1.Detection of P-selection,nitric oxide,and high-sensitive-C-reactive protein in type 2 diabetes mellitus with macroangiopathy and their significance

Objective To explore the relationship between P-selection,nitric oxide(NO),high-sensitive-C-reactive protein(hs-CRP)and type 2 diabetes mellitus(T2DM)with macroangiopathy.Methods In sernm of 125 T2DM subjects(60 subjects with macroangiopathy and 65 subjects without macroangiopathy)and 55 normal controls,the activity of P-selection,NO and hs-CRP was detected by ELISA,Nitrate Reductase Assay and scattering immunoturbidimetric method,respectivley.Results The activity of serum P-selection and hs-CRP was significantly higher in diabetic macroangiopathie group than in non macroangiopathic group and control normal.Serum NO was significantly lower in diabetic rnacroanglopathic group than in non macroangiopathic group and control normal.Serum P-selection was positively correlated with hs-CRP-systolic blood pressure(SBP),LDL-C,and was negatively correla- ted with NO:P-selection,hs-CRP,LDL-C and SBP were independently risk factors for T2DM with macroangiopathy. Conclusion Determination of P-selection and NO is very important for prevention and diagnosis of diabetic mac- roangiopathy in early stage.
2.The Effects of Telmisartan on Oxidative stress and the Activity of Protein Kinase C in Type 2 Diabetic Rat

Objective To observe the effets of telmisartan on oxidative stress and the activity of protein kinase C in type 2 diabetic Rat.Methods Thirty six Wistar rate were divided into three groups as normal rats、diabetic rats and diabetic rats treated with telmisartan.Blood glucose,blood insulin,blood lipid were measured after 4,5,11 and 17 weeks,serum creatinine,blood uria nitrogen,urinary albumin excretory rate, lipid peroxidation(MDA),the activity of protein kinase C(PKC) and of antioxidant enzymes including Cu/Zn superoxide dismutase(Cu, Zn-SOD),catalase(CAT) and glutathione peroxidase(GSH-Px) in renal tissue were measured after 11 and 17 weeks.In addition,the renal tissue were observed by light microscopy.Results The blood glucose and blood insulin were found no significant difference between that in type 2 diabetic rats with and without telmisartan treatment groups.Comparing with untreated group,the level of blood lipid,serum creatinine,blood uria nitrogen,urinary albumin excretory rate, renal MDA and the activity of cell membrane PKC in telmisartan treated group were significantly decreased,but the activity of renal Cu/Zn SOD,CAT and GSH-Px was increased.There was significant correlation between the change of renal MDA and the activity of antioxidase and the activity of cell membrane PKC. Conclusion Telmisartan could inhibit oxidative stress in diabetic rats, and its mechanism perhaps was decreasing the activity of cell membrane PKC.
3.Gravitational erythema:a case report

A 15-year-old female patient presented with orthostatic erythema in the limbs for half a year. Skin examination revealed that diffuse reticular erythema appeared in the limbs after standing with her arms hanging down for 30 seconds, while the erythema disappeared and skin color returned to normal after raising her arms or lying flat for 30 seconds. The rash in the limbs was reproduced by inflating a sphygmomanometer cuff to 60 mmHg for 2 minutes with the limbs in the horizontal position. Antinuclear antibodies were positive at a titer of 1 ∶1 000. Histopathological examination showed dilated small vessels in the superficial dermis without obvious inflammatory cell infiltration. The patient was diagnosed with gravitational erythema.
4.Expression and significance of VEGF and Bax in non-small cell lung cancer

Objective To investigate the expression of vascular endothelial growth factor (VEGF) and Bax in non‐small cell lung cancer (NSCLC) and clinical value .Methods Paraffin specimens were collected from 80 cases of NSCLC tissues(NSCLC group) and 20 cases of normal lung tissue adjacent to benign lesions(control group) .Western blot and immunohistochemistry were used to dectect the expression of VEGF and Bax .The relationship of the expression of VEGF and Bax with histological classifica‐tion ,stage ,lymph mode metastasis were analyzed .Results The expression of VEGF in NSCLC group was higher than that in con‐trol group ,the differences were statistically significant (P<0 .05) .The higher expression of VEGF in the group of stage Ⅲ‐Ⅳ was obtained than that in the group of stage Ⅰ‐Ⅱ (P<0 .05) .The higher expression of VEGF in the group with lymph mode metasta‐sis was obtained than that in the group without lymph mode metastasis (P<0 .05) .The expression of VEGF was not found to be related with histological classification (P>0 .05) .The expression of Bax in NSCLC tissue was lower than that in normal tissue ,the differences was statistically significant (P<0 .05) .The lower expression of Bax in the group of stage Ⅲ‐Ⅳ was obtained than that in the group of stage Ⅰ‐Ⅱ (P<0 .05) .The lower expression of Bax in the group with lymph mode metastasis was obtained than that in the group without lymph mode metastasis (P<0 .05) .The expression of Bax was not found to be related with histological classification (P>0 .05) .The expression of VEGF was negatively correlated with Bax in NSCLC (P<0 .05) .Conclusion The ex‐pression of VEGF played a promoting effect in the NSCLC ,and was negatively correlated with the expression of Bax protein ,and the expression of the two could provide a basis for the diagnosis and prognosis of NSCLC .
6.Modular analysis of target genes of microRNAs in plasma of coal-burning fluoride exposed population based on protein-protein interaction network

