Objective:To explore the approach to analyzing simple effect on factorial design with SAS.Methods:The mean square of error coming from factorial design was considered as standard error(SE),and simple effect analysis under Contrast statement in GLM was conducted by designing the contrast coefficient matrix of the parameter,and SAS program was realized based on an example of 3-way factorial design.Results:he procedures of designing the contrast coefficient matrix of the pa-rameter were achieved.Conclusion:It is convenient and feasible to conduct simple effect analysis under Contrast statement by constructing design matrix.
2.Pregnancy after Loop Electrical Excision Procedure of Cervical Intraepithelial Neoplasia.

Objective To investigate the effects of loop electrical excision procedure(LEEP)on pregnancy of patients with cervical epithelial neoplasia(CIN).Methods 83 patients with cervical intraepithelial neoplasia grade II~III have been diagnosed.Cervical loop trip were 38 cases for all of the observation group,did not undergo cervical loop were 45 cases for all the control group.observed pregnancy.Results There are significant differences in premature delivery between two groups(P0.05).Conclusion Reasonable 1oop electrical excision procedure will not increase the risk of pregnancy.
3.Expression of cathepsin B, cathepsin L and uPA mRNA levels in non-Hodgkin lymphoma and their significance

Objective To investigate mRNA expression levels of cathepsin B,cathepsin L and uPA in non-Hodgkin lymphoma (NHL) samples,and analyze the relationship between three genes and clinicopathological factors.Methods Real-time fluorescence quantitative RT-PCR was used to quantify the relative expression levels of cathepsin B,cathepsin L and uPA mRNA in 39 NHL patients without chemotherapy or radiotherapy and 15 patients with lymph node reactive hyperplasia.Relationship between their expression and clinico-pathological factors was analyzed.Results mRNA expression levels of cathepsin B,cathepsin L and uPA in NHL group were significantly higher than those in the control group (0.403 vs 0.128,0.209 vs 0.057,0.459 vs 0.031,all P < 0.05).The mRNA expression levels of cathepsin B,cathepsin L and uPA were correlated with each other in NHL.The mRNA expression levels of cathepsin B,cathepsin L and uPA in advanced staged NHL were markedly higher than those in early staged NHL (0.453 vs 0.341,0.204 vs 0.085,0.372 vs 0.196,all P < 0.05).mRNA levels of cathepsin B,cathepsin L and uPA were higher in NHL patients sensitive to chemotherapy than in those resistant to chemotherapy(0.401 vs 0.556,0.085 vs 0.205,0.316 vs 0.499,all P < 0.05).Conclusions The high expression of cathepsin B,cathepsin L and uPA could be detected in NHL,and their expression has a close relationship with the biological behavior characters of NHL.The dynamic examination of cathepsin B,cathepsin L and uPA may be used as clinical auxiliary diagnosis and prognosis of NHL.
4.Study on relationship between efflux pumps and Candida albicans resistance mechanism

Objective To investigate whether efflux mechanism is involved in azole resistant Candida albicans strains isolated in China. Methods We compared rhodamine 6G uptake and glucose induced efflux between azole sensitive and azole resistant strains to elucidate resistance to azole due to efflux pump, then measured mRNA level of efflux gene cdr1 and cdr2 by reverse transcriptase polymerase chain reaction (RT PCR). Results Rhodamine 6G moved from extra into the intracellular compartment in both azole resistant and azole sensitive strains quickly when in glucose free incubation condition. However, efflux of rhodanmine 6G was enhanced significantly in azole resistant strains and decreased in azole sensitive strains while adding glucose in the media. The mRNA level of efflux gene cdr1 and cdr2 was higher in clinical azole resistant strains and fluconazole induced resistant strains than strains reverted from resistance to sensitivity. However, all such strains had higher efflux gene expression than azole sensitive strains. Conclusions Efflux mechanism is associated with the resistance to azole in Candida albicans strains isolated in China. Fluconazole can induce efflux pumps expression leading to resistance to azole but the resistance to azoles can be reversed after withdrawing fluconazole.
5.Research progress on the relationship between microRNA and acute lymphoblastic leukemia

MicroRNA is a type of endogenous noncoding small molecular RNA with a length of 19-25 nucleotides. MicroRNAs are involved in almost all key biological processes, such as cell proliferation, differentiation, apoptosis, and hematopoiesis, via repres-sion of target gene expression. Acute lymphoblastic leukemia (ALL) is the most common type of pediatric cancer. It is a heterogeneous disease with different clinical signs, morphology, immunophenotype, and cytogenetics. Currently, several microRNAs have implicated aberrant expression in ALL and play an important role in cancer initiation and development. MicroRNA expression pattern is associated with clinical feature, prognosis, and treatment. Therefore, microRNAs could act as a novel marker to help scientists learn about the pathogenesis of ALL and to provide more effective treatment options.
6.Effect of leucine-rich α2-glycoprotein-1 on ischemic stroke by regulating transforming growth factor β signaling pathway

As one of the leucine-rich repeat protein family members, leucine-rich α-2 glycoprotein 1 (LRG1) affects many diseases by transforming growth factor (TGF)-β signaling pathway, and is closely associated with angiogenesis, endothelial cell apoptosis and autophagy, inflammatory reaction and blood-brain barrier damage after cerebral ischemia. It is expected to become a new marker and therapeutic target of ischemic stroke. However, at present, there are few studies on investigating the relationship between LRG1 and ischemic stroke, and the understanding of its molecular mechanism is not yet complete, resulting in controversy about the role of LRG1 in ischemic stroke. Therefore, this article reviews the research progress of LRG1-TGF-β signaling pathway and ischemic stroke, hoping to provide new ideas for the early diagnosis, prevention and treatment of ischemic stroke.
7.Protecting Effect of Porphyra Yezoensis Polysaccharide on Cerebrum Tissue of Diabetic Rats

