1.Awareness and infection of Toxoplasma gondii in married childbearing wom-en in Chengde Region

Objective To understand Toxoplasma gondii infection and awareness condition of married childbearing women in Chengde Region so as to provide the evidence for the establishment of control measures. Methods Totally 733 married childbearing women who took physical examination in Chengde Hospital of Traditional Chinese Medicine from July to December in 2013 were investigated by questionnaire to understand the awareness condition on T. gondii infection then 490 women among them from 3 counties and 2 districts were randomly chosen to detect the Toxoplasma antibodies by ELISA. Results A total of 733 questionnaires were returned and 126 women knew related knowledge about T. gondii infection and the awareness rate was 17.19% 126/733 . Sixty-three women were determined as infected cases and the infection rate was 12.86% 63/490 . The infection rates of the women who with higher educational level working as medical staff and living in urban were lower and the awareness rates of them were higher. Conclusions The infection rate of T. gondii among the married childbearing women in Chengde Region is high and the awareness rate of them is low. In order to decrease the infection rate as well as to increase the awareness rate of the population the health education should be strengthened.
2.Resistin concentration in healthy volunteers, patients with obesity, diabetes mellitus and hyperthyroidism

Conclusions There is an increasing trend of resistin level in healthy volunteers and patients with obesity, diabetes mellitus and hyperthyroidism in order Resistin also was a positive and an inverse correlation with BMI and resistance index respectively. These results support the hypothesis of Steppan that resistin causes insulin resistance and links obesity to diabetes, and that the high level of resistin in hyperthyroidism can help to explain the phenomenon of its insulin resistance.
3.Performance evaluation of nine commonly used urine albumin assay systems

Objective By investigating precision,linearity and accuracy of 9 commonly urine albumin assay systems (8 of immuno-turbidimetric assays and 1 of immuno-nephlometric assay),and comparing the concordance of measurement results,to elucidate the quality of the existing analytical systems.Methods Referring to Clinical and Laboratory Standards Institute (CLSI) EP15-A2,two mixed urines with U-Alb levels of 20 mg/L and 200 mg/L were made to validate precision; Referring to CLSI EP14-A2,fourty fresh urines were selected to evaluate matrix effect of saline diluted European Reference Materials (ERM) DA 470 and saline diluted urine,also to reflect the variation of measurement results among systems; Referring to EP6-A,saline diluted urines (10 levels) were made to validate linearity; Taking the theoretical concentration of precisely saline-diluted ERM-DA 470 as the target value,accuracy of each assay system was evaluated.Maximal allowable coefficient variation (CV) of ≤ 15% was taken as the acceptable precision for each assay system,as rccommcnded by International Federation of Clinical Chemistry (IFCC)-and National Kidney Disease Education Program (NKDEP) ; maximum allowable bias of ≤25% was taken as criteria for accuracy evaluation as used in Proficiency Test (PT) sponsored by College of American Pathologists (CAP).Results At level of micro-albuminuria(20-200 mg/L),all 9 systems total CVs were ≤ 15% ; No matrix effect or interference were found in saline diluted ERM DA 470 and saline diluted urine.For A,B,E,F,G and I systems,validated linear regions were close to those stated in kit instruction;For C,D and H systems,the lower limits of validated linear region (18.7,3.6 and 12.0 mg/L,respectively) were higher than those stated in kits instruction (0,0.9 and 5.0 mg/L,respectively) ;For B and C systems,the lower limits of validated linear region were close to the upper limits of reference interval stated in kit instruction.When urine albumin was ≤ 12.6 mg/L,A,E,F,G and I systems showed good accuracy,absolute biases at all dilution were below 3 mg/L,D system showed higher positive bias (5.0-14.4 mg/L),B,C and H systems' biases were not evaluated because of high in-batch CV (the CV of B system≥ 18.1%,of C system ≥ 14.5%,of H system ≥ 39.1%); when U-Alb ranged in 25.2-201.0 mg/L,all 8 systems' relative biases were ≤25%,except D systems,which showed an un-acceptable positive bias (15.9%-44.3%).Good concordance among systems' results was present at level of microalbuminuria(20-200 mg/L),with CV among systems < 15% ;when urine albumin was < 20 mg/L,CV among systems increased as allumin concentration decreased.The main contribution of variation came from B,C and H systems,which lower limits of linearity were relatively high.Conclusions At level of microalbuminuria(20-200 mg/L),except D system,the other 8 systems show good precision and accuracy;at low level of urine albumin(<20 mg/L,especially < 10 mg/L),precision and accuracy of some systems(B,C and H system) needs to be improved.

