1.Studies on inhibitive effect of low-voltage electrochemotherapy on head and neck cancer-bearing nude mice

Objective:To analyse the therapic effect of low-voltage electrochemotherapy (EP) on the head and neck cancer.Method:YCU-N861 human head and neck cancer cells were subcutaneously inoculated in the neck of nude mice to establish tumor-bearing models. Then the mice were treated with bleomycin(BLM, 20 μg per mouse), accompanied with electric stimulation(50 V or 100 V).Result:BLM treatment applied with EP can significantly inhibite the proliferation of tumor cells than BLM or EP treatment seperately(P<0.01) . The tumor volume, the mitotic index (MI) and the labeling index (LI) of Ki67 in BLM+EP group were all obviously decreased than BLM group, EP group and control group (P<0.01).Conclusion:Low-voltage electrochemotherapy can offer promising mothod in the local treatment of head and neck cancer.

we reviewed data from 547EEGs on 400 patients with epilepsy in order to determine the probability in finding the interictal epileptiform activity (IIEA) with serial EEGs. In 49.5% of our patients, the abnormaeity is present on the first record. In 53.5% by the second EEG, and in 54.25% by the third or above. There is relatively little yield to serial EEGs beyond this point. Patient's ages, types of seizures and causes are negatively correlated to IIEA yield

4.Discussion on the cultivation of research type clinical postgraduates of neurology

Neurology is a professional science. The content of neurotomia, sub-speciality knowl-edge and auxiliary examination are abundant, and meanwhile it is rather difficult. Clinical graduates after graduation are malnly engaged in clinical work, clinical services, and will play a variety of roles in the course of their clinical practice so it is necessary for them to learn comprehensive knowledge. This article introduced experience of clinical postgraduates tralning of neurology, such as doctor-patient communication skills, basic theoretical study (neurology, neurotomia and sub-speciality knowledge), clinical thinking, and assistant examination, clinical research capacity during these 18month. It will provide the theoretical and practical knowledge in the future work.
5.Clinical curative effects of internal sphincterotomy in the posterior midline and lateral position in treatment of patients with chronic anal fissure

Objective To investigate the curative effects of partial internal sphincterotomy in the posterior midline and lateral position in treatment of chronic anal fissure. Methods One hundred and fifty cases chronic anal fissure patients with stageⅡ or Ⅲ were randomly divided into observation group(80 cases)and control group(70 cases),who were hospitalized in the Shunyi District Hospital of Beijing from Dec. 2009 to Oct. 2012. The patients of two group were treatment by internal sphincterotomy either in the posterior midline or lateral position respectively. The efficacy,healing time,complications and recurrence of the two groups were recorded and compared. Results There were 76 cases with cure,6 cases with improvement and 2 with ineffective effect in the observer group. There were 49 cases with cure,12 cased with improvement and 9 cases with ineffective effect in control group. The difference of the efficacy was statistically significant( P =0. 025). Wound healing time of observation group and control group were(9. 9 ± 0. 8)d and(17. 1 ± 0. 9)d,and the difference was significant( P =0. 048 ). The incidence of anal exudates in observer group was lower than the control group(2. 5%(2/80)vs. 24. 3%(17/70);P=0. 032). The incidence of recurrence in observer group was 5. 0%(4/80),lower than the control group(24. 3%(17/70),P=0. 046). The degree of the postoperative pain,voiding dysfunction and wound edema in observer group were lower than the other group( P =0. 025, 0. 025,0. 049). Conclusion The effect of internal sphincterotomy in the lateral position in treatment of chronic anal fissure is better with quickly repair,fewer complications.
6.Efficacy of Cefoperazone/sulbactam on Severe Nosocomial Lower Respiratory Infection: A Clinical Analysis

OBJECTIVE To investigate the clinical and bacterial efficacy and safety of cefoperazone/sulbactam(Sulperazone) on severe nosocomial lower respiratory infections. METHODS The clinical materials in the treatment of severe nosocomical lower respiratory infections from Mar 2002 to Mar 2005 were analyzed retrospectively. RESULTS The overall clinical efficacy rate of Sulperazone was 83.9%,the bacterial eradication rate was 92.%,The sensitivity rate to Sulperazone was 88.2%.There was no adverse reaction in the observation. CONCLUSIONS Sulperazone is effective and safe in the treatment of severe nosocomial lower respiratory infection.
7.Cloning of rat B cell lymphoma-2 gene and construction of its lentiviral vector

