1.Research delta opioid receptor and its association with hepatocellular carcinoma

Opioid receptors have drawn highly attention to scientists since they were founded in the 1990s.As a member of G protein-coupled receptor family,opioid peptides are the endogenous ligands.It is well known to us that the basic structure consists of extracellular amino residues,seven transmembrane region and the hydroxyl residues of the cell.In recent years,the progress of science laid the foundation for further studies on the function of delta receptor.Aberrant expression of delta opioid receptor in a variety of tumors,and it plays an important role of the occurrence and development of tumors.This review describes the recent research advances on delta opioid receptor and its association with hepatocellular carcinoma.

Objective To study the effects of granisetron hydrochloride on vasovagal syncope(VVS) in rabbits.Methods Twenty-four healthy New Zealand rabbits were divided stochastically into control group and intervention group,12 in each group. The control group was injected intravenously with normal saline. The intervention group was injected intravenously with granisetron hydrchloiride.Rabbit VVS models were established,each was taken at 4 points in time in the bloodletting process:T1,T2,T3,T4,to compare the bloodletting time,the concentration of 5-hydroxytryptamine(5-HT) in T2,T3,T4 and the total blood volume between the groups,and monitor the heart rate, blood pressure during the entire process.Results 1.The time of intervention group in T2,T3,T4 was longer than the time of control group obviously(P
3.Clinical effect of meloxicam combined with alendronate in the treatment of knee osteoarthritis with osteoporosis

Objective To explore the clinical effect of meloxicam and alendronate in the treatment of knee osteoarthritis with osteoporosis. Methods A total of 64 cases of Kellgren-Lawrence grade 2-3 of knee osteoarthritis with osteoporosis patients were randomly divided into treat-ment group and control group.Treatment group was treated with meloxi-cam 15 mg, three times a day for three months, and alemdronate 70 mg, once three day for one month.Control group was treated with meloxicam only.The clinical effect, pain, stiff degree, daily activities difficulty and quality of life of the two groups were compared.Results After treat-ment, total effective rate of treatment group was 96.9%, significantly higher than 78.1% in control group ( P <0.05 ) .The pain degree in treatment group after treatment significantly decreased than that before treatment ( P<0.05).The degree of stiffness, difficulty of daily activi-ties and the changes in quality of life were significantly better than those of control group ( P <0.05 ) .Conclusion Meloxicam combined with alendronate in the treatment of knee osteoarthritis with osteoporosis can not only significantly reduce the pain, improve the stiff degree, but also improve the quality of life in early treatment of knee osteoarthritis.
4.Screening and verification of key Hub genes in esophageal squamous cell carcinoma based on bioinformatics analysis

Objective: To screen the Hub genes associated with the occurrence and development of esophageal squamous cell carcinoma (ESCC) and to analyze their biological functions by using various bioinformatics analysis tools. Methods: ESCC chip profile GSE100942 from GEO database was used as study subject; GEO2R tool was used to analyze the data and to screen the differentially expressed genes (DEGs), and the bioinformatics tools (DAVID, String, Cytoscape) were further used to construct protein-protein interaction (PPI) network and identify the key Hub genes. GO and KEGG were used for the biological function enrichment analysis. In the meanwhile, MiRDB was applied to identify the miRNAs that might regulate Hub genes and to construct Hub gene–miRNA network. Importantly, the expression of DEGs and the patient survival were verified by the GEPIA analysis tool. Results: By analyzing GSE100942 database, a total of 1229 DEGs with difference of 2 times and 223 DEGs with difference of 4 times were screened out. In addition, 20 Hub genes, which were all up-regulated in ESCC tissues, were also identified. The functional enrichment analysis showed that these DEGs were mainly enriched in cancer related pathways and involved in cell division and mitotic nuclear division. Among those 20 Hub genes, DLGAP5, BUB1B, TPX2, TTK, CDC20, CCNB2, AURKA and DEPDC1 were identified as 8 key Hub genes that related with ESCC, and involved in many important biological processes, such as cell proliferation, cell cycle and signal pathway. Five Hub genes, CEP55, ECT2, NEK2, DEPDC1 and NUSAP1, were identified to be highly regulated by the miRNA regulatory network. Conclusion: Microarray combined with bioinformatics can effectively analyze the DEGs associated with the occurrence and development of ESCC. The identification of the 20 Hub genes and the 8 key Hub genes can provide theoretical guidance for further research on the molecular mechanism and molecular marker screening of ESCC.

5.Effects of culture medium of amniotic cells on NO and NOS in retina tissues in vitro

Objective To observe the effects of culture medium of a mn iotic cells on NO and NOS in retinal tissues of rabbits in vitro in order to pro vide a protective method for antioxidation in retina transplantation. M ethods Thirty adult healthy rabbits (30 right eyes) were divided into 3 groups. Group Ⅰ: fresh retinal tissue; group Ⅱ: routine culture medium; group Ⅲ: culture medium of amniotic cells. The retinal tissues in group Ⅱ and Ⅲ we re cultured in the corresponding culture medium for 1 week. The content of NO an d NOS in retinal tissues in the 3 groups were determined. Results Compared with group I, the content of NO and NOS of group Ⅱ increased obvio usly (t=3.821, 3.854; P0.05) . Conclusion Culture medium of amniotic cells may remove free r adicals and enhance the ability of antioxidation.
6.Clinical efficacy and safety of atorvastatin combined with alendronate sodium in the treatment of senile osteoporosis