Urinary excretion of albumin indicates kidney damage and is recognized as a risk factor for progression of kidney disease and cardiovascular disease,resulting a widespread clinical utilization of urine albumin measurement.Considerable inter-method difference has been reported for urine albumin,there are no available reference materials and no reference measurement procedures for urine albumin.This review discusses the clinical utilization of urine albumin in recent years,the principles of existing measurement systems and difference of results among systems,and the current status of work about reference materials and reference measurement procedures.
5.Preliminary Study on the Relationship between the Spleen Apparent Diffusion Coefficient Value,Portal Hypertension and Hypersplenism

6.Macroscopic and microscopic identification of Chinese herb belonging to genus Senecio.

The medicinal herbs derived from genus Senecio have been commonly used in Chinese medicine and triggered attention in recent decades for that they contain the hepatotoxic pyrrolizidine alkaloids. Therefore the botanical pharmacognostic study to authenticate those herbs based on their macroscopic and microscopic characteristics is important for the assurance of safety when they are applied as raw material for extracts or for finished products. In this paper, 13 taxa (11 species and 2 varieties) of Senecio plants were collected and their macroscopic and microscopic characteristics were observed and described by digital microscopic illustration. The results showed that the distribution of collenchyma in the cortex, the level of development for pericycle, the location of the phloem, and the ratio of pith in transverse sections of the stems, and the morphology of the leaf epidermal cells, the stomatal types and the non-glandular hairs in leaf surface view were found to be the main microscopic characteristics for authentication of different Senecio species. The herbs derived from genus Senecio can be distinguished from each other on the basis of their macroscopic and microscopic characteristics, and those observation can be used for the identification of commercial crude drugs from Senecio plants.
7.MicroRNA expression in hepatocytes with hydrogen peroxide-induced oxidative stress-injury and alleviating effect of mesenchymal stem cell-conditioned medium

AIM: To evaluate the changes of microRNA (miRNA) in hepatocytes during hydrogen peroxide-induced oxidative stress injury, and to observe the alleviating effect of mesenchymal stem cell-conditioned medium (MSC-CM) in this progress.METHODS: The hepatocyte oxidative stress injury model was established using hydrogen peroxide and human normal liver cell line L02.MSC-CM was prepared using centrifugation and filter.The effects of MSC-CM on hepatocyte injury were evaluated by apoptosis analysis, cell viability detection, cell cycle, and mitochondrial membrane po-tential (MMP).Twenty-one differentially expressed miRNAs were selected by gene chip hybridization, in which miR-143, miR-145, miR-301a and let-7a were confirmed by RT-qPCR.Bioinformatics software was utilized to predict target proteins of these miRNAs, and then the proteins were verified by Western blot.RESULTS: MSC-CM markedly attenuated hydrogen peroxide-induced oxidative stress injury by reducing apoptosis, promoting cell viability and regulating cell cycle.The ex-pression of miR-143, miR-145, miR-301a and let-7a, indentified by RT-qPCR, increased under the condition of oxidative stress injury, while decreased after MSC-CM treatment.The expression of miR-143 predicted target proteins, HK2 and ADRB1, decreased under the hydrogen peroxide-exposure, while increased after MSC-CM treatment, which is consistent with the regulatory trend of miR-143.CONCLUSION: MSC-CM might attenuate hydrogen peroxide induced oxidative stress injury via inhibiting apoptosis and regulating some miRNA expression.
8.Effects of retinal ganglion cell apoptosis on delayed rectifier K+currents

