1.Clinical Observation of Ligustrazine Injection,α-ketoacid Combined with Levamlodipine in the Treatment of Chronic Renal Failure

OBJECTIVE:To observe the clinical efficacy and safety of Ligustrazine injection,α-ketoacid combined with levam-lodipine in the treatment of chronic renal failure. METHODS:92 patients with chronic renal failure were randomly divided into ob-servation group(46 cases)and control group(46 cases). All patients received electrolyte,vitamins and essential amino acid,low salt,low fat,hypoglycemic and hypolipemic,anti-infection,correcting anemia and other conventional symptomatic treatment. Based on it,control group received 8 Compound α-ketoacid tablets,3 times a day + 1 Benzenesulfonic acid levamlodipine tablet, once a day. Observation group additionally received 150 mg Ligustrazine hydrochloride injection,adding into 250 ml 5% Glucose injection,intravenous infusion,once a day. The treatment course for both groups was 3 months. Clinical efficacy,and urea nitro-gen (BUN),serum creatinine (Scr),serum cystatin (CysC),low-density lipoprotein cholesterol (LDL-C),total cholesterol (TC),triglyceride (TG) levels before and after treatment and the incidence of adverse reactions in 2 groups were observed. RE-SULTS:The total effective rate in observation group was significantly higher than control group,the difference was statistically sig-nificant(P<0.05). Before treatment,there were no significant differences in the BUN,Scr,CysC,LDL-C,TC and TG between 2 groups(P>0.05). After treatment,BUN,Scr,LDL-C,TC and TG in 2 groups were significantly lower than before,and obser-vation group was lower than control group,CysC was significantly higher than before,and observation group was higher than con-trol group,the differences were statistically significant (P<0.05). And there was no significant difference in the incidence of ad-verse reactions(P>0.05). CONCLUSIONS:Based on conventional treatment,Ligustrazine injection,α-ketoacid combined with le-vamlodipine show obvious efficacy in the treatment of chronic renal failure,they can improve the renal functions,with good safety.
2.Effects of pancuronium on nicotinic acetylcholine receptors in pheochromocytoma cells

Objective To investigate the effects of pancuronium on nicotinic acetylcholine receptors (nAChRs) in pheochromocytoma cells (PC12 cells) and to determine if pancuronium has direct effects on PC cells.Methods PC12 cells (purchased from Institute of Cytology, Chinese Academy of Science) were cultured in DMEM containing penicillin and glutamine. nAChR in PC12 cells were stimulated with different concentrations of Ach ( 10, 30, 100, 300, 1 000 ?mol?L-1 ). Ach-mediated inward currents were recorded using whole-cell patch clamp technique with holding potential set at - 80 mV. To investigate the effects of pancuronium on nAChR in PC cells, the PC12 cells were perfused with different concentrations of pancuronium (0.01,0.1, 1, 10, 100, 1 000 ?mol ? L-1 ) before Ach 1 ?mol?L-1 was added. Results Inward currents were elicited by stimulation of nAChR with Ach in a concentration-dependent manner. 93.7% of nAChRs could be activated by 1 ?mol ? L1 Ach. Pancuronium reversibly suppressed the currents in a concentration-dependent manner compared to the control currents elicited by 1 ?mol?L-1 Ach. 1 ?mol?L-1 pancuronium could almost completely suppressed the currents elicited by 1 ?mol ? L-1 Ach.Conclusion Pancuronium could inhibit nAChR in PC12 cells and reduce catecholamine release.
3.Investigation into the prevalence and suitable diagnostic criteria of gestational diabetes mellitus in China

