1.The roles of Musashil and Notch signaling pathways in neurogenesis after cerebral ischemia

Musashil is an evolutionarily conserved RNA binding protein. It plays an important role in the aspects of maintaining stem cell state, differentiation and tumorigenesis. Notch signal pathway plays an important role in the proliferation of neural stem cells after cerebral ischemia. This article briefly introduces the structure and molecular characteristics of Musashil and Notch, and reviews the interaction in the neurogenesis of Musashil and Notch signaling pathway after cerebral ischemic reperfusion.

Objective To explore the clinical value of false positive of Down’ s Syndrome Screening for premature rupture of membranes in the second trimester of pregnancy. Methods From Jan. 2009 to Jul. 2013, there were 321 cases who were in the second trimester of high risk pregnancy recieved Down’ s Syndrome Screening. Their fetals have been excluded chromosome or organ abnormality, and there was no fetal organ structure abnormal through sequence prenatal ultrasound examinations. Results of their pregnancy have been followed-up, and pregnancy outcomes of 346 cases who were of low risk were followed-up at the same time. Results Among the 321 cases, there were 14 ca-ses of abortion because of PROM ( gestational age less than 28 weeks);17 cases of premature delivery because of PROM, including 9 cases whose gestational age were from 28 to 34 weeks and 8 cases whose gestational age were from 34 weeks to 37 weeks;and 7 cases of neonatal asphyxia. In the low risk group, abortion because of PROM occured in 6 cases before 28 weeks, 3 cases between 28 to 34 weeks, and 4 ca-ses between 34 to 37 weeks, and neonatal asphyxia occured in 3 cases. Conclusion Comparing with Down’ s screening false positive preg-nant women and the low risk group, the false positive group have a higher occurrence of PROM, Down’ s screening is a potential high risk in-dex as a predictor of PROM.

5.Inhibitory effects of midazolam on amygdala kindling in rats and maximal electroshock seizure in mice

BACKGROUND: Typically antiepileptic drugs, such as phenobarbital, fenitoina sodica and diazepam, can inhibit amygdala kindling effect in rats. However, whether midazolam has the same effect is still unclear.OBJECTIVE: To investigate the inhibitory effect of midazolam on amygdala kindling onset in rats and maximal electroshock seizure (MES) in mice and effeots of antiepileptic drugs.DESIGN: This study was divided into three subexperiments, including the effects of midazolam on amygdala kindling onset, independent activities and incidence convulsion. All the three subexperiments were completely randomized,infra-group control or self-control studies.SETTING: Medical College of Qingdao University.MATERIALS: The experiment was carried out in the Comprehensive Experimental Room, Affiliated Hospital of Medical College of Qingdao University from August 2004 to March 2005. Nine Wistar rats weighing (250±10) g and 120 Kunming mice weighing (20±5) g and either gender were provided by Animal Center of Qingdao Institute of Drug Control.Midazolam (5 g/L) was provided by Xuzhou Enhua Drugs Co., Ltd. (batch number: 20030706).METHODS: ① Establishment of amygdala kindling models: Nine kindled rats were randomly selected and intraperitoneally injected with 0.25, 0.5 and 1 mg/kg midazolam, respectively. Quadri-pathway biological signal processing system (SMUP-PC) was used to measure discharge duration (ADD) and Racine's stage. ② Sixty mice were randomly divided into 5 groups, including saline group, 40 mg/kg phenobarbital group, 0.5, 1.0 and 1.5 mg/kg midazolam groups with 12 mice in each group. And then, numbers of activities in a unit time (times per 5 minutes) were determined by XZC-4A mini-animals independent activity instrument. ③ MES models were established to calculate incidence of convulsion.MATN OUTCOME MEASURES: Effects of midazolam on ADD, Racine's stage, numbers of independent activities and incidence of convulsion.RESULTS: All the 9 rats and the 120 mice were involved in the final analysis. ① Effect of midazolam on amygdala kindling onset: After intraperitoneal injection of 0.5 and 1.0 mg/kg midazolam, ADD and Racine's stage were obviously lower than those before administration (P < 0.05-0.01). After intraperitoneal injection of 0.25 mg/kg midazolam, ADD was obviously lower than that before administration (P < 0.05), but there was no significant difference in Racine's stage. ②Effect of midazolam on independent activities of mice: Numbers of independent activities were lower in the phenobarbital group and 0.5, 1.0 and 1.5 mg/kg midazolam groups than those in the saline group (P < 0.01), while numbers of independent activities were higher in 0.5 mg/kg midazolam group than those in the phenobarbital group (P < 0.05). ③Effect of midazolam on maximal electroshock seizure: Incidence of convulsion was lower in the phenobarbital group and 0.5, 1.0 and 1.5 mg/kg midazolam groups than that in the saline group (P < 0.05-0.01), while Incidence of convulsion was higher in 0.5 mg/kg midazolam group than that in the phenobarbital group (P< 0.05).CONCLUSION: Midazolam can significantly inhibit amygdala kindling onset, reduce numbers of independent activities,and antagonist MES in mice.
6.Ethics Problems in Administration of ICU

