1.Influence of regulation of host cell autophagy on the proliferation of Toxoplasma gondii in host cells

Objective To study the role of autophagy in the replication process of Toxoplasma proliferation. Meth-ods As the experimental groups, these cells were infected by Toxoplasma gondii tachyzoites at given MOI (2 ∶ 1, 4 ∶ 1 ,8 ∶ 1 ,16 ∶ 1 ) . Host cell autophagy was detected through acridine orange staining and MDC fluorescence stai-ning at different time points (1, 2, 4, 8, 24, 48, 96 hs post infection). Detect the condition of HEF cells autoph-agy with acridine orange fluorescence staining and MDC fluorescence staining, and detect the replication kinetics of Toxoplasma gondii infection at different time points using Giemsa staining. Results The results of acridine orange and MDC fluorescence staining showed that autophagy inhibitors and inducers could inhibit and promote the autoph-agy of HEF cells respectively. From the results of Giemsa staining, it was found that the proliferation of Toxoplasma gondii in HEF cells could be promoted with autophagy inducers and be inhibited with autophagy inhibitors. Conclu-sion The regulation on autophagy of host cell could regulate the proliferation and replication of Toxoplasma gondii.
2.Association of bacterial translocation with cachexia and its influence on the outcome of gastric cancer patients

ObjectiveTo investigate the association of bacterial translocation (BT) with cachexia and its impact on the outcome of gastric cancer patients.MethodsSixty cachectic gastric cancer patients,50 age- and sex-matched non-cachectic gastric cancer patients,and 55 healthy controls were enrolled in this study between January 2008 and July 2009.Polymerase chain reaction was performed to detect bacterial DNA in the peripheral blood of cancer patients and healthy controls,Cytokine levels were tested by enzyme-linked immunosorbent assay.Flow cytometry was used to detect immune indicators.All the enrolled patients were followed up for two years,and the two-year survival rate was calculated.ResultsThe BT ratio was significantly higher in cachectic patients than in non-cachectic patients (25.0% vs.8.0%,P =0.019) and healthy controls (25.0% vs.0.0%,P =0.000).BT-positive cachectic patients had significantly higher levels of IL-1α,IL-6,TNF-α,and IFN-γ compared with BT-negative cachectic patients ( P =0.012,0.003,0.036,and 0.017,respectively ) and BT-positive non-cachectic patients ( P =0.011,0.034,0.000,and 0.022,respectively).The two-year survival rate in BT-positive cachectic patients was significantly lower than in BT-negative cachectic patients (P =0.023 ).The levels of CD3 +T,CD4+ T,natural killer cells,and CD4 + T/CD8 + T in gastric cancer patients were significantly lower than in healthy controls ( P =0.023,0.031,0.016,0.041,respectively),whereas CD8 + T level was significantly higher in gastric cancer patients than in healthy controls (P =0.038).ConclusionBT may contribute to the development of cancer cachexia and influence the long-term survival of locally advanced gastric cancer patients.
3.Study of the algorithm for inversion of low field nuclear magnetic resonance relaxation distribution.

It is difficult to reflect the properties of samples from the signal directly collected by the low field nuclear magnetic resonance (NMR) analyzer. People must obtain the relationship between the relaxation time and the original signal amplitude of every relaxation component by inversion algorithm. Consequently, the technology of T2 spectrum inversion is crucial to the application of NMR data. This study optimized the regularization factor selection method and presented the regularization algorithm for inversion of low field NMR relaxation distribution, which is based on the regularization theory of ill-posed inverse problem. The results of numerical simulation experiments by Matlab7.0 showed that this method could effectively analyze and process the NMR relaxation data.

4.Radiosensitivity of AsPC-1 cell to γ-rays enhanced by up-regulation of PUMA induced by targeted Slug gene

Objective To explore the influence of PUMA on radiosensitivity of pancreatic cancer AsPC-1 cells after Slug gene inhibition by transfected short interferencing RNA(siRNA). Methods The AsPC-1 cells were infected with MOI 10,50,100 for 72 h, respectively. The expression of Slug and PUMA was analyzed by Western blotting and immunohistochemistry methods. The transfected and control cells were exposed to 4 Gy γ-rays. The cells inhibition rate was examined by MTT, Hoechst 33342 and IP double staining. DNA ladder and Giemsa staning was used to observe apoptosis. Results The relative value of Slug expression was 0.831 ±0.14,0. 546 ±0.12 and 0.178 ±0.08 after AsPC-1 was infected with Slug-siRNA ( MOI 10,50,100) for 72 h, significantly lower than that of control group ( F = 4. 992,P < 0.05 ).The relative value of PUMA was 0. 325 ±0. 07,0. 593 ±0. 11 and 0. 978 ±0. 12, after AsPC-1 was infected with Slug-siRNA ( MOI 10,50,100) for 72 h, significantly higher than that of control group ( F = 4. 324,P < 0. 05 ). The cell proliferation rate was ( 78.76 ± 9. 36 ) % in transfection combined with radiosensitivity group, significantly higher than that of transfection group [ ( 43.68 ± 6.71 ) % ] and radiosensitivity group alone [( 19.25 ± 3.72)% ] (F = 5.056, P < 0.05). The apoptosis of transfection combined with radiosensitivity group was significantly higher than that of others. Conclusions Slug gene targeting siRNA could inhibit the expression of Slug, and consequently increase the activation of PUMA expression, and so enhance the radiosensitivity to γ-rays.
5.Relationship between nuclear factor-κB as well as p53 up-regulated modulator of apoptosis and lung injury induced by severe acute pancreatitis and therapeutic effect of proline dithiocarbamate