Objective To investigate the incidence of glucose metabolism disorder during pregnancy and establish the diagnosis criteria for gestational diabetes mellitus (GDM) among Chinese patients.Method A prospective popolation-bused study of 16 286 pregnant women,who received 50 g glucose challenge test (GCT) for the first time between Apr 1,2006 and Sept 30,2006,was performed throughout 18 cities in China. Results According to national diabetes data group (NDDG)criteria,the overall incidence of GDM and glucose impaired glucose tolerance (GIGT) was 2.763% (450/16 286) and 3.862% (629/16 286),respectively; it changed to 5.078% (827/16 286)and 5.268% (858/16 286) when using American diabetes association (ADA) criteria.If the women who had 2 or more abnormal oral glucose tolerance test (OGTT) values meeting or exceeding ADA criteria was classified as group 1,and the women who had one or more meeting or exceeding NDDG criteria was group 2,the ratio of women who met both the criteria of ADA and NDDG in group 1 was 94.2%.The 95% CI of normal glucose when using ADA criteria were fasting glucose 5.3 mmol/L,1 hour 10.4 mmol/L,2 hour 8.7 mmol/L,3 hour 7.7 mmol/L,which is close to ADA criteria.Conclusions ADA criterion is more suitable for Chinese patients.According to NDDG criteria,it is reasonable to treat the patient with 1 or more abnormal OGTT values,and if choosing ADA criteria,2 or more abnormal OGrIT values is more reasonable.
4.Expression of angiotensin-converting enzyme 2 in rat pancreas

Objective To investigate the expression of angiotensin-converting enzyme 2(ACE2) in rat endocrine and exocrine tissue of pancreas and to compare its expression level with that in other visceral tissues. Methods The expression of ACE2 in rat visceral organs was detected by immunohistochemistry and reverse transcriptase polymerase chain reaction (RT-PCR). Results ACE2 was positively expressed in all the test tissues, including pancreas, heart, liver, kidney, lung, spleen, duodenum, ileum and bladder. The expression of ACE2 mRNA was detected in whole pancreas and ACE2 protein was found in both endocrine tissue and exocrine tissue of rat pancreas, while in the endocrine tissue much less ACE2 protein was expressed. The absorbance value of exocrine tissue of rat pancreas increased significantly as compared to that in pancreatic islet. Conclusion Our data demonstrated a tissue specific pattern of ACE2 expression in rat pancreas and this indicated ACE2 might be present in islet of pancreas of human, which might cause the elevation of blood glucose levels in SARS patients. Since ACE2 works as a negative regulator of the angiotensin system, the expression of ACE2 protein in endocrine tissue of pancreas may facilitate the treatment of diabetes.
5.Effects of rocuronium and vecuronium alone or in combination on adult and fetal type muscle nicotinic acetylcholine receptors

Objective To investigate the effects of rocuronium and vecuronium on the adult-type (?-nAChR) and fetal-type (?-nAChR) muscle nicotinic acetylcholine receptors. Methods HEK293 cells were obtained from Institute of Cytology, Chinese Academy of Sciences.?- and ?-nAChRs were expressed heterologously in HEK293 cells using transfection technique. Whole cell patch clamp technique was used to determine the potencies of the two muscle relaxants alone or in combination in blocking the function of the two types of nAChRs. Results Both rocuronium and vecuronium could competitively inhibit the activation of ?- nAChR and ?-nAChR by Ach. The IC50 of rocuronium and vecuronium for ?-nAChR was 169.2 ? 12.5 and (8.3 ? 2.7) ?mol?L-1 and for ?-nAChR was 8.6 ? 2.7 and (55.0?10.4) ?mol?L-1 respectively. The IC50 of rocuronium in combination with vecuronium was (0.7 ? 0.3) ?mol ? L-1 for ?-nAChR and (36.3 ? 14.2) ?mol ? L-1 for ?-nAChR.Conclusion The two muscle relaxants have different blocking action on the two types of nAChRs. Rocuronium has stronger inhibitory effect on ?-nAChR than on ?-nAChR while vecuronium has stronger inhibitory effect on ?-nAChR than on ?-nAChR. The inhibitory effects of the two muscle relaxants in combination was synergistic on ?-nAChR and additive on ?-nAChR.

Protein phosphatase 2A(PP2A)is the main serine/threonine protein phosphatase of the brain,and it is involved in many physiological and pathological processes in the cell. Particularly in the pathogenesis of Alzheimer′s disease(AD),PP2A is closely related to the main pathological features of AD. Deregulation of PP2A enzymes correlates with increased tau phosphorylation, the formation of amyloid precursor protein,and the missing of neurons. PP2A as the target of drug development may bring new hope for the treatment of AD. This review introduces the structure and activity regulation of PP2A,and the role of PP2A in AD.
7.Actions of vecuronium and atracurium on adult and embryonic-type nicotinic acetylcholine receptor at skeletal muscle cell membrane