Objective To evaluate the clinical efficacy and safety of atorvastatin combined with alendronate sodium in the treatment of senile osteoporosis.Methods Sixty-four cases of senile osteoporosis patients were randomly divided into control group ( n =32 ) and experimental group(n=32).The control group was treated with oral alendronate sodi-um 70 mg, once a week.The experimental group was treated with alen-dronate sodium 70 mg, once a week and atorvastatin 20 mg, once a day.The course of treatment for two groups was half a year.The clinical effi-cacy, bone density, visual analogue scale (VAS) and adverse drug reac-tions were compared between the two groups.Results After treatment, total effective rate in experimental group was significantly higher than that of control group (96.88%vs 78.13%, P<0.05).After the treatment, the bone mineral density of the forearm , femoral neck , lumbar spine and femoral neck in the experimental group were significantly higher than those in the control group ( P <0.05 ).After the treatment , the VAS score of the experimental group was significantly higher than that of the control group ( P<0.05 ).The adverse drug reactions rate in the experi-mental group was 46.88%, which was significantly lower than that in the control group ( P <0.05 ).Conclusion Atorvastatin combined with alendronate sodium has a definitive clinical efficacy for the treatment of senile osteoporosis , and it can effectively improve the patients′bone density.At the same time it can relieve the pain of patients and reduce the incidence of adverse drug reactions.
7.C-arm-based flat-panel detector cone-beam computed tomography venography in the diagnosis of iliac vein compression syndrome

Background::C-arm-based flat-panel detector cone-beam computed tomography (CBCT) venography has never been used in the management of iliac vein compression syndrome (IVCS). This study aimed to determine the technical feasibility and safety of CBCT venography in the diagnosis of IVCS compared with conventional venography (CV).Methods::Twenty patients with clinical manifestations of lower extremity venous insufficiency were prospectively enrolled between May 2018 and December 2018. Each patient underwent both CV and CBCT venography. The feasibility and safety of CBCT venography were assessed by technical success rate and complication rate. The relationships between the clinical indexes and the results of CBCT venography and CV were analyzed with correlation analysis. The consistency of the diagnosis of IVCS using each modality was analyzed by the kappa test.Results::The technical success rate was 100% for CBCT venography and for CV, without any complications. Compared with CV, CBCT venography was able to show more details of adjacent tissues which might be helpful for making etiological diagnosis. The stenosis rate under CBCT venography had excellent consistency with that under CV (kappa= 0.78, Chi-square test). The stenosis rate under CBCT venography was positively correlated with the presence of collateral veins (odds ratio 1.12, 95% confidence interval: [1.00, 1.26], P= 0.049), while the stenosis rate under CV was not. Unexpectedly, only one patient had a venous pressure gradient of more than 2 mmHg (1 mmHg = 0.133 kPa). Conclusions::For the diagnosis of IVCS, C-arm-based CBCT venography was technically feasible, with good safety. The presence of collateral veins on CBCT was clinically significant. A C-arm fluoroscopy-based technique that combines CV and CBCT might be a promising protocol for the management of IVCS during a single session.
8.Cloning of human bone morphogenetic protein-2 gene and the construction of its eukaryotic expression vector.

OBJECTIVETo clone human bone morphogenetic protein-2 (hBMP2) gene and construct its eukaryotic expression vector pcDNA3.1 -hBMP2.
METHODSHuman BMP2 gene was amplified by RT-PCR method from human osteosarcoma cells and constructed into eukaryotic expression vector pcDNA3.1-hBMP2. The gene in the vector pcDNA3.1-hBMP2 was identified by PCR amplification, enzyme digestion and DNA sequencing.
RESULTSThe cloned DNA was confirmed to be hBMP-2 gene.
CONCLUSIONIn this study, hBMP2 gene is successfully cloned and its eukaryotic expression vector pcDNA3.1-hBMP2 is constructed, which provides the foundation of using BMP2 gene therapy to accelerate new bone formation in distraction osteogenesis.

10.Predictors of testicular injury secondary to incarcerated inguinal Hernia in children

Objective:To find out predictors of the testicular ischemia caused by incarcerated inguinal hernia and evaluate the ischemic injury of the testis more accurately, which can indicate testicle exploration in time or prevent unnecessary testicle exploration.Methods:Pediatric patients (median: 9 months) undergoing operation of unilateral incarcerated inguinal hernia and ipsilateral testicular exploration from 1 Jul. 2013 to 30 Jun. 2019 were retrospectively investigated. Age at surgery, incarcerate duration, degree of intestinal and testicular injury, times of manual reduction and preoperative ultrasound data were collected. Statistical analysis was performed by SAS 9.4 (Copyright ? 2016 SAS Institute Inc.Cary, NC, USA) .Results:460 patients (median: 9 months) , of which 57 (12.39%) (median: 1.4 months, interquartile range 0.8-10.7 months) had severe testicular injury, and their average incarceration time was (23.9±9.3) h. Univariate logistic regression revealed that increased times of manual reduction, ultrasound scores, incarcerate duration and degree of intestinal injury were positively correlated with the degree of testicular ischemia, while age at surgery was negatively correlated with the degree of testicular ischemia ( P<0.05) . A model for calculating the probability of severe testicular ischemia injury was established: P= through multivariate analysis with backward stepwise logistic regression and 10-fold cross-validation was used for preliminary verification of the model. Conclusion:This study provides a relative reliable model to predict the risk of irreversible testicular ischemia due to incarcerated inguinal hernia using readily available clinical characteristics in young pediatrics with testicular ischemia.
