1.Drug Use Analysis of the Chinese Traditional Patent Medicine in Our Hospital during the Period of 1999~2003

OBJECTIVE:To provide references and information about the research and development,manufacture and use for the related departments.METHODS:The sales amount,sequence of sales and etc.of the Chinese Traditional Medicine in our hospital from1999to2003were analyzed statistically with the help of database and electronic table software.RESULTS:Percentage of sales amount of the Chinese Traditional Medicine has been lowered from43.81%to33.70%.The top5drugs on the consumption list were the blood-regulating agents,tonifying agentss,drugs for bone fractures and wounds,febrifugal ag ents and gynaecological drugs respectively;the blood-regulating agents had a big share on the top10consumption volume list.CONCLUSION:There is a decrease in the consumption of the Chinese Traditional Medicine.
2.Optimal Controlover Drug Stock in Hospital

OBJECTIVE:To make the economic analysis on drug stock in hospita l.METHODS:The database technique and Excel software were used to analyze the sale and stock of the drugs in our hospital from1999to2003,the business turnover rate was evaluated to measure the efficiency of drug stock.RESULTS:The turnover rates increased continuously in5years.CONCLUSION:The turnover rate should be increased in a practical way to keep the optimal control over drug stock in hos?pital.
3.Optimization of the Extraction Technology for Gengniankang Granules with the Multiple Guidelines Grading Method

Objective To optimize the extraction technology for Gengniankang Granules.Methods An orthogonal test was adopted in the study.The contents of Paeoniflorin and Astragaloside Ⅳ were used as indices of appraisal.Data processing was carried out with the multiple guidelines grading method for optimizing the extraction condition.Results The optimized extraction technology for Gengniankang Granules was shown to be:eighteen fold volume of 70% alcohol should be added to 1 volume of the granules for 3 successive reflux extractions with 1.5 h of each.Conclusion The optimized extraction technology was rational and the extraction rate of the active components was high.
4.Optimization of Alcohol Extraction Process of Yiqigutuo Granules by Orthogonal Test

Objective:To optimize the alcohol extraction process of Yiqigutuo granules. Methods: Using the L9 (34 ) orthogonal design method, the active ingredients astragaloside and ferulic acid in the formula were quantitatively analyzed by HPLC. The concen-tration and amount of ethanol, extraction times and extraction duration and the other parameters affecting the extraction volume were ex-amined. Results:Analysis of variance showed that the ethanol concentration and extraction times had significant influence on the ex-tract content and the extraction of ferulic acid. The best alcohol extraction process was as follows:70% ethanol of 10-fold volume, ex-tracting 3 times with 1. 5h for each time. Conclusion:The process is stable, and suitable for the industrial production.

Objective: To investigate the mechanism of tea polyphenol in inhibiting microsatellite instability (MSI) of colorectal cancer. Methods: Using LoVo cells and SW480 cells treated with aqueous solution of tea polyphenol, cell proliferation was detected by methyl thiazolyl tetrazolium (MTT) method, changes in microsatellite sequences were detected by genescan method and changes in gene expression of LoVo cells were detected by illumina expression arrays and quantitative real-time polymerase chain reaction (PCR). Results: The proliferation inhibition rates of LoVo and SW480 cells treated with tea polyphenol increased with the increasing of drug concentration and showed an increasing tendency with time. The proliferation inhibition rate of LoVo cells with tea polyphenol was higher than that of SW480 cells, and there was a significant difference in the proliferation inhibition rates at 24 h, 72 h and one week. The microsatellite sequence of LoVo cells treated with tea polyphenol remained stable. The gene expression arrays and quantitative real-time PCR suggested that tea polyphenol inhibited the gene expressions of MT2A, MAFA, HES1 and JAG1 nearly two-fold over controls. It was also found that tea polyphenol inhibited the BAX and p38 genes with a more than two-fold difference but did not significantly inhibit the nuclear factor-κB pathway. Conclusion: Tea polyphenol significantly inhibited the proliferation of MSI colorectal cancer cells and stably maintained the microsatellite state in MSI colorectal cancer. Tea polyphenol inhibited the gene expressions of HES1, JAG1, MT2A and MAFA, up-regulated the gene expression of BAX and down-regulated that of P38. Further research is required to investigate how these pathways are interrelated.
6.Study on the Quality Standard of Huoxuesan

