1.Studies on optimizing extraction technology of Euphorbia hirta L.by central composite design-response surface methodology

Objective To optimize the extraction process of Euphorbia hirta L.by Central composite design/response surface methodology with ethanol by the ultrasonic extraction technology.Methods Based on the single factor experiment and central composite design-response surface methodology,the content of quercitroside was regarded as evaluation index,select ethanol concentration,extraction time,and solid-liquid ratio as three evaluation factors to achieve the purpose. Results The results indicated that the optimal extraction process was obtained as follows:10 times the amount of 60% ethanol under ultrasound for 2 times,50 min each;the average content of the quercitroside was 2.39 mg/g.Conclusion The method to optimize the extraction process is simple and reasonable.
2.Effects of obestatin on amylase release of pancreatic acinar cells and lobules of rats

ObjectiveTo investigate the effects of different doses of obestatin on amylase secretion of pancreatic acinar and lobules of rats in vitro.MethodsPancreatic acinar cells of rats were separated in vitro and incubated with different doses of obestatin (0,0.1,1,10,30 nmol/L) for 1h,another group of pancreatic acinar ceils was incubated with obestatin for 30 min,then was incubated with 100 pmol/L CCK-8 for another 30 min.Pancreatic lobules,which containing intrapancreatic nerve terminals and islets,were prepared and were incubated with different concentrations of obestatin for 30 min at 37℃ with or without 75 mmol/L KCl.Amylase levels in the supernatants,acinar cells and pancreatic lobules were calculated as a percentage of total amylase content.ResultsObestatin (0,0.1,1,10,30 nmol/L) produced no significant change in basal amylase release of acinar cells [ (3.48 ± 1.44) %,(3.70 ±- 1.39) %,(3.36 ± 1.24) %,(3.86 ± 1.41 ) %,(4.54 ± 2.01 ) % ].CCK-8 significantly increased amylase release of acinar cells [ ( 13.84 ± 2.63 ) % vs (3.48 ± 1.44)%,P <0.05],but obestatin (0.1,1,10 nmol/L) has no effect on the amylase release inducedby CCK-8 [(14.55 ± 1.7)%,(13.79 ± 1.81)%,(14.39 ± 1.12)%].Obestatin (0.1,1,10,30 nmol/L) did not affect the amylase release of pancreatic lobuole.KCl significantly increased anylase release,which was ( 1.84 ± 0.29 ) folds higher than that of control group ( P < 0.05 ),but obestatin has no effect on the amylase release induced by KCl,which were (2.01 ± 0.30 ),( 1.89 ± 0.41 ),( 1.74 ± 0.14 ),( 1.88± 0.33) folds higher than those of control group.ConclusionsExogenous obestatin has no effects on pancreatic exocrine of acinar cells and lobules of rats in vitro and cannot block or assist the increased amylase release induced by CCK-8 or KCl.
3.The change of serum inflammatory mediators in patients undergoing tension-free inguinal hernia repair

ObjectiveThis study was to evaluate the changes of inflamatory mediators in patients undergoing tension free hernioplasty(LH) using expanded polytetrafluoroethylene (e PTFE) mycromesh biomaterial or conventional Bassini hernia repair(BH). MethodTwenty one patients randomized into two groups receiving LH or BH. Peripheral venous blood samples were collected 24 hours prior to surgery and then 6,24,48 and 168 hours postoperatively.ResultLH patients showed a higher increased serum level of fibringen,C reactive protein,alpha 1 antitrypsin, and interleukin 6 than BH patients. Postoperative visual analogue scales for pain reduced on mobilization for patients undergoing LH compared with BH. Neutrophils significantly increased only in LH compared with baseline. ConclusionOur data show that although LH induces less pain and rapid postoperative recovery,it is associated with an higher inflammatory response compared with BH, likely due to mycromesh biomaterial.
4.The Survey and Study on HBsAg Carrier at Different Period After Administered Hepatitis B Vaccine

