1.Impacts of Ruangan-Lidan tang on the the immune function and survival for liver cancer patients receiving TACE combined with 3D conformal radiotherapy

Objective To observe the impacts of Ruangan-Lidan Tang on immune function for liver cancer patients receiving TACE combined with 3D conformal radiotherapy, evaluation of short-term efficacy, survival, to explore the changes of cellular immune function in the treatment. Methods 86 cases of liver cancer patients were randomly divided into treatment group and control group were taken in 3D conformal radiotherapy to 50-60GY/5-6 week for fourth weeks after TACE, the treatment group taking Ruangan-Lidan Tang intervention, control group without the use of traditional Chinese medicine intervention.Before the start, after 1 weeks of TACE, within 3 days before radiotherapy,end of radiotherapy, a total of 4 times for T cell differentiation antigen were measured, and between two groups were compared with t test, 2 months after radiotherapy CT or MRI evaluation of curative effect, survival. Results 5 cases of loss of cases, the total cases were 81 patients, including 40 cases of treatment group, 41 cases in After 1 weeks of TACE, CD3+, CD4+, CD8+of the treatment group were (55.15±4.76)%, (31.88±5.50)%, (22.00±3.18)% of the control group were (53.39±5.00)%, (30.49±5.94)%, (23.39±3.41)%. There was no significant difference between two groups (P>0.05); Before radiotherapy, CD3+, CD4+, CD8+ of the treatment group were (59.05±5.91)%, (40.55±6.17)%, (18.63±3.14)%, of the control group were 55.88±4.65%,33.88±4.41%,22.00±3.78%. There was significant difference between two groups(P<0.01);At the end of radiotherapy, CD3+,CD4+,CD8+ of the treatment group were (61.03±5.54)%, (42.65±4.57)%, (18.10±3.20)%, of the control group were (55.56±5.14)%, (32.95±4.01)%, (23.32±2.69)%. There was significant difference between two groups (P<0.01). Short-term efficacy(CR+PR)of the treatment group was higher compared with the control group(82.50%, 75.61%), but no significant difference (U=1.177, P>0.05). In the treatment group, the median survival time was 22 months,the average survival time was 21.7 months, the half year survival rate was (97.5± 2.5)%(39/40);In the control group, the median survival time was 20 months, the average survival time was 19.3 months, the half year survival rate was (94.9±3.5)% (37/41). Follow-up survival analysis, treatment group than in the control group increased, but there was no statistical difference (P=0.132, P>0.05). Conclusion Ruangan-Lidan Tang can improve the immune function of liver cancer patients receiving TACE combined with radiotherapy, but in the Short-term efficacy and survival did not reflect advantage.
2.Effect of platform switching technique on bone loss in the posterior area of the mandible in the first year following dental implantation

BACKGROUND: Peri-implant crestal bone resorption is considered as a normal reaction according to traditional butt-joint prosthesis around two-piece implants. OBJECTIVE: To evaluate bone loss around two-piece implants restored according to the platform switching concept in the posterior area of the mandible. METHODS: Eighty-eight implants were consecutively placed in 50 patients following submerged surgical protocol. In the test group, 40 Ankylos implants were placed using platform switching technique, while 48 3i implants using traditional butt-joint in the control group. All the implants were positioned at the crestal level. Definitive prosthesis was delivered after 3 months. RESULTS AND CONCLUSION: All implants were judged to be successfully osseointegrated. The success rate was 100%. Twelve months after restoration, radiographic analysis showed an average bone reduction level of (0.31±0.39) mm in the test group that was statistically significantly different (P < 0.01) from the average reduction in the control group [(0.94±0.43) mm]. In the follow-up time, no implants were loose or lost and healthy gingival was seen. These findings show that the platform switching technique can reduce peri-implant crestal bone resorption and preserve alveolar bone level around dental implants 12 months after restoration.Oral Implantology Center, Affiliated Hospital of Medical College, Qingdao University, Qingdao 266003, Shandong Province, China
3.The clinical observation in the prophylactic effect of EEN on pulmonary infection in patients with severe traumatic brain injury

Objective: To investigate the prophylactic effect of EEN on pulmonary infection in patients with severe traumatic brain injury.Methods: 60 cases of young adults with severe brain injury were randomized into the early enteral nutrition group(experimental group) or the control group.The experimental group was feeded 12~24 hours after injury or surgery and the control group was feeded 24 h~5 d after injury or surgery.The double-sugar test method was used for determination of intestinal barrier function,and the duration of pulmonary infection was recorded.Results: The lactulose/mannitol ratio in experimental group was significantly lower than in control group on the 7th postoperative day.The average body temperature and duration of pulmonary infection in experimental group were significantly lower than in control group.According to ADL scores,the daily capacity of convalescent patients in experimental group was significantly better than in control group.Conclusion: The early enteral nutrition can improve mucosal barrier function,reduce the incidence of pulmonary infection and improve overall prognosis in patients with severe traumatic brain injury.
4.Relationship between human membrane associated sialidase ( Neu3 ) and multidrug resistance (MDR) in K562 cell line

