fetal DNA level of patients with preeclampsia at 20 weeks of gestation was(316?61)copy/ml.They were(266?79)copy/ml,(396?91)copy/ml,(165?43) copy/ml for light and severe preeclampsia women and normal pregnant women,respectively.Maternal blood fetal DNA levels in pregnant women with preeclampsia at 20-weeks of gestation were significantly higher than those normal pregnant women(P

3.P53 regulation of leukemia cells with the blockage of MDM2 by antisense oligonucleotides.

The changes of expression and function of MDM2 and P53 by MDM2 specific antisense oligonucleotides were investigated in HL60 cells. Cells were divided into control group, AS group (MDM2 specific antisense oligonucleotides group), cisplatin group, and combined treatment group. FCM analysis and Western blot and RT-PCR were used to estimate apoptosis and the expression of MDM2 and P53. Our results showed that the transfection of MDM2 specific antisense oligonucleotides obviously inhibited MDM2 expression (P < 0.01) and increased the expression of P53 (P < 0.05). Apoptosis rate were reduced by MDM2 specific antisense oligonucletides and cisplatin (P < 0.01). It is concluded that MDM2 specific antisense oligonucletides can inhibit the expression of MDM2, induce the expression of P53 and increase the apoptosis of leukemia cells after chemotherapy.
4.Dynamic monitoring of fat emboli on transesophageal echocardiogram during total knee arthroplasty

Objective To detect fat emboli in cardiac chamber of the patients undergoing total knee arthroplasty(TKA) by transesophageal echocardiography(TEE), and to discuss the relevant clinical value.Methods Eleven female patients with 12 osteoarthritis(OA) knees were underwent TKA. According to whether using tourniquet or not,12 knees were divided into 2 groups at random:tourniquet-related group (6 knees) and control group(6 knees). Echo intensity and ultrasonic characteristics of fat emboli in cardiac chamber were studied dynamically by multiplane TEE in various periods of the whole operation. Results All of the patients were implanted knee prostheses successfully. In different periods of TKA, fat emboli appeared as isoechoic or hyperechoic particles, which were found in right atrium, but nothing could be found in left heart. The imaging of the particles was flowing with duration time of 10~600s, like dots, lines, "moving star" or "shower". While femur expanding(0~3mins),the particles in right atrium and ventricle became more and more, and the echo intensity became significantly higher than those in other periods. After releasing tourniquet(0~5mins),most particles with highest echo intensity were observed in right heart,showing like "snowstorm" in tourniquet-related group, and more emboli were still detected at the end of monitoring (10 min after releasing tourniquet) by TEE,compared to the control group. No patient suffered from fat embolism syndrome(FES) with clinical manifestation. Conclusions Fat emboli in cardiac chamber can be observed sensitively by real-time TEE dynamically during TKA. It should be encouraged and promoted to analyze ultrasonic characteristics to facilitate the early detection, early diagnosis and early clinical intervention for the subclinical type of FES.
5.Effect of non-invasive labor analgesia on postnatal depression among primiparas

Objective To explore the effect of non-invasive labor analgesia on postnatal depression among primipara. Methods Three hundred primipara of spontaneous labor in our hospital from February 2014 to April 2014 were assigned as control group and another 300 primiparas of spontaneous labor from May 2014 to August 2014 as experiment group. In the control group, one-to-one responsibility system was conducted for the delivery with an accompanying midwife and in the experiment group, non-invasive labor analgesia was done apart from one-to-one responsibility system. The two groups were compared in terms of analgesic effect at three stages of labor, postnatal depression scores and incidence of postnatal depression. Result The analgesic effect of the experiment group was better than that of the control group during the three stages of labor (all P<0.01), and postnatal depression scores and the incidence of postnatal depression in the experiment group were significantly lower than those in the control group (both P<0.05). Conclusion Non-invasive labor analgesia can relieve the labor pains and decrease the incidence of postnatal depression among primiparas.
6.The study of mouse lymphoma cell EL4 transfected with mB7-2 gene

Objective:To assess the potential of mouse immune-costimulating signal B7-2 in inducing immune effect.Methods:(1)The specific mB7-2cDNA fragment from LPS-stimulated mouse splenocytes was obtained by reverse transcription and polymerase chain reaction.The obtained fragment was inserted to pLXSN plasmid.Then the plasmid pLXSN-mB7-2 was packaged into PA317 cells;(2)The EL4/mB7-2 was obtained by infecting the EL4 by the concentrated virus particles produced by PA317/mB7-2;(3)In vitro,the secretion of IL-2 of EL4/mB7-2 stimulated-lymphocytes was detected by using mixed lymphocytes culture.Results:pLXSN-mB7-2 and transgenetic EL4/mB7-2 cells were obained successfully.IL-2 production in 24h in the supernatant stimulated by EL4/mB7-2 was much higher than wild EL4 cells.Conclusion:EL4/mB7-2 can activated T cell to produce IL-2.This research lay foundation for the research of the function of immune-costimulating signal in the tumor immunity and treatment,the mechanism of autoimmune disease and organ transplantation.

