The kinetic alterations of plasma prekallikrcin(PKA) and antithrombin Ⅲ (AT-Ⅲ) following myocardial infarction induced by 2 injections subcutaneously of 85 mg isoproterenol/kg at 24 hours interval in rats were detected in this study, and the relationship between PKA, AT-Ⅲ and myocardial ischemia induced by isoproterenol were also discussed. It was found that the activity of PKA decreased significantly and AT-Ⅲ tended to decrease at 4 hours after the first injection of isoproterenol, both of the PKA and AT-Ⅲ at 24 hours were lower than those at 4 hours and evidently lower than those of normal control, and at 48 hours after the first injection of isoproterneol the PKA was still maintained at the same level of 24 hours but the AT-Ⅲ almost returned to the normal level.

Objective To investigate the expressions and clinical significances of Runt-domain-related 3 (Runx3) and chromodomain helicase DNA-binding protein 5 (CHD5) in colorectal cancer.Methods Ninety-six colorectal cancer tissue samples and matched adjacent normal tissues and 72 colorectal adenoma tissues were collected.Real-time quantitative polymerase chain reaction (qRT-PCR) and Western blotting were used to detect the mRNA and protein expression of Runx3 and CHD5.The associations of Runx3 and CHD5 expression with clinical pathological characteristics,diagnostic value and prognosis relationship of patients were further analyzed.Results Runx3 and CHD5 relative expressions of mRNA and protein were 0.35 ± 0.00,0.28 ± 0.02 and 0.26 ± 0.02,0.31 ± 0.01,which were significantly lower than those in the matched adjacent tissues 0.95 ± 0.02,0.92 ± 0.02 and 0.89 ± 0.03,0.93 ± 0.02 (t =2.36,P ＜ 0.05;t =1.25,P ＜ 0.05;t =1.37,P ＜ 0.05;t =1.13,P ＜ 0.05) and colorectal adenoma tissues 0.89 ± 0.02,0.90 ± 0.02 and 0.85±0.02,0.87±0.04 (t=2.27,P＜0.05;t=2.16,P＜0.05;t=1.25,P＜0.05;t=2.65,P＜0.05).Runx3 and CHD5 expressions differed significantly between tumors with different TNM stages (x2 =4.65,P =0.031;x2 =7.89,P =0.005),depths of tumor invasion (x2 =4.17,P =0.041;x2 =4.86,P =0.028),lymph node statuses (x2 =4.20,P =0.040;x2 =7.02,P =0.008),or histological differentiation (x2 =7.31 P =0.036;x2 =9.54,P =0.023).Linear correlation analysis showed that the expressions of the two genes were positively correlated (r =0.572,P =0.001).Receiver operating characteristic (ROC) curve showed that Runx3 and CHD5 had diagnostic value (AUC were 0.712,0.745;sensitivity and specificity were 45.9％,52.5％ and 83.6％,81.4％ respectively).Runx3 and CHD5 both low expression group compared with the other patient groups in overall survival time (x2 =8.156,P ＜ O.05) and progression-free survival (x2 =6.325,P ＜ 0.05) had statistically significant differences.Conclusion Runx3 and CHD5 are low expressed in colorectal cancer and may prove useful as biomarkers for diagnosis target and prognostic indication in patients with colorectal cancer.

Objective: This study was undertaken to explore the effect of left ventricular assist device (LVAD) on failing heart after myocardial ischemia. By detecting the changes of plasma atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP) and cardiac troponin I (cTnI) levels within 6 hours after the implantation of LVAD, we review the basis on which neurohormones may be used to determine prognoses of failing heart and choose an optimal predictor. Methods: 15 adult healthy dogs were divided into 3 groups randomly. The LVAD was implanted in LA-AO bypass in all three groups. Myocardial ischemia was induced by ligating the main left anterior descending coronary artery (LAD). In group A, after a ligation of 10 minutes, the myocardium was reperfused for 6 hours. In group B, after a ligation of 40 minutes, the myocardium was reperfused for 6 hours. In group C, after a ligation of 40 minutes, the LVAD was used for LV support for 6 hours. Results: After 6 hours reperfusion,in group C, the hemodynamics was significantly improved, the ANP, BNP and cTnI were return to normal level, and myocardial ultrastructure was recovered significantly. While in group B, the hemodynamic, the neurohormones, and myocardial ultrastructure were worse. Relational analysis demonstrated that ANP and cTnI levels were influenced by hemodynamics obviously, but there was a weak relationship between circulating BNP and hemodynamics. Plasma BNP level was able to identify the cardiac function status. Conclusion: LVAD can be beneficial to improve cardiac function and can reduce the plasma levels of ANP, BNP and cTnI. Plasma BNP level can identify the cardiac function status. Those findings indicated that plasma BNP determination provides important prognostic information about cardiac function and may be a better prognostic indicator.