Objective To observe the effect of porphyra yezoensis polysaccharide (PYP) on cerebrum tissue of diabetic rats and apoptosis. Methods 60 Wistar rats were randomly divided into normal control group, diabetic model group, and treated groups respectively with PYP (0.25 g/kg × d), PYP (0.5 g/kg × d), and PYP (1 g/kg × d). The cerebrum pathologic changes were observed by HE staining method, and the number of apoptotic cells was observed by electrophoresis and the mRNA expression of p53 and bcl-2 was confirmed by RT-PCR. Results PYP improved the cerebrum pathologic changes of diabetic rats. With comparison to the diabetic model group, the cerebrum pathologic change in the treated groups also improved. And the number of apoptotic cells, the expression of p53 in cerebrum tissue decreased and expressions of bcl-2 rose. Conclusion PYP can decrease the expression of p53 in cerebrum tissue and rise expressions of bcl-2 so as to protect brain tissues from cerebral ischemia.

9.Distribution and antimicrobial resistance of pathogens causing urinary tract infection in patients with prostatic hyperplasia

Objective To explore the distribution and antimicrobial resistance of pathogens causing urinary tract infection(UTI)in patients with prostatic hyperplasia,and provide reference for clinical diagnosis and rational anti-microbial therapy.Methods Urine culture results of 171 patients with prostatic hyperplasia complicated with UTI from February 8,2012 to October 12,2013 were analyzed retrospectively.Results A total of 192 bacterial isolates were isolated from 171 patients,gram-negative bacteria accounted for 79.17%,37.50% of which were Escherichia coli (E.coli )strains,and 18.75% were Klebsiella pneumoniae (K .pneumoniae );gram-positive bacteria and Candida accounted for 18.23% and 2.60% respectively.Of E.coli and K .pneumoniae ,the proportion of extend-ed-spectrumβ-lactamase (ESBLs)strains were relatively higher,which was 72.77% and 63.89% respectively,re-sistant rates of ESBLs-producing E.coli to piperacillin.cefuroixme,ceftazidime,ceftriaxone,aztreonam,and levo-floxacin were all significantly higher than non-ESBLs strains,resistant rates of ESBLs-producing K .pneumoniae to piperacillin,cefoxitin,aztreonam,levofloxacin,and compound sulfamethoxazole were all significantly higher than non-ESBLs strains.Resistant rates of Pseudomonas aeruginosa to imipenem and meropenem were both 50.00%, susceptibility rates of Enterococcus to penicillins and aminoglycosides were both low,linezolid-and vancomycin-re-sistant strain was not found.Conclusion Organisms isolated from patients with UTI associated with prostatic hy-perplasia have high antimicrobial resistance,clinical therapy should be based on characteristics of bacterial distribu-tion and antimicrobial resistance.
10.Overtraining induces renal tubular cells apoptosis through activating caspase-related signal pathway by impairing the balance of Bax and Bcl-2 in exhaustive swimming rats

Objective To observe the expression changes of renal tissue Bax,Bcl-2 and caspase-3,to wxamine the correlation between the ratio of Bax to Bcl-2,caspase-3 and renal tubular cells apoptosis,and to investigate the role of caspase-related signal pathway.Methods Forty-eight male Wistar rats were randomly divided into three groups: control (CN,n=8),exhaustive swimming (ES,n=24) and inula britannica (IB,n=16) group.The rats of CN were quiet without swimming.The rats of ES swam to exhaustive state and were sacrificed at immediately(ESI),6 hour (ES 6 h) and 24 hour (ES 24 h) after exhaustive swimming respectively.The rats of IB took orally inala britannica at the dose of 25 ml/kg body weight at 24 h before swimming and then swam to exhaustive state.The rats of IB group were sacrificed at 6 hour (IB 6 h) and 24 hour (IB 24 h)after exhaustive swimming.The animal model of overtraining-induced acute kidney injury was developed by exhaustive swimming.The renal cell apoptosis was measured by the method of TUNEL.The expressions of Bax,Bcl-2,caspase-3 in renal tissue were observed by immunohistochemistry.The expression of caspase-3 protein was examined by Western blotting.The correlation between the ratio of Bax to Bcl-2 and caspase-3 was analysed by Pearson method,and the correlation between the ratio of Bax to Bcl-2,caspase-3 and renal tubular cell apoptosis was analysed by Spearman method.Results The number of renal tubular apeptotic cells was increased progressively in ESI to ES 24 h rats by TUNEL (P<0.05).Immunohistochemistry staining showed that the ratio of Bax to Bcl-2 and caspase-3 in renal tubular cells were increased progressively at 0 h,6 h and 24 h after exhaustive swimming compared with control group (P<0.05).The change of renal tissue caspase-3 was also revealved by Western blotting analysis.The ratio of Bax to Bcl-2 and caspase-3 in renal tubular cell was correlated positively (r=0.865,P<0.05),The ratio of Bax to Bcl-2,and caspase-3 was also correlated positively to renal tubular cell apoptosis (r=0.674,r=0.837,P<0.05) in ES rats.Pretreatment with inula britannica inhibited the up-regulation of the ratio of Bax to Bcl-2,caspase-3 and cell apoptosis in renal tubular cell induced by exhaustive swimming.Conclusion Overtraining can induce renal tubular cells apoptosis through activating caspase-related signal pathway by impairing the balance of Bax and Bcl-2,which may be one of the important molecular mechanisms of overtraining-induceed renal tubular cells apoptosis.