OBJECTIVE:To establish the quality standard of Huoxuesan. METHODS:Microscopic identification method was adopted for the qualitative identification of Rhizoma pinelliae,Eupolyphaga seu,Phellodendron amurense and Fructus kochiae in the preparation. HPLC was adopted for the contents determination of phellodendrine,polydatin,jatrorrhizine,berberine and emo-din:the column was ZORBAX Eclipse Plus C18 with methanol-0.1% phosphoric acid(gradient elution)at a flow rate of 0.4 ml/min,the detection wavelength was 240 nm,column temperature was 30 ℃,and the injection volume was 5 μl. RESULTS:Micro-scopic identification map of R. pinelliae,E. seu,P. amurense and F. kochiae was clear. The linear range was 0.159-5.073 μg(r=0.997 4)for phellodendrine、0.149-4.761 μg(r=0.999 9)for polydatin、0.239-7.649 μg(r=0.995 5)for jatrorrhizine、0.165-5.268 μg (r=0.997 2)for berberine、0.012-0.382 μg(r=0.999 9)for emodin;RSDs of precision,stability and reproducibility tests were low-er than 3.0%;recoveries were 98.9%-104.1%(RSD=1.9%,n=6),96.1%-101.7%(RSD=2.5%,n=6),99.6%-105.1%(RSD=2.6%,n=6),90.3%-98.2%(RSD=2.9%,n=6)and 92.9%-96.4%(RSD=2.0%,n=6)respectively. CONCLUSIONS:The es-tablished standard can be used for the quality control of Huoxuesan.
7.Optimization of Extraction Process of Volatile Oil from Wenyang Purgation Granules by Orthogonal Design

Objective To optimize the extraction conditions of volatile oil from Angelica Sinensis and Cassia Bark in Wenyang purgation granules.Methods The volatile oil was extracted from Angelica Sinensis and Cassia Bark by the method of steam distillation light oil device with diethyl ether extraction. The yield of the volatile oil was chosen as the evaluation index. The time of dip in water, the ratio of water to herbal medicine material and the time of distillation were used as the main factors. The optimum extraction conditions were investigated by the L9(34) orthogonal design. Results The optimal conditions for extraction process of volatile oil from Angelica Sinensis and Cassia Bark in Wenyang purgation granules were as follows:the time of dip in water was 3 h;the ratio of water to herbal medicine material was 10∶1;the time of distillation was 6 h. Conclusion The optimized conditions of extraction process are stable and feasible.
8.Simultaneous Determination of Rutin, Hesperidin,Arctiin and Liquiritin in Sangxing Oral Liquid by RP-HPLC

OBJECTIVE:To establish a method for the simultaneous determination of rutin ,hesperidin ,arctiin ,liquiritin and glycyrrhizinate in Sangxing oral liquid. METHODS:RP-HPLC was performed on the column of Kromasil C18 with mobile phase of acetonitrile-0.1% phosphoric acid solution(gradient elution) at a flow rate of 1.0 mL/min,the detection wavelength was 276, 348 nm ,column temperature was 30 ℃,and the injection volume was 5 μL. RESULTS:The linear range was 1.068-10.68 μg/mL (r=0.9993)for rutin,9.320-93.20 μg/mL(r=0.9992)for hesperidin,46.91-469.1 μg/mL(r=0.9994)for arctiin,0.5110-5.110μg/mL(r=0.9992) for liquiritin;RSDs of precision , stability and reproducibility tests were less than 3%;recoveries were 95.10% -101.90%(RSD=2.1%,n=9),96.75%-101.80%(RSD=1.4%,n=9),95.69%-100.00%(RSD=1.5%,n=9), 98.22%-104.60%(RSD=2.1%,n=9),respectively. CONCLUSIONS:The method is accurate,sensitive and reproducible,and can be used for the simultaneous determination of rutin,hesperidin,arctiin and liquiritin in Sangxing oral liquid.