8.Effects of transient receptor potential V6 silence on proliferation and apoptosis of trophoblasts

Objective To investigate the effects of the transient receptor potential V6 (TRPV6) gene silencing on the proliferation and apoptosis of trophoblasts HTR-8/SVneo cells.Methods siRNA sequences targeting the TRPV6 gene were constructed and then transfected into HTR-8/SVneo cells mediated by liposome.The cells were divided three groups,including blank control (add the reagent of transfenction),negative control groups (transfecting nonspecific siRNA) and experimental groups (transfecting TRPV6-siRNA).Those cells in every group were collected at 24,48,72 hours after transfecting.The expression levels of TRPV6 mRNA were detected by reverse transcription (RT) PCR at different times after transfecting.The effects of siRNA on the proliferation and apoptosis of the cells were assayed by methyl thiagolyl tetragolium (MTT) and flow cytometry at different times after transfecting.Results siRNA TRPV6 transfection could inhibit the expression of TRPV6 mRNA in the HTR-8/SVneo cells.The expression was decreased with the extension of time,by 0.72 ± 0.02,0.54 ± 0.02 and 0.29 ± 0.01 after 12,48 and 72 hours of siRNA transfection as compared with the blank control and the negative control groups (P <0.01).The rates of proliferation inhibition were (19.29 ± 1.23) %,(32.12 ± 1.35) % and (46.51 ±1.42) % at 24,48 and 72 hours respectively when compared with the blank control (2.12 ± 0.03)%,(2.42 ± 0.02) %,(3.13 ± 0.04) % and the negative control groups (2.37 ± 0.01) %,(2.61 ± 0.05) %,(2.93 ± 0.03) % (P < 0.01).The apoptosis rates of HTR-8/SVneo cells was 16.21% at 48 hours after transfected with siRNA TRPV6,which were significantly higher than 3.27% in the blank control and 5.34% in the negative control groups (P < 0.05).Conclusion Silenceing of TRPV6 genen could inhibit the proliferation and increase the apoptosis of extravillous trophoblas of human placenta.
9.Role of three vessels and trachea view in ultrasonic diagnosis of fetal heart disease and great artery abnormality

Objective To evaluate the role of three vessels and trachea view (3VT) in diagnosis of fetal heart disease and great artery abnormality.Methods 39 pregnant women undergoing fetal ultrasound scan and diagnosed with great artery abnormality were enrolled in this study.Then the image characteristics and significance of 3VT in these 39 cases were analyzed and evaluated.Results In these 39 cases proved by autopsy,persistent truncus arteriosus(7 cases),transposition of the great arteries(7 cases),coarctation of the aorta(5 cases),tetralogy of Fallot (4 cases),pulmonary stenosis (3 cases),hypoplastic left ventricle syndrome(2 cases),persistent left superior vena cava (2 cases),pulmonary valve stenosis with single ventricle(2 cases),Ebstein anomaly with pulmonary stenosis (1 case),interruption of the aortic arch( 1 case),hypoplastic right ventricle(1 case),dextroaortic arch (1 case),and ductus arteriosus stenosis( 1 case) were detected.All of them had anomalies performance on 3VT.While in the 2 cases of double outlet right ventricle,only one eoncurrenting with pulmonary artery stenosis showed reduced inner diameter of pulmonary artery on 3VT.According to the performance of 3VT in great artery abnormality,it could be divided into 4 categories:abnormal location,abnormal ratio,abnormal amount and abnormal blood direction of great artery.Conclusions 3VT is a useful supplement to the 4-chamber view,and has significant value in diagnosis of fetal heart and great arteries abnormities.
10.Effects of partial liquid ventilation on acute lung injury in piglets

AIM: To observe the effect of partial liquid ventilation with perfluorocarbon on gas exchange, hemodynamics and lung histological change in the piglets with surfactant depletion-induced acute lung injury. METHODS: 12 piglets were treated by lung lavage to a partial pressure of oxygen in arterial blood (PaO 2) below 100 mmHg for one hour and randomly divided into gas ventilation group (control group) and partial liquid ventilation (PLV) group, in PLV group, piglets received PFC (FC3280) intratracheally at doses of 15 mL/kg. The parameters of gas exchange and hemodynamics were measured before lung lavage, after lung lavage when the acute lung injury (ALI) was established, and 1 hour, 2 hours after ALI. Animals underwent euthanasia at the end of the study, lung histologic analysis followed. RESULTS: Surfactant depletion by lung lavage induced a stable acute lung injury.Gas exchange increased markedly in the animals that underwent PLV, less hemodynamic damage was observed in PLV group compared with the animals in GV group. Lung histologic analysis demonstrated a less lung damage, including atelectasis, neutrophil excudation, intra-alveolar hemorrhage and interstitial edema in PLV group compared with control group. CONCLUSION: In piglets with surfactant depletion-induced acute lung injury, partial liquid ventilation with perfluorocarbon can improve the gas exchange with less adverse hemodynamic effect and less lung injury compared with conventional gas ventilation.
