1.A Study of the acupuncture point embedding therapy on slow transit constipation along with colon transmission function,plasma SP and VIP

Objective To explore the clinical efficacy of acupuncture point embedding therapy in the treatment of slow transit constipation (STC)and the influence on the colon transmission function,plasma substance P(SP), vasoactive intestinal peptide(VIP). Methods A total of 128 slow transit constipation patients were involved in the study and divided randomly into two groups by using stochastic indicator: a treatment group (treated with acupuncture point embedding therapy) and a control group (treated with lactulose oral liquid). The clinical effects, colon transmission function, the influence of SP, and VIP were observed for STC patients. Results In the treatment group, CCS score was (13.52±3.93) before treatment and(8.15±3.77) after the treatment. In the control group CCS score was (14.11±4.24) and (9.01±2.93) respectively before and after the treatment, CCS score in both groups showed significantly improvement after the treatment (P<0.01), and there was no significant difference between groups (P>0.05). The plasma SP and VIP level of the two groups after the treatment [treatment group (46.73±1.50) ng/L,(206.10±12.14) ng/L, the control group(40.92±6.25)ng/L,(214.73± 7.62)ng/L] were also improved than before [treatment group was(26.66±0.88)ng/L, (332.68±17.84)ng/L, the control group was (23.32±1.75)ng/L,(306.22±20.01)ng/L], with statistically significant(P<0.05), and the difference between the two groups were also significant(P<0.05).Conclusion The acupuncture point embedding therapy is effective for STC, and it can regulate the level of SP and VIP.
2.Familial Hepatits B Virus Genotypes and Clinical Prognosis

OBJECTIVE To investigate the distribution of familial hepatitis B virus (HBV) genotypes and analyze the relationship between genotypes and clinical outcome. METHODS A total of 136 cases of familial hepatitis B were involved in this study,including 25 cases of asymptomatiec HBV carriers (ASC),85 cases of chronic hepatits B (CHB),24 cases of liver cirrhosis (LC) and 4 cases of hepatocellular carcinoma (HCC). Nested polymerase chain reaction-restriction fragment length polymorphism (ntPCR-RFLP)was used to identify the genotypes of HBV. RESULTS There were 124 cases with hepatitis genotype C,7 cases with genotypes B+C mixed,3 cases with genotype B and 2 cases that did not conformed to be any of genotypes from A to H. Genotype C in the HCC was predominant genotype (100%),next in the LC(96%),CHB (95.3%) and ASC(66.7%).Compared with genotype C,genotypes B and B+C only existed in ASC and CHB,the proportion was 9.5% vs 23.8% and 1.2% vs 2.3%,respectively. There were no other HBV genotypes from A to H seen in familial hepatitis B in Shanxi province. CONCLUSIONS HBV genotype C is the major one in familial hepatitis B in Shanxi. There are a few genotypes B and B+C existed. Genotype C induces a relatively bad prognosis.
3.Clinical Observation of Brucea Javanica Oil Injection Combined with Oxaliplatin and Xeloda in the Treat-ment of Elderly Patients with Advanced Gastric Cancer

OBJECTIVE:To observe the clinical short-term efficacy and safety of brucea javanica oil injection combined with oxaliplatin and xeloda in the treatment of elderly patients with advanced gastric cancer. METHODS:82 elderly patients with ad-vanced gastric cancer were randomly divided into control group and observation group. Control group was treated with Oxaliplatin injection 130 mg/m2 added into 5% Glucose injection 250 ml for 2 h,iv,d1+Xeloda tablets 1 000 mg/m2,orally,twice a day, d1-14;based on the treatment of control group,observation group was additionally treated with 10% Brucea javanica oil injection 30 ml added into 0.9% Sodium chloride injection 250 ml,iv,once a day,d1-14. 21 days were as a treatment course,and it lasted 3 courses. The short-term efficacy,life quality and toxicity reactions were evaluated in 2 groups. RESULTS:The short-term efficacy in observation group was significantly higher than control group,life quality was significantly better than control group,and the in-cidences of leucopenia and liver damage were significantly lower than control group(P<0.05). CONCLUSIONS:Brucea javanica oil injection combined with oxaliplatin and xeloda has better efficacy than oxaliplatin combined with xeloda in the treatment of elder-ly patients with advanced gastric cancer,with good safety.
4.The high-risk factors for bronchopulmonary dysplasia in premature infants of mechanical ventilation