Objective To clone the full-length cDNA of rat B cell lymphoma-2(bcl-2) gene,then construct and identify the cytomegavirus-mediated lentiviral expression vector of bcl-2 gene,and assess the gene expression in 293T cell,which is a human embryonic kidney cell line.Methods The full-length bcl-2-cDNA fragment was amplified by reverse transcription polymerase chain reaction(RT-PCR) from the kidney tissue of a Wistar rat.The double-stranded oligonucleotides(dsOligoe) were then cloned into the pMD18-T plasmid.After confirmation of a correct construction by sequencing,the positive clone was subcloned into pGC-FU vector with enhanced green fluorescent protein(EGFP),and then transformed into DH5a competent cells.The restricted endonuclease and T4 DNA ligase were used to construct the lentiviral expression vector plasmid pGC-FU-bcl-2 which,combined with the lentiviral packing materials(pHelper 1.0,pHelper 2.0),was then transfected into 293T cell line to form the recombinant lentivirus pGC-FU-bcl-2,and it was used to transfect the 293T cells.The expression of pGC-FU-bcl-2 was further verified by detecting EGFP and bcl-2.Results 1) It was verified by DNA sequencing that the sequence of rat bcl-2 gene was consistent with reported sequence in GenBank.2) The bcl-2 gene was successfully combined in pGC-FU-bcl-2 recombinant plasmid which could be transfected into human embryonic kidney cells.3) The recombinant virus pGC-FU-bcl-2 could be obtained from the 293T cells by co-transfection of pGC-FU-bcl-2 and packing plasmids.4) Targeting gene could be cloned into 293T cells by the recombinant lentivirus with steady expression.The fluorescent protein could be observed under microscope and the expression of bcl-2 protein was detected by Western blotting.Conclusions The lentiviral expression vector containing EGFP and bcl-2 gene has been successfully constructed,with which the transfected 293 T cells can lead to a steady expression of bcl-2 protein.The present study provides a basis for the further study of the function of bcl-2 gene and a potential therapy for the diseases relating to apoptosis.
8.Cloning and sequencing of rat bcl-2 gene riched guanine and cytosine

BACKGROUND: Excessive apoptosis of ovary granulosa cell is a dominant cause of premature ovarian failure and bcl-2 gene is able to inhibit cell apoptosis. But studies demonstrate that,the guanine and cytosine (GC) content reaches 60% in the rat bcl-2 gene sequence. This gene cannot be amplified using routine polymerase chain reaction method. OBJECTIVE: To clone and identify the bcl-2 gene riched GC. DESIGN,TIME AND SETTING: Open experiment was finished in the Laboratory of Medicine and Molecular Biology,Life Science School of Sun Yat-sen University from May to December in 2007. MATERIALS: Wistar rats were purchased from Experimental Animal Center of Sun Yat-sen University. Ecoli DH5?was preserved by Laboratory of Medicine and Molecular Biology,Life Science School of Sun Yat-sen University; pMD18-T vector was purchased from Takara Biotechnology (Dalian) Co.,Ltd. METHODS: The bcl-2-cDNA,in which GC accounted for 60.6%,was obtained by modified reverse transcription-polymerase chain reaction from kidney tissue of Wistar rats,and was cloned into vector-pMD18-T. Characterizations and sequencing of the pMD18-T-bcl-2 were carried out by polymerase chain reaction screening of individual bacterial colonies. MAIN OUTCOME MEASURES: Cloning and purification of bcl-2 gene cDNA; results after connecting bcl-2 gene cDNA to pMD18-T vector and transducting Ecoli DH5?,identification of positive clone and results of sequencing. RESULTS: The bcl-2 gene was identified by the clone and DNA sequencing. DNA sequence analysis was consistent with Genebank sequence,with a 99% homology. CONCLUSION: The gene riched GC is difficult to be amplified,bcl-2-cDNA can be cloned and constructed into cloning vector pMD18-T successfully by the efficient technique for other genes riched GC.
9.Clinical study of amoxicillin combined with pectin bismuth in the treatment of chronic atrophic gastritis

Objective To investigate the clinical efficacy of amoxicillin combined with pectin in the treatment of chronic atrophic gastritis. Methods According to the different treatment methods will be January 2015 to March 2017 patients with chronic atrophic gastritis in our hospital 80 cases groups:control group with clarithromycin+amoxicillin+bismuth pectin bismuth pectin treatment,observation group treatment;detailed records of two groups before and after treatment(pathological atrophy,intestinal metaplasia,dysplasia,chronic the inflammatory activity and the integral changes),the levels of inflammatory factors(IL-2/IL-6/CRP)incidence level changes and adverse reaction,the obtained related data for comparative analysis. Results Amoxicillin and bismuth pectin(observation group)in treatment of chronic atrophic gastritis is better than clarithromycin+bismuth pectin(control group)treatment,patients with pathological score decreased than the control group,the levels of inflammatory factors is better than the control group,the incidence rate of adverse reaction was lower than the control group,the difference was statistically significant(P<0.05). Conclusion Chronic atrophic gastritis patients choose amoxicillin and bismuth pectin treatment effect significantly,can greatly reduce the pathological integral,improve clinical symptoms,and can reduce the level of inflammatory factors in patients with a low incidence of adverse reactions,it is worthy of widely used to treat patients with chronic.
10.Comparison of sedative effects of different fractions from patrinia scabiosaefolia fisch in mice

Objective To investigate the sedative effects of different fractions of patrinia scabiosaefolia fisch. in mice and provide some valuable information for isolating components with sedative effect.Methods Petroleum, acetic ether and n-butanol fraction was isolated from the patrinia scabiosaefolia fisch.. The sedative effects were determined with locomotor activity assay, hole board test and sodium thiopental-induced sleeping time assay, and it was compared with the effects of total 70% ethanol extract. Results N-butanol fraction showed better sedative effect compared with others, and it was in a dose-dependent manner. Conclusion The results showed that the main active principles of patrinia scabiosaefolia fisch. were the n-butanol fraction, and the promising active chemical might be obtained from this fraction.
