Objective To investigate the effects of Xanthoceraside on the learning and memory impairment induced in mire by innacere-broventricular injection of aggregated amyloid β peptide _(1-42)(Aβ_(1-42)). Methods Learning and memory functions in mice were examined us-ing step-through test and water maze test. Biochemical determination of acetylcholinesterase (AchE) and choline acetyl transferase (ChAT) were measured with spectrophotometric melhod. Results Administration of Xanthoceraside reduced number of error and prolonged the laten-cy in step-through test in mice impaired by Aβ_(1-42) (P < 0.05,P< 0.01,respectively). In water maze test, the swimming time decreased in mice treated with Xanthoceraside compared with the model mice impaired by Aβ_(1-42) (P< 0.05,P< 0.01,respectively). The results of bio-chemical determination showed that decrease level of AchE and ChAT in mice impaired by Aβ_(1-42) were significantly ameliorated by Xantho-ceraside administration (P < 0.05 and P < 0.01). Conclusion Xanthoceraside has the effect of improving learning and memory impairment in mice inducel by intracerebroventricular injection of Aβ_(1-42) via enhancing the cholinergic system functions.

Objective To compare the blood glucose level and associated hypoglycemia risks between group of insulin glargine combined with repaglinide and group of mixed insulin in the treatment of type 2 diabetes in the elderly. Methods Sixty four type 2 diabetes patients (age≥60 years) with inadequate glyeaemic control by drugs were divided into 2 groups randomly: glargine group (32 cases) and mixed insulin group (32 cases). In glargine group, 32 patients were given repaglinide before breakfast, lunch, supper respectively and injection of glargine hypodermically once at 22 o'clock every night, while the 32 patients in mixed insulin group were injected with the mixed insulin (Novolin 30R) hypodermically before breakfast and supper respectively for 16 weeks. The dose of repaglinide and insulin were adjusted every 3 days according to the level of fasting glucose (FPG)and postprandial glucose (PPG), reaching the aim of FPG less than 7.2 mmol/L and PPG less than 10mmol/L. The blood glucose level and the incidence of hypoglycemia were observed. Results The daily glucose profile and the level of HbAlc of the 2 groups dropped obviously after 16 weeks treatments (all P<0. 05). At the end of the experiment, the PPG of lunch and supper, and the level of HbAlc were markedly lower in glargine group than in mixed insulin group (all P<0. 05), and the body mass index (BMI) increased markedly in mixed insulin group compared with pre-experiment (P<0. 05), but no significant change was found in glargine group (P>0. 05). The incidence of hypoglycemia in glargine group was lower than that in mixed insulin group (2 patients in glargine group, 12 patients in mixed insulin group, P<0.05). Conclusions Both of the mixed insulin and glargine combined with repaglinide have visible effects on controlling the blood glucose, but the latter has better efficacy, lower risk of hypoglycemia and lower increase of BMI than the former.

Objective To prepare and identify monoclonal antibody against LRR-WSC domain of polycystin-1 and to investigate the distribution of polycystin-1 in kidney tissues and kidney cell lines. Methods BALB/c mice were immunized with fusion protein PC1-e of polycystin-1 LRR-WSC domain. The splenocytes were fused with myeloma cells by PEG 4000 and the hybridomas were selected in HAT medium. The hybridoma clones secreting antibodies against polycystin-1 LRR-WSC domain were detected by enzyme-linked immunosorbent assay ( ELISA) and cloned by limiting dilution. The specificity of anti-polycystin-1 LRR-WSC domain monoclonal antibody from hybridoma was verified by ELISA and Western blot. The distribution of polycystin-1 in tissues and cells was detected by immunohistochemical method. Results One cell line of hybridoma secreting monoclonal antibody against polycystin-1 was established. Western blot analysis showed that the monoclonal antibody reacted strongly and specifically to polycystin-1 LRR-WSC domain. Distribution of polycystin-1 in fetal kidney was localized in tubular epithelium. In normal adult kidney tissues, our study showed that polycystin-1 was mainly expressed in the medullary collecting ducts and distal convoluted tubules. Positive staining was also found in the majority of cyst-lining epithelial ceEs of cystic tissue from autosomal dominant polycystic kidney disease ( ADPKD) patients. Expressions of polycystin-1 were found in either ADPKD cyst-lining epithelia cell line and LLC-PK1, clearly plasma membrane and intracytoplasmic staining of polycystin-1 were observed. Conclusion Specific monoclonal antibody against polycystin-1 LRR-WSC domain were obtained. The antibody is important to researching the mechanism of ADPKD. The distribution of polycystin-1 in kidney tissues and cells show that polycystin-1 was important in tubular elongation and the maintenance of tubular architecture.

In this research, a new set of training methods was created and put into practice to train the body's sit-to-stand balancing ability, and controlling ability of body's gravity centre of patients with cerebrovascular diseases. It contains a series of motion trainings, including sitting position training, standing position training, sit-to-stand training and game training, etc. There is also a series of trainings of feedback included in this set of methods, such as internal feedback, external feedback, bandwidth feedback, conclusive feedback, visual feedback, acoustical feedback and so on. The trainings resulted in better controlling ability of body's centre of gravity in the sit-to-stand process of patients after a period of training. It indicated that these targeted-designed trainings, especially the feedback trainings, could effectively help the under-trained patients to have a better knowledge of their current situation, and in turn to have a better knowledge of which aspects they should take more training to achieve a better therapeutic effect.

Journal of Biomedical Engineering 2013;30(2):320-326
This article is aimed to present a design of a testing device for knee joint kinematics parameters of patients with strokes. We used Auto CAD to design the knee joint sensor device, and applied multifunction data acquisition card to collect sensor data. Then we transferred the data to the computer to quantitatively detect and analyze the joint angle, angular velocity and other parameters. The software system used Visual C+ + MFC frame and shared database Access, and used ADO technology to realize the collection between the software system and patient information system. After a preliminary test of 20 healthy subjects, the result showed that intraclass correlation coefficient (ICC) was more than 0.8, indicating a good reliability of the instrument.