Objective To analyze the positive rate of HBsAg in different population at different periods after widely using Hepatitis B Vaccine and provide strategy for Hepatitis B Control.Methods Using ELISA method to detect the HBsAg carrier in polulation under 18 years old in local area.Results After six year investigating,the HBsAg carrier rate in tested population decreased.For 0-4,4-8,8-12,12-15,15-18 age group,the carrier rate were 3.76% vs 0.30%,4.74% vs 1.39%,5.59% vs 2.52%,8.51% vs 3.03%,and 12.58% vs 2.98% in 2000 and 2006.Conclusion The reduction in carrier rate had a direct relationship with Hepatitis B Vaccine inoculation.Hepatitis B Vaccine had important effect to Hepatitis B control.We should gradually intensify Hepatitis B Vaccine inoculation.It is not only for newborn but also for adult.
5.Identification of the nanobacteria in prostatic fluid of CPPS

Objective To identify the nanobaeteria in prostate fluid of patients with CPPS.Methods Expressed prostatic secretion(EPS)and urine specimens were collected by Meares-Stamey way from CPPS patients(n=100)and normal controls(n=100).The specimens were cultured and nanobacteria was identified by indirect immunofluoreseenee staining with rnonoelonal antibody.The morphological features were observed by using transmission electron microscopy(TEM). Results The positive rate of nanobaeteria in the EPS cdture of CPPS patients and controls were 43% and 5% respectively,with significant statistical difference(X2=39.58,P<0.01).By TEM,the sizes of NB ranged from 100 to 500 nm and appeared eoccoid-ccccobacillary shape. Conclusion Nanobaeteria infection may exist in EPS of CPPS patients.
6.Influence of rotation manipulation on Raman spectral characteristics of stable plaques in a rabbit model of early carotid atherosclerosis

BACKGROUND:It is generaly recognized that cervical rotation manipulation can increase the risk of detachment of unstable plaques in carotid atherosclerosis, but few studies are reported on the influence of cervical rotation manipulation on the stable plaque in early carotid atherosclerosis. OBJECTIVE:To explore the influence of the cervical rotation manipulation on the lipid contents in carotid atherosclerotic plaque in a rabbit model of early carotid atherosclerosis. METHODS:After being fed for 15 days with normal diet, 30 male New Zealand White rabbits were further fed for 18 weeks with normal diet (n = 10; control group) or a high-fat diet containing 2% cholesterol, 10% lard and 88% normal granules to build rabbit models of early carotid atherosclerosis with stable carotid plaque (n = 20). At 14 weeks of feeding with high-fat diet, the experimental rabbits fed with high-fat diet were randomly divided into a cervical rotation manipulation group (n = 10) and a model group (n = 10). The rabbits in the cervical rotation manipulation group underwent cervical rotation manipulation to the left and right sides, once each side, with the maximal range of rotation. Total five cervical rotation manipulations, once every 3 days, were performed. RESULETS AND CONCLUSION:The 1 450 cm-1 and 1 660 cm-1 peaks of the Raman spectrum of lipid in the carotid atherosclerotic plaque of rabbit models were not distinctly present in the control group, however, they were obviously observed in the cervical rotation manipulation group and model group. Nevertheless, the relative intensity differences at spectrum characteristic peaks were not significant between cervical rotation manipulation group and model group (P > 0.05). The environmental findings indicate that the lipid content in carotid atherosclerotic plaque of rabbit models of early carotid atherosclerosis cannot be increased after short-term administration of cervical rotation manipulation.
7.Relationship of blood glucose, blood lipids, blood uric acid and fibrinogen level, and prognosis of acute lacunar cerebral infarction patients

ObjectiveTo investigate the relationship of blood glucose,blood lipids,serum uric acid,fibrinogen level and prognosis of patients with acute lacunar cerebral infarction.MethodsClirical data of 127 cases with acute lacunar cerebral infarction were retrospectively analyed.Patients with normal blood glucose,blood lipid,uric acid and fibrinogen level were enrolled into A group of 64 cases,63 cases with abnormal levels were selected as group B,an average of 2 years of follow-up,observing the two groups of neurologic,re-infarction incidence and prevalence of vascular dementia were obsenved in two groups.ResultsAfter 4 weeks of treatment,A group of neural function defect score(9.6 ±7.3) was significantly lower than group B( 14.3 ±7.4) (t =3.8696,P <0.05) ;95.4% total effective rate in A group was significantly higher than B group of 82.0% ( x2 =5.8623,P < 0.05 ).After an average follow-up of 2 years,A group of recurrent cerebral infarction rate( 18.8% ),vascular dementia rates(7.8% ),were significantly lower than group B(28.6%),17.5%(11/63)(x2 =6.5421,5.9623,P<0.05).ConclusionA number of abnormal serum biochemical indexes had a significant impact on short and long-term prognosis of acute lacunar cerebral infarction.