Objective To explore the incidence rate and high-risk factors for bronchopulmonary dysplasia(BPD) in premature infants of mechanical ventilation.Methods From Jan 2008 to Dec 2009,196 very low birth weight infants with gestational age less than 32 weeks in our NICU were enrolled in the study.61 cases had the histories of mechanical ventilation.The clinical materials of 21 cases with BPD were compared to 41 cases without BPD in retrospective analysis.Results The total incidence of BPD in the very low birth weight infants with gestational age less than 32 weeks was 10.7%,and 34.4% in those infants with mechanical ventilation.The gestational age and birth weight of BPD group were lower than those without BPD,and the rates of premature rupture of fetal membrane and complicated neonatal respiratory distress syndrome (NRDS),postnatal infection,with long time intravenous nutrition in BPD group were higher than those without BPO control (P<0.05).Gender,asphyxia history,the use of pulmonary surfactant (PS),positive end-expiratory pressure,and mean airway pressure were not different significantly between two groups (P>0.05).Peak inspiratory pressure (PIP),the time of mechanical ventilation and oxygen administration were different significantly between two groups (P<0.05).NRDS,postnatal infection,the duration of ventilation,and hyperoxia were the risk factors for BPD,and the regression coefficient were 3.683,1.541,1.188 and 1.647.Conclusion Preventing premature,low birth weight,NRDS,shortening the duration of high peak inspiratory pressure and hyperoxia with mechanical ventilation and the use of the parenteral nutrition,managing the infection of the prenatal and postnatal are the key points of preventing BPD.
5.Research on the relationship between ecchymosis physique and the uterus myoma

Objective To summarize the tending physique of hysteromyoma patients and thus to provide the clinical prevention and treatment methods for the disease.Methods Clinical epidemiology investigation and study methods were adopted to design the unified investigation and study table.Questionnaire survey were performed stochastically to 150 hysteromyoma patients and 150 normal females.Various physique characteristics were recorded.Results Ecchymosis physique were commonly seen in hysteromyoma patients,accounting for 36.7% in all patients.Conclusion The occurrence of hysteromyoma was closely related to ecchymosis physique.

8.Comparation of gastrointestinal absorption studies of specnuezhenide with salidroside in rats.

The experiment was designed to study the mechanism of increasing efficiency of Ligustrum lucidum steamed with wine. Rats in vivo with gastrointestinal perfusion model were used. The contents of salidroside and specnuezhenide in the fluid of gastrointestinal perfusion of rats were measured by HPLC at different time points after dosing. Then the K(a) and absorption percentage were calculated. Specnuezhenide could be detected in the fluid of gastrointestinal perfusion of specnuezhenide. The K(a) of the specnuezhenide and salidroside in the fetal intestines are 0.055 3 and 0.144 2 h(-1) respectively and the total absorptivity are 24.46% and 60.14% respectively after 4 hours. The K(a) in the stomach are 5.70 and 8.26 h(-1) respectively and the total absorptivity are 34.21% and 47.23% respectively after 4 hours. The experiment proved that specnuezhenide can be metabolized into salidroside which is more beneficial for gastrointestinal absorption. The experiment proved that specnuezhenide can be metabolized into salidroside both in the rat's stomach and the fetal intestine and compared with the specnuezhenide salidroside is more conducive to gastrointestinal absorption. The results suggested that the increasing efficiency on liver and kidney of L. lucidum steamed with wine has business with the fact that Specnuezhe nide is more conducive to the body after it is changed into salidroside.

