1.Study on the Genotoxicity of Organic Extracts From Fishpond Water Containing Sewage

Objective To evaluate the genotoxicity of the organic extracts in the fishpond water containing the sewage. Methods Healthy Kunming mice were randomly divided into 6 groups according to the body weight, 8 mice in each group. The mice were treated with organic extracts through intraperitoneal injection at the doses of 25 L/kg, 12.5 L/kg, 6.25 L/kg, 3.125 L/kg water sample respectively for 3 consecutive days. Genotoxicity induced by organic extracts from the fishpond water containing the sewage was detected by the mice bone marrow micronucleus test and un-fluorescent staining comet assay of mice peripheral blood lymphocytes. Objective:To evaluate the changes of urinary and sexual function after pelvic autonomic nerve preservation with total mesorectal excision operation in patients of rectal cancer . Methods:Fifty-two patients of Dukes B stage received The PANP following TME with follow-up was complete in 38 cases while TME in 35 cases of Dukes B from January 2004 to August 2006. The urinary and sexual dysfunctions of patients in case and control group were analyzed retrospectively by case-control study. Urinary and sexual function were evaluated by means of questionnaires before and after operation.Results:The micturation dysfunction rate was 15.7%(6/38) and 40%(14/35) ,the sexual dysfunction rate was 18.4%(7/38) and 51.4%(18/35)in case and control group respectively,with the difference being significant(P=0.013,P=0.002).Conclusion:The PANP following TME for rectal cancer could reduce sexual and micturition disorder rate and improve the quality of life of the patients after operation.
4.Transgenic Atractylodes macrocephala with double defense genes exhibiting resistanceto Rhizoctonia solani

Objective The transgenic Atractylodes macrocephala resistance to Rhizoctonia solani was obtained by gene engineering. Methods On the base of the efficient regeneration system of Baizhu via shoot organogenesis, the rice chitinase gene (RCH10) and the alfalfa ?-1, 3-glucanase gene (AGLU) were tandem-inserted into the transformation vector pB101, which was transformed into A. macrocephala with gene gun. Transformants were confirmed by PCR, GUS assay, and disease resistant. Results Twenty-five independent transformants possessed desired genes were observed by PCR detection, and among them five transformants exhibiting resistance to Rhizoctonia solani. Conclusion The disease resistant variety has been obtained by transformation, which provides a shorten avenue for the direct introduction of novel traits into A. macrocephala through genetic engineering without the need for numerous back-crossing in breeding programs that slow down cultivar improvement, meanwhile it improves disease resistant gene pool.
5.Role of hepatic microcirculatory disturbance induced by intestinal endotoxemia in liver injury

Objective To investigate the effect of hepatic microcirculatory disturbance induced by intestinal endotoxemia in liver injury. Methods The model of rats with intestinal endotoxemia induced by thioacetamide(TAA) was established. 25 Male Wistar rats were divided into 4 groups as normal control group (N), Heparin control group(H), TAA treated group(T), and TAA + heparin treated group(T +H). The changes of plasma biochemistry and hepatic histopathology were measured. Results The plasma endotoxin level, alanine transaminase (ALT) activity and malondialdehyde (MDA) content in TAA treated rats were markedly higher than that in the control ones (P < 0.05), while plasma endotoxin level was lower ( P > 0.05) and ALT as well as MDA were decreased significantly ( P < 0.05) in TAA + heparin group than in TAA group. Conclusion Intestinal endotoxemia could induce disturbance of hepatic microcirculation, which results in ischemia and hypoxia of liver cell. Heparin could not only correct disturbance of hepatic microcirculation induced by intestinal endotoxemia, but also reduce liver injury induced by ischemia and hypoxia.

7.Effect of neuregulin-1βcombined with captopril on myocardial cell apop-tosis and related gene expression in rats with chronic heart failure

AIM:To study the effect of neuregulin-1 ( NRG-1β) or captopril alone and their combination on myocardial cell apoptosis and related gene expression in rats with chronic heart failure .METHODS:Young male Sprague-Dawley rats were randomly divided into sham-operated group, heart failure group, NRG-1βgroup, captopril group and combined treatment group .The volume overloaded heart failure model in the rats was established by aortocaval fistula .The cardiac function was evaluated by determining the changes of hemodynamics and plasma BNP level .The apoptotic index ( AI) of myocardial cells was assayed by TUNEL .The protein levels of p-Akt, Bax and Bcl-2 in left ventricular tissue were detected by Western blot .RESULTS: The LVEDP, AI and the expression of Bax were significantly lower in NRG-1βgroup, captopril group and combined treatment group than those in heart failure group ( P<0.05 ) , while the values of ±dp/dtmax and the protein levels of p-Akt and Bcl-2 were significantly higher in NRG-1βgroup, captopril group and com-bined treatment group than those in heart failure group (P<0.05).The effect of combined treatment group was more signifi-cant ( P<0.05) than that in NRG-1βgroup and captopril group .CONCLUSION: Neuregulin-1βor captopril alone and the combination of them effectively improve the cardiac function and inhibit myocardial cell apoptosis in chronic heart failure rats.The therapeutic effect of combination of NRG-1βand captopril is better than that of NRG-1βor captopril alone .
8.Effect of pravastatin on expression of HCN channel in post-myocardial infarction rats