Objective:To establish a protein-protein interaction (PPI) network of 5 microRNA (miRNA) target genes differentially expressed in the plasma of coal-burning fluoride exposed population, and to screen genes and gene modules with important roles.Methods:Five miRNA (hsa-miRNA-3131, hsa-miRNA-4516, hsa-miRNA-6501-5p, hsa-miRNA-10b-5p, hsa-miRNA-4683) target genes differentially expressed in the plasma of coal-burning fluoride exposed population screened by our previous study were mapped to the STRING online database (https://string-db.org), and the PPI network was screened. The Cytoscape v3.6.0 software was used to visualize the PPI network, the topological attribute values degree and betweenness centrality were obtained by the NetworkAnalyzer plug-in, and the central node was filtered in the network (the node with the highest degree and the highest betweenness centrality). At the same time, the maximal clique centrality (MCC) analysis method in the CytoHubba plug-in was used to determine the important nodes in the PPI network. The cluster analysis was conducted by the MCODE plug-in, and the gene modules were screened in the PPI network. The protein names contained in the gene modules were submitted online to the KOBAS v3.0 database (http://kobas.cbi.pku.edu.cn/), and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis was performed on the gene modules selected by the MCODE plug-in.Results:The PPI network of target genes was consisted of 1 035 nodes and 4 346 edges. The degree (101) and betweenness centrality (0.010 723 89) of ubiquitin A-52 residue ribosomal protein fusion product 1 (UBA52) were the highest, which was the central node of the PPI network. According to MCC analysis, UBA52 was an important node in the PPI network. The top 5 gene modules were selected from the PPI network, and the highly enriched gene pathways in the KEGG pathway enrichment analysis of the 5 gene modules included ubiquitin mediated proteolysis, spliceosome, endocytosis, neuroactive ligand-receptor interaction and vesicular transport.Conclusion:The PPI network of 5 miRNA target genes differentially expressed in the plasma of people exposed to coal-burning pollution of fluoride is successfully established, and the UBA52 gene and the 5 main pathways of gene modules are selected.
7.Exosomal microRNAs and their roles in tumors

Exosomes contain many components including variety kinds of lipids,proteins,microRNAs (miRNAs) and so on.They can transport their loaded substances to the recipient cells and play a role of biology.Exosomal miRNAs play important roles in the occurrence and development of tumors,and they are closely related to the tumor growth,metastasis,drug resistance and immune regulation.Exosomes and exosomal miRNAs can be served as reference indexes for tumor metastasis and prognosis,which provide new targets for tumor therapy.
8.Clinical analysis of maintenance hormonal treatment for patients with advanced breast cancer after response to chemotherapy

ObjectiveTo observe the efficacy of maintenance hormonal treatment after response to chemotherapy in advanced breast cancer. Methods8 patients with advanced breast cancer were treated with chemotherapy,maintenance hormonal therapy were given after response to chemotherapy.The efficacy was evaluated every 2 cycles of chemotherapy and 2 months of endocrine therapy according to RECIST standard.Results8 patients received chemotherapy for 2-8 cycles (median 4 cycles). All patients got PR, the duration of chemotherapy was 1-6 months (median 2 months), the time to failure of chemotherapy was 4 months. Until the last follow-up day (31th December 2010), the time to progression was 6-86 months (median 13.5 months).Survival was 6-86 months(median 21.5 months).Seven patients quit the chemotherapy due to severe side effects of hematologic toxicity,fatigue or nausea vomiting.One patient died because of allergy to paclitaxol.Conclusion Maintenance hormonal treatment after patients with metastatic breast cancer response to chemotherapy may prolong the duration of effective therapy and improve the QOL.
9.The role of matrix metalioproteinase-9 and high sensitive C-reactive protein in type 2 diabetes mellitus combined with coronary heart disease

Objective To investigate the effect of matrix metalloproteinas-9 (MMP-9) and high sensitive C-reactive protein (hs-CRP) in type 2 diabetes mellitus (T2DM) combined with coronary heart disease(CHD). Methods Thirty-one patients with T2DM combined with CHD(T2DM combined with CHD group) ,50 patients with CHD (CHD group) and 30 healthy volunteers (control group) were studied. Serum MMP-9 was measured by ELISA, and serum hs-CRP was measured by scattering immunoturbidimetric assay. Results The level of MMP-9 in T2DM combined with CHD group (409.62 μg/L) was significantly higher than that in CHD group (263.40 μg/L) and control group [(196.15 ±44.89) μg/L] (P < 0.05).The level of hs-CRP in T2DM combined with CHD group (17.20 mg/L) was significantly higher than that in CHD group (4.57 mg/L) and control group [(1.52±0.78) mg/L] (P <0.01). MMP-9 wa significantly related with hs-CRP in T2DM combined with C HD group (r = 0.482,P < 0.01). Conclusion The serum M MP-9 and hs-CRP levels are closely associated with the occurrence and development of diabetes related coronary atherosclerosis.
10.Oxidative modification of rat β2-glycoprotein Ⅰ

Objective:To investigate oxidative modification of β2-glycoprotein Ⅰ in vit ro.Methods:Rat β2-glycoprotein Ⅰ was purified and characterized,then oxidize d by hypoxanthine plus xanthine oxidase as a supreroxide free radical generating system;carbonl groups of β2-glycoprotein Ⅰ were detected by the reaction w ith 2,4-dinitrophenylhudrazine.Results:There was a significant increase of carbonyl groups formation in β2- glycoprotein Ⅰ oxidized in comparison with native β2-glycoprotein Ⅰ (P <0.05). Conclusion:Carbonyl groups have been formed in vitro on rat β2-glycoprotein Ⅰ after oxidative modification using hypoxanthine plus xanthine oxidase system.