9.Extracelluar matrix stimulates neurite outgrowth of dorsal root ganglion neurons differently depending on whether extracted from degenerated or normal intervertebral disk

BACKGROUND:Compared with the normal intervertebral disk, the density of nerve fibers and number of nerve endings and neuropeptides appear to be more in the degenerated intervertebral disk. However, this phenomenon does not occur in the normal y aged disk.
OBJECTIVE:To evaluate the axonal growth and induction of a painful neuropeptide and substance P using rat dorsal root ganglion neurons and degenerated human disc cells in vitro.
METHODS:The human intervertebral discs were harvested from patients with discogenic low back pain and normal people. And extracelluar matrix extracted from human degenerative intervertebral discs was cultured with rat dorsal root ganglion neurons. The promotion of axonal growth and induction of substance P of dorsal root ganglion neurons in extracted medium were evaluated through morphology observation and enzyme-linked immunosorbent assay, respectively.
RESULTS AND CONCLUSION:Compared with the normal group, the content of nerve growth factor in the degenerative group was significantly higher and the average length of neuritis was significantly longer in the experimental group (P<0.05). After intervention with anti-nerve growth factorβ, the average length of neuritis became remarkably shorter. The percentage of substance P-immunoreactive cells was significantly higher in the degenerative group compared with the normal group (P<0.001). Nerve growth factors that highly express in the extracellular matrix from the degenerative intervertebral dick can promote neurite outgrowth of dorsal root ganglion neurons and induce release of neuropeptides related to pain transmission.
10.Mesenchymal stem cells derived from adipose and nucleus pulposus tissue differentiate towards nucleus pulposus-like cells induced by transforming growth factor-beta 1

BACKGROUND:A large number of studies have shown that adult stem cels derived from multiple tissues are available to differentiate towards nucleus pulposus-like celsin vitro. It is unclear whether mesenchymal stem cels derived from nucleus pulposus tissues have the ability to differentiate towards nucleus pulposus-like phenotypes induced by transforming growth factor-beta 1. Up to now, there are few reports on the difference between the differentiation ability of mesenchymal stem cels derived from nucleus pulposus tissues and adipose-derived mesenchymal stem cels. OBJECTIVE:To compare the ability of mesenchymal stem cels derived from nucleus pulposus tissues and adipose-derived mesenchymal stem cels differentiating into nucleus pulposus-like cels under induction of transforming growth factor-beta 1. METHODS:The groin fat tissue and the coccygeal spine of rats were taken respectively to isolate and culture mesenchymal stem cels derived from nucleus pulposus tissues and adipose-derived mesenchymal stem cels by mechanical enzyme digestion method. Flow cytometry was employed to detect the expression of CD105, CD90, CD29, CD45, CD44, CD34, and CD24 of both two kinds of stem cels. Mesenchymal stem cels derived from nucleus pulposus tissues and adipose-derived mesenchymal stem cels were divided into complete induction group (complete induction medium with transforming growth factor-beta 1), incomplete induction group (complete induction medium without transforming growth factor-beta 1) and control group(DMEM/F12 containing 10% fetal bovine serum and 100 mg/L penicilin/streptomycin), respectively. After 14 days of culture, real-time PCR was used to detect the expression of colagen type II, Aggrecan and SOX-9 in each group. RESULTS AND CONCLUSION:CD105, CD90, CD29 expressed positively and CD45, CD44, CD34, CD24 negatively in both two kinds of stem cels. After 14 days of induced differentiation, the expressions of colagen type II, Aggrecan and SOX-9 in the two kinds of cels were significantly higher in the complete induction groups than in the control groups (P < 0.05). Under the induction of transforming growth factor-beta 1, the expression of colagen type II, Aggrecan and SOX-9 in mesenchymal stem cels derived from nucleus pulposus tissues was significantly higher than that in adipose-derived mesenchymal stem cels (P < 0.05). These findings suggest that both two kinds of mesenchymal stem cels have the ability to differentiate towards nucleus pulposus-like cels induced by transforming growth factor-beta, and mesenchymal stem cels derived from nucleus pulposus tissues may be more suitable as seed cels for nucleus pulposus tissue engineering research.
