Objective To assess the mutagenicity of polycyclic aromatic hydrocarbons(PAHs) extracted from the atmospheric particulates collected from different aerodynamic sizes, seasonal variation and the sampled sites, and to detect the effect of these factors on the mutagenic activity of PAHs.Methods PAHs were extracted from the atmospheric particulates(PM10 and PM2.5) collected from two different sites, business area and industrial area(summer and winter respectively). The mutagenicity of these PAHs samples was evaluated by the Salmonella mutagenicity assay(Ames assay),using Salmonella typhimurium strains TA98 and TA100 with and without metabolic activation(S9).Each sample was assayed in three doses(125,250 and 500?g per plate). Results All samples induced the number of revertants to be higher measured with TA98 with and without S9 mix.The level of specific mutagenic activity measured with strain TA98 was higher than TA100.Metabolic activation with the S9 fraction increased the mutagenicity of the industrial area samples studied for the strain TA98.All groups showed a good dose-response relationship of mutagenicity(P

Objective:To observe the effect of protoscolex on Th subsets and correlative cytokine in mice spleen cells in vitro.Methods:Co-culture spleen cells from BALB/c mice with protoscolices, then IL-4,IFN-γand TGF-βproduction in cell culture supernatants were analyzed by ELISA.The percentage of Th subsets were detected by Flow Cytometry analysis. Results:Secretion levels of IL-4 and TGF-βwere significantly increased in spleen cells at different time point in co-culture system with protoscolices.Ratios of Th2 and Treg cells were also significantly increased in co-culture system at different time points than the control groups. However, there was no statistical significance for ratio of Th1 cells at different time points. Conclusion:The protoscolex can increase the ratios of Th2 cells and Treg cells from spleen cells.Secretion levels of IL-4 and TGF-βwere also increased in spleen cells co-cultured with protosco-lices.The results suggest that these Th cell subsets play a role in the immune escape of the hydatid disease.

Objective:To investigate the expression levels of PPARα/γin RAW264. 7 cells in the early stages of co-cultivation with Echinococcus granulosus in vitro. Methods:RAW264. 7 cells were co-cultured with E. granulosus and collected at 12,24,36,48, 72 h. The mRNA levels of PPAR-γ,PPAR-α,M1 macrophages-associated cytokines including TNF-α,MCP-1 and IL-1β,and M2 mac-rophages-associated cytokines including Arg-1,TGF-β and Fizz-1 were detected by qRT-PCR. The protein levels of Arg-1 and MR were analyzed by ELISA. Results:The expression levels of PPAR-γ, PPAR-αand the M2 macrophages-associated cytokines including Arg-1,TGF-β,Fizz-1 and MR were significantly increased, especially at 72 h (P<0. 05). M1 macrophages-associated cytokines including TNF-α,MCP-1 and IL-1β were decreased at 72h although increased at first. Conclusion:During the early stages of co-cultivation with Echinococcus granulosus in vitro, the levels of PPAR-γ/α are up-regulated in RAW264. 7 cells, which may drive macrophage polarization and play a role in the immune escape.

Objective To investigate MRI features of cystic oligodendrocytoma. Methods MRI features of 10 patients with oligodendroglial tumors proved with operation and pathology were analyzed retrospectively. Results Among 10 patients, lesionslocated on the supratentorium in 8 patients, while on the cerebellar vermis in 2 patients.Solitary lesion was found in 9 cases, and multiple lesions were found in 1 case.Complete cystic lesions were observed in 4 cases, large cystic lesions with solid nodules were found in 3 cases, and multi-cystic lesions were found in 3 cases.Smooth cyst wall of lesions were found in 7 cases, locally depressed and rough wall were found in 3 cases.Calcifications were found in 3 cases.Mild peritumoral edema was found in 9 cases, moderate to severe edema was found in 1 case. The lesions significantly enhanced in 3 cases, ring enhanced in 3 cases, and locally enhanced in 1 case. Conclusion COD is difficult to diagnose, but has certain MRI characteristics.COD should be considered when supratentorial complete cystic lesion or large cystic lesion with septum and/or solid nodule was found with MRI.

As precious Chinese medicinal materials, Moschus and Bovis Calculus are often used in the treatment of acute and severe patients. However, due to their scarce sources and high prices, wild animal resources are in urgent need of protection, natural Moschus and Bovis Calculus can no longer meet the needs of preparation production. The use of substitutes such as Moschus Artifactus, Bovis Calculus Artifactus and Bovis Calculus Sativus has alleviated the shortage of original medicinal materials to some extent, and has been widely used in the production and use of traditional Chinese medicine (TCM) preparations. According to statistics, the 2020 edition of Chinese Pharmacopoeia contains 75 TCM preparations containing Moschus and 95 preparations containing Bovis Calculus, but in the quality standards of these 134 TCM preparations, the difference between natural medicinal materials and their substitutes is not very obvious, and some quality control projects are relatively simple. Based on this, the author intends to sort out the quality standards of TCM preparations containing Moschus or Bovis Calculus in the 2020 edition of Chinese Pharmacopoeia (volume Ⅰ), including the type, drug form, prescription dosage, maximum daily (time) dosage and the quality control items of Moschus and Bovis Calculus in the preparation, in order to explore the rationality of the quality standard of TCM preparations containing Moschus or Bovis Calculus, and to give some suggestions on standardizing the use types of Moschus and Bovis Calculus, improving the quality control items of Moschus and Bovis Calculus in TCM preparations combining modern research achievements and advanced technology.

Objective To investigate the effect of Echinococcus granulosus cyst fluid(EgCF) on the cytoskeletal rearrangement and phagocytosis and the migration of macrophages induced by lipopolysaccharide(LPS). Methods Peritoneal macrophages of C57BL/6 mice were isolated and cultured in vitro, and divided into control group and LPS group and LPS combined with EgCF group. After 48 hours of treatment, filamentous actin (F-actin) changes were observed with rhodamine-labelled phalloidin staining and fluorescence microscopy; TranswellTM chamber was used to test cell migration ability and flow cytometry to test cell phagocytosis. After 1 hour of treatment, PI3K and AKT, phosphorylated AKT (p-AKT), Rac1, guanosine triphospho-Rac1 (GTP-Rac1), WASP and Arp2 protein expressions were detected with Western blot analysis. Results Compared with the control group, after LPS stimulation, macrophages were deformed significantly; pseudopodia increased; actin cytoskeleton increased and was more distributed in pseudopodia; the ability of migration and phagocytosis were significantly improved, and the expression of PI3K, p-AKT, GTP-Rac1, WASP and Arp2 proteins significantly increased. EgCF treatment caused cell shrinkage and disappearance of pseudopodia protrusions of LPS-activated cells, and led to the reduced phagocytic and migratory of cells; the protein expression of PI3K, p-AKT, GTP-Rac1, WASP and Arp2 decreased significantly compared with the LPS group. Conclusion LPS induces the migration and enhances phagocytosis of macrophages while EgCF inhibits these effects, which is related to actin cytoskeleton rearrangement.