Objective To investigate the role of human membrane associated sialidase (Neu3) in multidrug resistance ( MDR) of K562 cells. Methods K562 cells and K562/ADM cells were treated with DNR alone or with combination of DNR and NeuAC2en. The cell survival rate was measured by MTT assay; the expression of MDR related factors and apoptosis related factors were determined by Western blot and RT-PCR; Neu3 activity was detected by TBA reaction. Results The survival rate of K562/ADM cells was higher than that of K562 cells; NeuAC2en showed synergistic effect with DNR(P <0. 01) ; Neu3 activity of K562 and K562/ADM cells decreased after DNR or NeuAC2en induction and the decrease was most significant in the combination treatment group (P <0. 01). Under the identical condition, the protein expression of P-gp and Neu3 and the mRNA expression of MDR1, Neu3, BCL-xl and BCL-2 increased comparing with K562 cells. The mRNA level of BAX in K562 and K562/ADM groupsdid not changed significantly. MDR1, Neu3,BCL-xl and BCL-2 decreased after DNR induction and down-regulated most obviously in the groups treated by DNR combined with NeuAC2en. Conclusion Neu3 may be related with multidrug resistance of K562/ADM cell through regulating apoptosis and the expression of MDR1.
5.Effects of prolonged heavy load swimming and the Panaxadiol saponins on expression of ?-actin of quadriceps in rats

AIM: To study the effects of 7 weeks prolonged haevy load swimming and Panaxadiol Saponins (PDS) on the expression of ?-actin in quadriceps muscle of thigh in rats. METHODS: After 7 weeks prolonged heavy load swimming training, the expression of ?-actin in quadriceps muscle of thigh was quantitatively examined by Northern blotting analysis in the condition with or without PDS treatment. RESULTS: The expression of ?-actin in rat quadriceps muscle of thigh in exercise+PDS group was significantly higher than that in sedentray control group and exercise+saline group at 24 th hour after intensive training. No statistical difference between exercise+androsan group and exercise+saline group was observed.CONCLUSION: PDS enhanced ?-actin expression in quadriceps muscle of thigh in rats.

In order to provide experimental data for the development and application of drug in clinic, we determined the effects of extracts from different parts of folium perillae (L. ) Britt. on hemorheological parameters, extracted from leaves (folium perillae), seeds (fructus perillae) and peduncles (caulis perillae). The results showed that all extracts from different parts of folium perillae (L. ) Britt. can significantly reduce the whole blood viscosity at low shear rate (10 s(-1)), erythrocyte aggregation index, erythrocyte electrophoresis index (P<0.05), and the whole blood reductive viscosity at low shear rate (10 s(-1)) (P<0.01). Extracts from folium perillae and caulis perillae can significantly decrease erythrocyte deformation index (P<0.05), whereas extracts from fructus perillae can not. Extracts from fructus perillae and caulis perillae can significantly decrease plasma viscosity at low shear rate(10 s(-1)), but extracts of folium perillae can not. Aspirin can only decrease the whole blood reductive viscosity at low shear rate and plasma viscosity (P<0.05). All extracts from different parts of folium perillae (L. ) Britt. had no significant effects on hematocrit, erythrocyte rigidity index, fibrinogen concentration , the whole blood viscosity and the whole blood reductive viscosity at middle and high shear rate (60 s(-1),120 s(-1)).

8.Reconstruction and expression of green fluorescent protein and aquaporin 7 fusion recombinant vector.

OBJECTIVETo study the membrane mobility of aquaporin 7 (AQP7) by cloning stably transfected CHO cells with expression of pEGFP-C1-AQP7, in which AQP7 cDNA was fused downstream and in frame to pEGFP-C1 gene.
METHODSThe full sequence of AQP7 was amplified by RT-PCR and then recombined in the downstream of the green fluorescent protein gene in the pEGFP-C1 vector. The recombinant vector pEGFP-C1-AQP7 was stably transfected into CHO cells. With fluorescent microscopy, immunocytochemical stain and Western blot, pEGFP-C1-AQP7 showed a predominant intracellular vesicular localization.
RESULTS(1) The sequence of AQP7 cDNA of the Wistar rat was logged into the GenBank (access number: AY157737). (2) Identification demonstrated that pEGFP-C1-AQP7 fusion protein stably expressed in CHO cells. (3) With fluorescence microscopy, pEGFP-C1-AQP7 showed a predominant intracellular vesicular localization.
CONCLUSIONThe CHO cell line with stable pEGFP-C1-AQP7 expression was set up successfully for advanced research.

9.Discovery of thiosemicarbazone derivatives as effective New Delhi metallo-β-lactamase-1 (NDM-1) inhibitors against NDM-1 producing clinical isolates

New Delhi metallo-β-lactamase-1 (NDM-1) is capable of hydrolyzing nearly all β-lactam antibiotics, posing an emerging threat to public health. There are currently less effective treatment options for treating NDM-1 positive “superbug”, and no promising NDM-1 inhibitors were used in clinical practice. In this study, structure–activity relationship based on thiosemicarbazone derivatives was systematically characterized and their potential activities combined with meropenem (MEM) were evaluated. Compounds 19bg and 19bh exhibited excellent activity against 10 NDM-positive isolate clinical isolates in reversing MEM resistance. Further studies demonstrated compounds 19bg and 19bh were uncompetitive NDM-1 inhibitors with Ki = 0.63 and 0.44 μmol/L, respectively. Molecular docking speculated that compounds 19bg and 19bh were most likely to bind in the allosteric pocket which would affect the catalytic effect of NDM-1 on the substrate meropenem. Toxicity evaluation experiment showed that no hemolysis activities even at concentrations of 1000 mg/mL against red blood cells. In vivo experimental results showed combination of MEM and compound 19bh was markedly effective in treating infections caused by NDM-1 positive strain and prolonging the survival time of sepsis mice. Our finding showed that compound 19bh might be a promising lead in developing new inhibitor to treat NDM-1 producing superbug.