OBJECTIVE:Aspirin sustained release capsules(A-SRC) were prepared The release rate and bioavailability of A-SRC were studies METHODS:To study release mechanism by determining dissolution rate and to study relative bioavailability of A-SRC by determining serum concentrations in rabbits RESULTS:The release profile of A-SRC fitted zero-order kinetics This sustained release preparation had good release stability and high bioavailability in comparison with commonly-used aspirin sustained release capsules CONCLUSION:A-SRC showed obviously sustained release effect The drug concentration in blood kept steady and lasted long after oral administration,therefore the times of administration could be decreased Because the light-excipients were used in A-SRC,which had floating properties and could make drug release slowly A-SRC could decrease the irritations to gastrointestinal tract and other side effects
9.The gamma knife radiosurgery for cavernous sinus meningiomas

Objective To analyse the therapeutic outcome of cavernous sinus meningiomas by gamma knife radiosurgery. Methods Retrospective analysis was conducted of 37 patients with cavernous sinus meningiomas. The median maximum diameter was 2.6 cm (1.4~3.2 cm) with the median dose to the tumor margin of 15.7Gy(8~22 Gy), the isodose curve to the tumor margin of 40%~60% and the target numbers of 1~9. The radiosurgical dose to the optic nerve and brain stem was not more than 10 Gy and 15 Gy, respectively. Results After an average follow up of 22.6 months(ranging 4~43 months), unchanged symptoms were found in 22 cases(59.5%), alleviated in 12 cases(32.4%) and complications of optic and trigeminal nerve in 3 cases(8.1%). By MR image analysis, unchanged size was found in 18 cases(48.6%), smaller size in 15 cases(40.5%) and larger size in 4 cases(10 8%). 10.Study on the molecular mechanism of C-phycocyanin from Spirulina platensis induced apoptosis in HeLa cells

Aim To investigate the influence and molecular mechanism of C-phycocyanin(CPC) from Spirulina platensis on apoptosis of HeLa cells in vitro.Methods Firstly,the effect of purified CPC on proliferation of HeLa cells in vitro was determined by MTT assay,and then electron microscope was exploited to observe the characteristic apoptotic features of cells treated with CPC.Subsequently,genomic DNA changes of HeLa cells were observed by agarose electrophoresis.Flow cytometric analysis revealed the influence of different concentrations of CPC on cell cycle of HeLa cells.The expressions of apoptosis related genes of CPC treated HeLa cells were determined by immunohistochemistry analysis.In addition,the activities of caspases and the release of cytochrome c from mitochondria into the cytosol were detected.Results Compared with control cells untreated with CPC,a significant decreased in the numbers of HeLa cells in survival treated with CPC and concentration dose effects existed.CPC could induce characteristic apoptotic features including cell shrinkage,membrane blebbing,microvilli loss,chromatin margination and condensation into dense granules or blocks.DNA of HeLa cells treated with CPC showed fragmentation pattern(DNA ladder of oligomers of 180~200 bp) typical for apoptotic cells.HeLa cells treated with different concentrations of CPC demonstrated an increasing percentage of cells in sub-G0/G1 phase.In addition,CPC could promote the expression of pro-apoptotic gene(Fas and ICAM-1);meanwhile,held back the anti-apoptotic gene(Bcl-2) expression,and then facilitated the transduction of tumoral apoptosis signals that resulted in the apoptosis of HeLa cells in vitro.In CPC treated HeLa cells,CPC treatment of HeLa cells also resulted in activation of caspases and release of cytochrome C from mitochondria into the cytosol.Conclusion C-phycocyanin from Spirulina platensis can induce the apoptosis of HeLa cells in vitro.By virtue of the promotion of the apoptosis signals transduction in HeLa,CPC realizes its antitumor activities.
