1.Therapeutic value of immune cell therapy based on T memory stem cells and central memory T cells for hematological malignancies

In recent years,cellular immune therapy represented by antigen-chimeric receptor T cells (CAR-T) has made a great breakthrough in the treatment of hematological malignancies.T memory stem cells (TSCM) and central memory T cells (TCM) can be used as most powerful carrier for T cell immunotherapy,however,how to regulate their differentiation in vitro and in vivo as well as how to construct a strong treatment system while without potential side effect become quite interesting.This article summarized the studies from the 58th America Society of Hematology Annual Meeting regarding to values and associated research progress about TsCM and TCM in hematological malignancies.
2.Cost-effectiveness Analysis of Levofloxacin and Gatifoxacin in the Treatment of Lower Respiratory Tract Infections

Objective:To evaluate the cost-effectiveness of levofloxacin and gatifoxacin in the treatment of lower respiratory tract infections.Method:The clinical data of levofloxacin and gatifoxacin in the treatment of lower respiratory tract infections were collected with the evidence-based medicine method and were evaluated with pharmacoeconomic cost-effectiveness analysis.Result:The cure rates of two drugs were 64.29%(57.70%,70.88%),and 81.43%(70.05%, 92.81%)and the cost-effectiveness ratios were 1.90 and 5.79 respectively.The?C/?E was 20.37.Conclusion:The cost-effectiveness analysis indicated that the cost-effectivenss ratio of levofloxacin is superior to that of gatifoxacin though the cure efficacy of gatifoxacin for lower respiratory tract infections is better.
3.Systematic Review of Gemcitabine in the Treatment of Non-small-cell Lung Cancer

OBJECTIVE:To evaluate the clinical efficacy,safety and status of gemcitabine for non-small-cell lung cancer (NSCLC) systematically.METHODS:Included studies retrieved from Medline,EMbase,Cancerdit,CBM and CNKI were evaluated by two reviewers alone and cross-checked.The Meta-analyses of studies were conducted using RevMan 4.2.2 software.RESULTS:12 studies containing 1,715 patients with NSCLC were included.12 studies were all reported randomly(RCT).Hidden information and application of blind method in therapy were not reported.Results of meta-analysis indicated that:(1)After chemotherapy reaction rate and the level of median disease development in GP therapy (gemcitabine combined with cisplatin) were higher than in GC therapy (gemcitabine combined with carboplatin) with similar median survival time and strong reaction of digestive tract;(2)Reaction rate;the level of median disease development and median survival time in GP therapy than in EP therapy (etoposide combined with cisplatin) but with strong toxicity in bone marrow;(3)Reaction rate of GP therapy was same to NP therapy (vinorelbine combined with cisplatin) with strong toxicity in bone marrow;(4)Reaction rate,median survival time and one-year survival rate in GP therapy were similar to in gemcitabine alone therapy with intense reaction in digestive tract;(5)Reaction rate in VGP therapy (vinorelbine,gemcitabine combined with cisplatin) was higher than in VG therapy (vinorelbine combined with gemcitabine) with strong toxicity in bone marrow but they had same the level of median disease development and median survival time;(6)Reaction rate,median survival time in DG therapy (gemcitabine combined with taxotere) were higher than in taxotere alone therapy with advanced the quality of life and strong toxicity in bone marrow;(7)Reaction rate,median survival time,the level of median disease development and one-year survival rate in therapy of gemcitabine (50 mg?min-1?30 min,i.v) were similar to that of gemcitabine (10 mg?min-1?150 min,i.v) with intense toxicity in bone marrow.17 cases of related death were reported in all researches.CONCLUSION:Gemcitabine has exact clinical efficacy and causes strong toxicity in bone marrow and reaction in digestive tract.

6.Protective effect of tetrahydroxystilbene glucoside on hippocampal neurons damage induced by glutamate in rats

ObjectiveTo observe the protective effect of tetrahydroxystilbene glucoside (TSG) on rats' hippocampal neuronal damage induced by glutamate (Glu) in the culture.MethodsHippocampus was isolated from newborn SD rats and dispersedly cultured in the medium for 9 days. Neurons were incubated with TSG (5—100μmol/L) for 24h, the cells were washed twice with Lock's solution without Mg2+,then Glu 500 μmol/L was added. Thirty min later, the reaction was terminated by washing the monolayer cells twice with the Lock's solution and then cultures were kept at 37℃ for 24h. Cell viability was measured by MTT method and cell membrane damage was determined by LDH leakage; with Fluo-3/AM as an intracellular calcium indicator and added into the bathing medium, fluorescent intensity of intracellular free calcium were observed through laser scanning confocal microscopy (LSCM).ResultsAfter the treatment with 5—100μmol/L TSG for 24h, the decrease of cell viability and the increase of LDH leakage caused by Glu was obviously resisted dose dependently. TSG inhibited increase of Ca2+ in cytoplasm, compared with model group.ConclusionTSG can significantly promote the cell viability and reduce the cell membrane damage in Glu treating hippocampal neurons. The neuroprotective activities of TGS is mediated by inhibiting Ca2+ overload in cytoplasm.
7.Effects of rehabilitation training on the expression of Fos in the central nervous system of rats with cerebral infarction

Objective To study the effect of rehabilitation training on the expression of Fos in the central nervous system(CNS) of rats with cerebral infarction. Methods 60 SD rats were made as cerebral infarction models and divided into 2 groups at random 24 hours later: rehabilitation group was given balancing, grasping, rotating and walking and other trainings everyday, while immobiligation group was fixed in cages. Immunohistochemistry was used to detect the Fos expression 1 d, 7 d, 14 d, 21 d and 28 d after infarction respectively. Results Fos-positive immune reaction appeared in cortex,thalamus, hypothalamus, reticular form and posterior gray horn of the spinal cord of both groups .Rehabilitation group responded stronger than the immobilization group significantly (P<0.01).Conclusion Rehabilitation training can activate cortical neurons around the infarted focus and in its opposite side of the CNS, and thus promote the recovery of rats' motor functions.

Objective BALB/C mice were inoculated subcutaneously with hepatoma 22 (H22) cells, and the treatment effect of thalidomide and intratumor injection of ethanol extract of Scutellaria Barbata (SB) on tumor growth were evaluated. Methods In vitro, mouse H22 liver cells were cultivated, and H22 cells in the logarithmic growth phase were tested. SB was extracted in the ethanol recirculation. A total of 28 BALB/C male mice inoculated subcutaneously with H22 cells were randomly divided into 4 groups, 7 mice per group. Group A:intragastric administration with thalidomide. Group B:SB intratumor injection. Group C:combination of thalidomide and SB with the same administration as Group A and Group B. Group D (control group):1% Ethanol 0.2~0.5 mL intratumor injection. Tumor volumes of four groups were observed and the tumors were weighed. The microvessel densities (MVD) of all transplantation tumors were measured by immunohistochemical staining with anti-CD31 monoclonal antibody. A thin layer chromatograph-fluorescent method was used to assay content of scopoletin in Erycibe obtusi folia Benth.. The method showed that it not only can minimize analytical errors in thin layer scan and column contamination in HPLC,but also eliminated mutual interference of components in the preparation. It provides a good assay technique for quality control of hebal preparations.