Objective To investigate the expression of nuclear factor-κB (NF-κB) and p53 up-regulated modulator of apoptosis (PUMA) in acute lung injury (ALI) induced by severe acute pancreatitis (SAP), and the therapeutic role of proline dithiocarbamate (PDTC). Method SD rats weighed 200~ 250 g were randomly(random number) divided into sham operation group (A group, n = 18), ALI group (B group, n = 18) and PDTC treatment group (C group, n = 18). The model of SAP was eastablished by injecting 1 mL/kg of sodium tauarocholate into the pancreatic capsule of the rats in B group and C group. The model rats in C group were treated with PDTC one hour after modeling. Six rats of each group were sacrificed 6 h,12 h, and 24 hours after modeling. The histopathological changes in lung and pancreas were observed. The levels of NF-κB p65 and PUMA in lung were detected by using Western blotting, and the expressions of bcl-2, bax and caspase-3 mRNA in the lung were detected by using RT-PCR. The lung tissue was taken for examination under transmission electron microscope. TUNEL was used for detection of apoptotic alveolar epithelial cells. Results Six to 24 hours after modeling, the pathological scores in lung of ALI group were significantly higher than those of control group and PDTC group after sodium taurocholate injection ( P < 0.05). The levels of NF-κB p65 and PUMA, and the expressions of bax and caspase3 mRNA in ALI group at different intervals were higher than those in control group and PDTC group ( P < 0.05),whereas the expression of bcl-2 mRNA in ALI group was lower than that in control group and PDTC group ( P <0.05). The NF-κB p65 was correlated closely and positively with PUMA ( r= 0.987, P < 0.01). Higher activity of caspase-3 acrtive units was seen in ALI group than that in control group and PDTC group ( P < 0.05). The microvilli disappeared in ALI group 24 hours later. The apoptosis index in ALI group was higher than that in control group and PDTC group ( P < 0.05). Conclusions The apoptosis of alveolar epithelial cells of rats in ALI group is caused by PUMA activated by NF-κB. PDTC treatment can inhibit apoptosis of alveolar epithelial cells of rats in ALI group by inhibiting the activation of NF-κB.
6.Design of active shimming coils on mini-type permanent magnetic resonance imaging system.

The homogeneity of magnetic field is a key parameter in nuclear magnetic resonance system (NMRI); it can be refined by active shimming. In this work, the authors analyze the theory of active shimming, carry out data processing and simulation, and design an active shimming system which can work on permanent mini-type MRI system. The shimming coils are placed on both out-sides of the receiving shielding box, so the gap of the coils is constant. The system improves the homogeneity of magnetic field (i. e. the decrease of heterogeneity from 10 ppm to 1.4 ppm) on NMI20 mini-type MRI instrument.

In clinical practice, incubators can contribute to better diagnosis and treatment of diseases. Currently, insulation medical equipment used for the newborn are mainly warm-boxes. With the use popularity of warm-boxes in the country, medical accidents caused by improper use are on the increase. According to neonatal insulation technology and standardized management of practice, this article puts forward technical requirements of infant incubators with constant temperature and humidity, operation regulations and management specifications to better regulate the use of such devices. Meanwhile, it also suggests national related departments to formulate "incubator technology management norms" and "warm-box technology and practice", intending to standardize industry behaviors and ensure medical safety.

9.The value of third-generation dual-source CT low-pressure voltage combined with ADMIRE in children′s nasopharynx examination

Objective Exploration of the application value of the third-generation dual source CT low tube voltage (70 kVp) scanning combined with advanced modeling iterative reconstruction (ADMIRE) technique in children with adenoid examination. Methods CT scans were performed in patients with clinically suspected adenoid hypertrophy. They were divided into two groups according to the time of treatment. Group A (40 cases) : low tube voltage (70 kVp) scan, reference tube current 163 mAs, reconstruction with ADMIER, Intensity 3; Group B (40 cases) : conventional 100 kVp, reference tube current 163 mAs, conventional (filtered back-projection, FBP) reconstruction;rest of the scanning parameters remained unchanged. The subjective scores and objective quality indicators of the images (CT value, image noise, signal noise ratio (SNR) , contrast noise ratio (CNR)) and radiation dose of the two groups were compared. Results The difference of radiation dose between group A and group B was statistically significant (P < 0.05). The radiation dose of group A was lower than that of group B by 77.58%.Compared with group B, the image noise of group A increased by 0.002%; the SNR decreased by 0.01%; CNR increased by 0.03%; there was no significant difference in objective quality evaluation index and subjective score between two groups in the image quality (P> 0.05). Conclusion The third-generation dual-source CT low-tube voltage (70 kVp) combined with ADMIRE reconstruction technique for children with adenoid scan can effectively reduce the radiation dose while ensuring image quality.
10.Dynamic change of flavonoids' content in various extent of cortex moutan carbonisatum.

OBJECTIVEStudy on the of content variety of flavonoids in the course of processing Cortex Moutan,and discuss the preparation mechanism of Cortex Moutan Carbonisatum (CMC).
METHODHPLC method was developed for the determination of flavonoids in various extent of CMC, the sample was extracted by ultrasound 30 min twice along with ethanol 50 mL. Chromatographic conditions were as follows: wavelength 360 nm, gradient eluant of methanol--0.5% per hundred trifluoroacetic acid.
RESULTThe content of the three flavonoids cuts down along with the processing time and the rising temperature.
CONCLUSIONThe impact of various extent of processing on flavonoids content of the CMC is very great. The overall trend is that high temperature and long time lead to the lower of the content of flavonoids. This provides a basis data for the further study on the hemostatic mechanism and quality control of CMC.