Objective To establish an effective method for inducing mouse bone marrow mononuclear cells(MNCs) to differentiate into hepatocytes.Methods MNCs were isolated by gradient density centrifugation,and the cells were cultured in Iscove's Modified Dulbecco's medium supplemented with 10 % new bovine serum(NBS),20 ng/ml hepatocyte growth factor(HGF),10 ng/ml fibroblast growth factor-4(FGF-4) and 10 ng/ml oncostatin m(OSM) for 21 days' induction.The medium was changed every 3 days.Results When MNCs were cultured with HGF,FGF-4 and OSM,cuboidal morphology was observed,and cells also expressed marker genes specific for liver cells in a time-dependent manner.?-fetoprotein(AFP) and cytokeratin 19(CK19) were expressed on the day 7,and CK18 and TAT were detected on the day 14-21,in common with the immunofluoresence results.Differentiated cells further demonstrated these cells also acquired functional characteristics of hepatocytes.Conclusion Mouse MNCs can differentiate into hepatocytes when induced by HGF,FGF-4 and OSM.

Objective:To develop a new type of microelectrode used to measure free fluoride in minim solution. Methods:F--ion sensitive field transistor( F--ISFET) microelectrode was developed based on H+-ISFET combined with an Ag/AgCl referential electrode of microtube. The F--ISFET was used to determine the free fluoride ions in saliva and dental plaque fluid in microliter. And its property was investigated. Results:The test limit of F--ISFET was 10 -6 mol/L-10 -1mol/L, the linear test limit was 10 -6mol/L-10 -4mol/L, the sensitivity was 30-55 mV/pF, the responding time was less than 30 seconds, r2 was 0.97, the reclamation was 94%-104%. Conclusion:F--ISFET can be used to measure free fluoride ions in dental plaque.

Objective:To study the biological characteristics of new retrograde filling materials iRoot BP plus and iRoot FS.Methods:(1 )The roots were cut into 3 mm in length,and the root canals were pre-pared to 1 mm in diameter,followed by being filled with iRoot BP plus,iRoot FS,or mineral trioxide ag-gregate (MTA).The specimens were immersed in simulated body fluid (SBF).The ability of minerali-zation in vitro was detected through three studies.First,the mineralization of specimens was analyzed through scanning electron microscope observations and energy dispersive X-ray spectrometer.Then,the pH of SBF was monitored using pH meter.(2)The extracts were gained by immersing blocks of iRoot BP plus,iRoot FS,and MTA (8 mm diameter and 2 mm height)into dulbecco’s modified eagle medium (DMEM).The effects of the extracts on proliferation of MG63 cells were detected through MTT assay. The gene expression level of alkaline phosphatase (ALP)was analyzed by quantitative real-time PCR, and the expression of ALP activity was observed by ALP activity staining.Results:(1 )The formation of minerals could be observed on the surfaces of iRoot BP plus,iRoot FS,and MTA at the end of 24 h,and there were more amounts of apatite aggregated after 14 days.The values of Ca/P ratios of apatites were 1.43,1.39,and 1.51,respectively.(2)The pH of SBF could be raised to 8.09 ±0.07,7.91 ± 0.06,and 8.1 1 ±0.06,respectively,significantly higher than the blank.(3)The extracts of iRoot BP plus,iRoot FS,and MTA of dilutions of 1∶5 and 1∶10 presented no effect of proliferation of MG63 cells. (4)iRoot BP plus and iRoot FS could significantly up-regulated the levels of ALP messenger RNA ex-pression,while there was no obvious difference in ALP staining among the iRoot BP plus,iRoot FS, MTA,and the blank.Conclusion:The present study shows that iRoot has displayed good mineralization capability in vitro and capability to promote differentiation and mineralization of MG63 cells,inferring that iRoot may have good bioactivity.

OBJECTIVE:To observe therapeutic efficacy of dexmedetomidine combined with propofol for sepsis infection deliration in patients underwent continuous intravenous blood replacement,and to study pharmacokinetic study. METHODS:60 patients with sepsis infection deliration underwent continuous intravenous blood replacement were selected and randomly divided into observation group and control group with 30 cases in each group. Control group was given propofol 0.5 mg/kg,and observa-tion group given dexmedetomidine and propofol. Therapeutic efficacy of 2 groups were analyzed comparatively,and pharmacoki-netic parameters of propofol were studied. RESULTS:The pharmacokinetic parameters of propofol as elimination half-life period (t1/2β),slow distribution half-life period (t1/2α) were shorter than literature reference value,with statistical significance (P<0.05),while the distribution rate constants(K21,K13,K10),the central compartment volume of distribution(VC),systemic clear-ance(CL),rapid distribution half-life period(t1/2pi)had no statistical significance,compared with literature reference value(P>0.05). After treatment,ICDSC score of 2 groups were decreased significantly,with statistical significance(P<0.05),while the observation group was significantly higher than the control group,with no statistical significance before and after treatment(P>0.05);the average hospital stay of observation group was significantly shorter than that of control group,with statistical signifi-cance(χ2=6.263 4,P=0.000);the incidence of ADR in control group was significantly higher than the observation group,with statistical significance(P<0.05). CONCLUSIONS:Dexmedetomidine combined with propofol have good effect on sepsis infec-tion deliration in patients underwent continuous intravenous blood replacement,and can reduce the length of hospital stay and the incidence of ADR.