Objective To investigate the effects of pravastatin on the expressions of hyperpolarization-activated cyclic nucleotide-gated channel subunits,HCN2 and HCN4 of right ventricular after myocardial infarction (MI) in rats. Methods Sprague-Dawley rats MI model was induced by ligating the left anterior descending coronary artery,and then 20 MI rats were randomly and equally divided into 24-hour group,1-week group,4-week group and pravastatin group (20 mg?kg-1?d-1 for 4 weeks by gavage). Another 5 rats served as sham-operation group. Real-time PCR and Western blotting were used to measure the mRNA and protein expressions of HCN2 and HCN4 in the heart tissues of each group. Results The mRNA and protein expressions of HCN2,and mRNA expression of HCN4 were up-regulated in 4-week group and pravastatin group compared with sham-operation group. The expression of HCN2 in pravastatin group were markedly attenuated compared with 4-week group. The protein expression of HCN4 was not detected in all groups. Conclusion The expression of HCN2 and HCN4 were up-regulated in myocardial infarction rats. Pravastatin may reverse the up-regulated expressions of HCN2 at mRNA and protein levels.
9.Clinical value of dual-source CT in assessing adult congenital heart disease

Objective To investigate the clinical value of dual‐source CT (DSCT ) in assessing adult congenital heart disease (ACHD) .Methods The data in 29 ACHD patients with DSCT were retrospectively analyzed .Results Among 29 cases ,the major‐ity suffered from two or more cardiovascular malformations ,in which 20 cases had intracardiac malformations ,including 4 cases (13 .79% ) tetralogy of Fallot ,6 cases (20 .69% ) were atrial septal defect ,4 cases(13 .79% ) were bicuspid aortic valve (13 .79% ) and 8 cases (27 .58% )were ventricular septal defect ;19 cases had extracardiac malformations ,in which 3 cases(10 .34% ) were co‐arctation of the aorta ,4 cases (13 .79% ) were patent ductus arteriosus ,3 cases (10 .34% ) were transposition of the great arteries and 5 cases (17 .24% ) were anomalous pulmonary venous connection .Conclusion DSCT could not only clearly demonstrate the cardiovascular malformations in ACHD patients ,but also conduct the observation on the lung ,trachea and bronchia ,which can pro‐vide detailed anatomic information for surgery .
10.Clinical Observation of Torasemide in the Treatment of Chronic Heart Failure

OBJECTIVE:To observe clinical efficacy and safety of torasemide in the treatment of chronic heart failure(CHF). METHODS:94 patients with CHF were selected and randomly divided into control group and observation group,with 47 cases in each group. Control group were treated with routine treatment,such as bed rest,oxygen inhalation,control of total body fluid in-take,routine anti-heart failure therapy and use of diuretic in 24 h. Based on the above treatment,observation group were treated with Torasemide injection with initial dose of 5-10 mg,qd,gradually increasing to 20 mg/d,maximal does of 40 mg/d. Both group re-ceived 7 days of continuous treatment. Clinical efficacies were observed in 2 groups as well as serum potassium and sodium,Scr,24 h urine volume and the rate ofⅠ-Ⅱgrade cardiac function before and after treatment. The occurrence of ADR was compared between 2 groups. RESULTS:The total effective rate of observation group was 61.70%,which was significantly higher than that of control group (46.81%),with statistical significance (P<0.05). There was no statistical significance in serum potassium and sodium and Scr between 2 groups before and after treatment(P>0.05). There was no statistical significance in 24 h urine volume and the rate ofⅠ-Ⅱgrade cardiac function between 2 groups before and after treatment(P>0.05);after treatment,24 h urine volume and the rate ofⅠ-Ⅱgrade cardiac function of 2 groups were increased significantly,the observation group was higher than the control group, with statistical significance (P<0.05). There was no statistical significance in the incidence of ADR between 2 groups (P>0.05). CONCLUSIONS:Torasemide is effective for CHF,and can promote diuresis and the recovery of cardiac function with good safety.