Objective To investigate and compare the actions of vecuronium and atracurium on adult and embryonic-type nicotinic acetylcholine receptors (?-and ?-nAChR) at skeletal muscle cell membrane. Methods The adult and embryonic nAChRs were heterologously expressed in HEK 293 cells. Peak currents induced by actions of vecuronium and atracurium at ?-and ?-nAChR were recorded in HEK 293 cells, using whole cell patch clamp technique. Results Vecuronium and atracurium competitively inhibited ?-and ?-nAChR in HEK 293 cells. The inhibitor concentrations for half-maximal response (IC50) for vecuronium and atracurium ate-nAChR were (8.3 ? 2.6) ?mol/L and (24.2 ? 10.5) ?mol/L respectively; The IC50 values for vecuronium and atracurium at e-nAChR were (55.0 44 28.4) ?mol/L and (183.2 ? 39.2) ?mol/L respectively.Conclusion According to IC50 values for both adult and embryonic type nAChR, vecuronium is more potent than atracurium on e-and 7-nAChR. Embryonic nAChR is less sensitive to vecuronium and atracurium than adult-type nAChR.
8.Inhibitory activity of substance in seed and sarcocarp of Phellodendron amurense

Objective Looking into the causes of self-regeneration obstacle of the wild Phellodendron amurense population in order to provide the reference for the sutdy of the germination mechanism. MethodsThe germination and seedling growth were observed by taking cabbage, wheat, and P. amurense as the tested objects and the sarcocarp and seed of P. amurense were extracted by alcohol and ethyl ether. Results The extract in sarcocarp and seed of P. amurense could decrease the germination rate of cabbage, wheat, and P. amurense seed, also inhibit the seedling growth in different levels. The inhibitory effect was getting stronger and stronger following the concentration increased. The ethyl ether extract showed the best inhibitory effect, while the water extract had no significant effect. The embryo, emdosperm, and cotyledon of P. amurense were extracted by distilled water, and among them the cotyledon had the strongest inhibition. Conclusion There are some substances with the better inhibitory effect in the sarcocarp and seed of P. amurense, which can inhibit the growth of itself and other plants significantly. The substance with the inhibitory effect exits mainly in cotyledon.
9.Study on the provincial legislation of medical institution management

After The Medical Institution Management Regulations was enacted by the State Council in 1994, 26 Chinese provinces and municipalities have enacted the local legislation of medical institution management.These local legislations make a full use of the space of legislation which the host law gives, reasonably determined the health administrative departments'management responsibilities at all levels, detailed the responsibility division for examina-tion and approval, and defined the medical institution practice behavior requirement.The local legislation has played a positive role for the rational medical resources allocation and standardized the local medical institutions practice or-der.But as time goes by and reform deeply, the weakness of these legislations has emerged.This is seen for the fact that they have not been repaired for many years, blindly repeat host law, heavily rely on the regulations with poor sta-bility , do not keep the pace with the system design times, violate the license law, and pay attention to the administra-tive examination and approval instead of the occupational behavior management.This paper makes full investigation on the existing relevant medical institution management laws, regulations and policies.Through the analysis of advan-tages and disadvantages of provincial legislations, some thinking on the medical institutions management is put for-ward.
10.Expression and significance of Six1 in cervical cancer

Objective To investigate the clinical significance of protein expression of Six1 in cervical cancer. Meth?ods The immunohistochemical (IHC) staining was applied to detect the expression of Six1 protein in normal cervical tis?sues (n=32), cervical intraepithelial neoplasia (CIN) tissues (n=49) and cervical cancer tissues (n=123). The localization of Six1 protein was detected in vitro cultured HeLa cells using immunofluorescence (IF) staining. Results The positive rate of Six1was significantly higher in cervical cancer (72.3%) than that of CIN tissues (28.6%) and normal cervical tissues (15.6%,χ2=13.118 and 10.058 respectively, P<0.01). There were significance differences in expression levels of Six1 protein be?tween different tumor sizes and metastasis of cervical cancer (P < 0.01). The Six1 protein showed positive signals in cyto?plasm and nucleoli in HeLa cells. Conclusion Six1 expression is associated with cervical cancer, which may be a potential biomarker for invasion and metastasis of cervical cancer.