AIM: To investigate the effects of rat retinal ganglion cell (RGC) apoptosis on delayed rectifier K+currents (IK).METHODS:The retinas of 2~3 d newborn Sprague-Dawley rats were dissociated into cell suspension by trypsin digestion .The RGCs were cultured and divided into control group , pressure 0.5 h group, pressure 1 h group, pressure 1.5 h group and pressure 2 h group.The cells were cultured regularly for 6 d in control group , and the cells in other groups were cultured regularly for 6 d and gave pressure of 80 mmHg for 0.5 h, 1 h, 1.5 h and 2 h, respectively. The rhodamine 123 fluorescence from labeled RGC mitochondrion was detected by continuous wavelength multifunctional microplate detection instrument.The membrane capacitance (Cm) in different groups and IK in the pressure 1 h group were recorded from the RGCs by whole-cell patch-clamp technique .RESULTS:No difference of rhodamine 123 fluorescence in the RGC mitochondria between control group and pressure 0.5 h group was observed .Rhodamine 123 fluorescence in the other 3 groups was significantly lower than that in control group (P<0.05).No difference of the Cm between control group and pressure 0.5 h group was found, and the Cm in the other 3 groups was significantly lower than that in control group (P<0.05).The amplitudes of IK were higher than that in control group .At the test potential from -10 mV to 60 mV, the current density in pressure 1 h group was significantly higher than that in control group (P<0.05).The maximal conduc-tion ( Gmax ) in pressure 1 h group was significantly higher than that in control group .The voltage for IK channel half-activa-tion ( V1/2 ) in pressure 1 h group declined comparison with control group ( P<0.01 ) , and the k value had no significant difference between the 2 groups.CONCLUSION:Retinal ganglion cell apoptosis accompanies with delayed rectifier K +current enhancement .
9.Evaluation of an immunochemiluminescence assay for human plasma B-type natriuretic peptide(BNP) and the reference interval of BNP

Objective To evaluate an immunochemiluminescence assay for human plasma B-type natriuretic peptide(BNP),and to investigate the reference interval of BNP in healthy Chinese.Methods EDTA anti-coagulant plasmas from 421 healthy volunteers(202 men and 219 women) were included.BNP was measured with Triage BNP kit developed by Biosite on the Beckman Coulter Access II.Referring to the EP documents,a series of tests were designed to investigate the assay′s linearity,detection limit,sensitivity,precision,interference factors,and BNP′s stability under different storage conditions.Results The assay had a linearity of 15~5 040 ng/L,and the detection limit was 9.2 ng/L.The concentration at 10% of CV was 30 ng/L.There was no obvious interference in the assay when bilirubin was 75 years old persons was 100.7 ng/L.Conclusion The immunochemiluminescence assay for human BNP is highly sensitive.The characteristic of its wide detection linearity,precision,and anti-interference ability meets the clinical requirement.We recommend that the 97.5% percentile be a reference interval;when the age is 55 years old,the value is
10.Change and Clinical Significance of Peripheral Blood DN Tregs in Patients with Systemic Lupus Erythematosus

Objective To study the expression and function of TCRαβ+ CD3+ CD4-CD8-T (Double negative regulatory T cells,DN Tregs) in peripheral blood of Systemic lupus erythematosus (SLE) patients,and investigate their function in pathogenesis of SLE.Methods TCRαβ+ CD4-CD8 T cells were quantified as percentage of total CD3+ T cells in peripheral blood from 20 SLE patients and 20 healthy controls by flow cytometry.Used ELISA to test the cytokine levels (IFN-γ,TNF-α,IL-6,IL-17A) in the plasma of SLE patients.And analyzed the relationship between the percentage of DN Tregs and cytokines levels.Results The ratio of DN Tregs in TCRαβ+ T cells was significantly increased in SLE patients compared to healthy donors (t=3.54,P<0.01).The levels of IFN-γ,TNF-α and IL-6 in plasma of SLE patients were higher than healthy donors (t=2.824,2.085,2.304,P<0.05).The frequency of DN Tregs was found to correlate with IFN-γ (r=0.52,P=0.02) but not TNF-α (r=0.17,P=0.16) and IL-6 (r=0.47,P=0.49).Conclusion This study reveals frequency of DN Tregs in peripheral blood of SLE patients was higher than healthy controls,the frequency of DN Tregs was also found to correlate with IFN-γ levels,which means that DN Tregs may play an important role in pathogenesis of SLE.