ICU is a special department for critically ill person.For the reasons of the closed-type of administration and the peculiar psychological state of patient,some ethics issues came out.The patients and their relative can't receive the due respect and humanism care,and the ethics issue hasn't drawn enough attention.The author holds that to solve the ethical problems in the administration,we should pay attention to the basic principle of ethics of life,reinforce the humanistic morals of the medical personnel in ICU,and apply the principle of people first in clinic.

Objective To investigate the effects of atorvastatin on cardiovascular remodeling in renovascular hypertensive rats. 9.A new method for screening latent tuberculosis infection in Beijing army recruits

Objective To investigate the Mycobacterium tuberculosis infections in 2014 among Beijing army recruits, and evaluate a new method for screening latent tuberculosis infections.Methods A total of 194 army recruits were subjected to chest X-ray examination purified protein derivative(PPD) skin test, antibody detection, and interferon gamma release assay(IGRA) by ELISA combined with recombinant protein CFP10-ESAT6 and latent infection protein Rv2628.Results The positive rates of PPD skin test and antibody test were 49.7% and 15.5%, respectively.The latent infection rate of IGRA test was 22.2% in 194 cases after CFP10-ESAT6 stimulation.After stimulation of latent tuberculosis infection(LTBI) with Rv2628, IFN-γ level was significantly higher than that in healthy control group (P<0.05).The weak positive group of TST (5 mm≤diameter<15 mm) had a significantly higher level of IFN-γ than the strong positive group(diameter≥15 mm)(P<0.05),but after stimulation with CFP10-ESAT6,IFN-γ levels were not significantly different between the two groups(P>0.05).There was no significant difference between antibody negative and positive groups after stimulation by CFP10-ESAT6 and Rv2628 (P>0.05).The area under the ROC curve of Rv2628 diagnosis of tuberculosis infection was 0.84.When Youden index was 0.621,the specificity was 94.7% and sensitivity was 67.4%.ConclusionCombined detection of antigens Rv2628 and CFP10-ESAT6 specific IFN-γ values can be potentially used for differential diagnosis of active or latent tuberculosis infections.
10.Inhibition of miR-375 decreases lipoapoptosis in NIT-1 β cells

Objective To investigate the effect of miR-375 inhibited by 2'-O-me-375 on lipoapoptosis of NIT-1 pancreatic β cells. Methods NIT-1 cells were divided and treated according to the optimal condition: mock (without lipofectamine) ,lipofectamine( transfected only with lipofectamine) ,NC-miRNA (transfected with negative control miRNA) ,and 2'-O-me-375( transfected with 2'-O-me-375) groups. 72 hours later, all cells in each group were cultured with 500 μmol/L palmitate for 48 h. The percentage of apoptotic cells was detected by Hochest33342 staining and terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end-labeling (TUNEL). The protein expression of myotrophin ( V1 ) , a target gene of miR-375, was detected by Western blotting. Results Compared to the other three groups,the cell apoptosis rate of 2'-O-me-375 group was the lowest (P<0.01) .along with the highest VI expression level(P<0. 01). Conclusion Inhibition of miR-375 decreases pancreatic (3-cell lipoapoptosis.
