In the light of developing trend and reality of domestic pharmaceutical supervision and management we propose the guidance of drug market, using the scientific method, making supervision as center, taking emphasis on basic units, strengthening in vertical section, popularizing in horizontal section, expanding scale etc as guiding thoughts. In order to strengthen the basic pharmaceutical supervision and management the suggestion of ten kinds of strategic changes must be realized.
6.Regulation of genistein on the levels of ER?, ER? mRNA in uterine endometrial cancer cells
Journal of Peking University(Health Sciences) 2004;0(03):-
Objective: To explore the mechanism of phytoestrogen (genistein) on uterine endometrial cancer cells and provide the theory foundation for using phytoestrogen in clinical practice. Methods: Uterine endometrial cancer cell lines (ishikawa) were cultured in vitro at different genistein concentration, and the expressions of the two estrogen subtypes of endometrial cancer cells were observed by real-time PCR. And two treatment groups, estradiol and progestin, were used as positive controls. Results:(1) Low concentration of genistein could enhance the expression level of ER? mRNA of endometrial cancer cells, which was obviously weaker than that of the estradiol group. On the other hand, low concentration of genistein could decrease the expression level of ER? mRNA of endometrial cancer cells, however estradiol had hardly any effect on this .(2) High concentration of genistein could decline the expression levels of ER? and ER? mRNA of endometrial cancer cells, whose effect was weaker than that of progestin. Conclusion: The expression levels of ER? mRNA and ER? mRNA of endometrial cancer cells vary with genistein concentrations. The experimental results suggest that genistein may act as estrogen receptor regulator.
8.Changes of tear film after trabeculectomy in glaucoma
International Eye Science 2015;(7):1201-1203
AlM: To learn the changes of the tear film before and after the trabeculectomy of glaucoma and explore the incidence of dry eye and the prevention and control measures.METHODS:The 36 patients (60 eyes) of glaucoma were examined in detail before 3d of trabeculectomy and after the surgery at 3, 7, 14 and 30d. The examinations include lower eyelid central river of tears, break-up time ( BUT) , Schirmer I test ( S I t ) and staining scores of corneal fluorescein under slit lamp microscope.RESULTS:The tear meniscus height of central lower eyelid was increased and the tear film BUT was shortened at the same time, the scores of S I t was reduced and corneal fluorescein staining score was increased at postoperative 3 and 7d compared with that of preoperation. The tear meniscus height of central lower eyelid, tear film BUT and Slt and score of corneal fluorescein staining began to recover in most of the affected eyes after surgery 14d. At 30d after surgery, 22%of patients tear film failed to recover to the preoperative level; dry eye occured in 18% preoperative eyes with normal tear film.CONCLUSlON:Trabeculectomy of glaucoma may affect the stability of the tear film and some patients showeing obvious dry eye and should be intervened and treatmented timely.
9.Effects of melatonin on retinal oxidative stress in short-term diabetic retina
Chinese Journal of Experimental Ophthalmology 2011;29(3):215-219
Background Oxidative stress is one of the pathogenesis mechanisms of diabetic complications.Melatonin is the most powerful antioxidant in living organism.Seldom study on the effects of melatonin on diabetic complications is found in China now. Objective This study aimed to detect the expression and alteration of malonadehyde(MDA),glutathione(GSH)and glial fibrillary acidic protein(GFAP)in retina with experimental diabetes and explore the effects of melatonin on retinal oxidative stress in diabetic rats. Methods The animal models of diabetes were established by injection of 2% streptozocin via caudal vein(STZ,45 mS/kg)in 48 6-week-old clean SD rats.Isometric citric acid buffer solution wag injected in 24 normal.rats as control group.Seven days after injection of STZ,melatonin(10 mg/kg)was injected into the abdominal cavity of 24 diabetic rats in melatonin group dailv, and the equal volume of normal saline was injected in other 24 diabetic rats(model rats)and 24 rats in normal control group. The animals were sacrificed and retinas were obtained in three groups in 4,6 and 8 weeks respectively.Immunohistochemistry and Western blot were used to detect the expression of GFAP in rat retina. The levels of MDA and GSH in retina homogenate were assayed by spectrophotometer. This experiment complied the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission. Results The expression of GSH in model group was lower than that in melatonin group or control group at 4,6 and 8 weeks (all P<0.05).However,no statistically significant difference was seen in the expression level of GSH between melatonin group and normal control group at 4 and 6 weeks(P>0.05),and in the eighth week, expression of GSH in melatonin group was lower than that in control group(P<0.05).The expressions of MDA and GFAP were more prominent in the model group than in melatonin group and control group in 4,6 and 8 weeks(P<0.05),but no significant difference was found between melatonin group and control group at 4 and 6 weeks(P>0.05).The expressions of MDA and GFAP in melatonin group were higher than those in control group at 8 weeks(P<0.05).Conclusion Melatonin plays a powerful effect in protecting the retina from oxidative stress in diabetes retina by downregulating the MDA level, upregulating GSH level and inhibiting GFAP expression.
