1.Ramipril/Atorvastatin for Diabetic Nephropathy:Clinical Observation
China Pharmacy 2001;0(11):-
OBJECTIVE: To observe the renal protective effects of ramipril and atorvastatin used in combination or alone for diabetic nephropathy. METHODS: 54 patients were randomly assigned to receive ramipril 5 mg q.d (Group A), atorvastatin 10 mg q.d (Group B) or ramipri 5mg qd + atorvastatin 10mg q.d (Group C) for 18wk. RESULTS: After 18-week treatment, levels of both urine protein and blood lipid decreased in all the three groups (P0.05). CONCLUSION: Either Ramipril or Atorvastatin alone had satisfactory effect on diabetic neph-ropathy, but a more remarkable protection effect on kidney was noted when they used in combination.

Objective To study the protective mechanism of lycopene by modulating lipid metabolic disturbance in rats. Method The hyperlipidemic rats were induced by high fat diet and were divided into groups given different doses of lycopene. The serum total cholesterd(TC), triglyceride(TG), low density lipoprotein cholesterol(LDL-C), high density lipoprotein cholesterol(HDL-C), the hepatic lipase(HL) and lipoprotein lipase(LPL)activity in liver were tested by biochemical method, and the expression of low density lipoprotein receptor(LDL-R)in liver was detected by Western blot analyses. Results Lycopene decreased serum TC, TG, LDL-C, and the activity of LPL and HL in three lycopene groups were higher than those in model group, and the high dose group was the highest. Conclusion Lycopene may increase the lipases’activity and the expression of LDL-R, and decrease the deposit of TG and LDL in liver to prevent lipid metabolic disturbance.

Aim To investigate the anti-EAC breast cancer effects of viable human umbilical vein endothe-lial cells ( HUVECs) vaccine. Methods Subconflu-ent HUVECs were mixed with OK432 to prepare HU-VECs-OK432 vaccine, and the anti-tumor efficacy of HUVECs-OK432 was investigated using a subcutaneous tumor model of EAC breast cancer. ELISA, splenic lymphocyte proliferation assay and cytotoxic T lympho-cytes ( CTL ) killing assay were adopted to detect the humoral and cellular immune responses after HUVECs-OK432 immunization. Results HUVECs-OK432 im-munization significantly inhibited the growth of EAC tumor in mice in the prophylactic procedures. High ti-ter of anti-HUVEC antibody was elicited by HUVECs-OK432 immunization. The proliferation activity of splenocytes from mice immunized with HUVECs-OK432 was significantly increased. T lymphocytes iso-lated from HUVECs-OK432-immunized mice were dose dependently killing HUVECs in vitro. Conclusion Strong humoral and cellular immune responses targeting HUVEC are elicited by HUVECs-OK432 immuniza-tion, which results in a significant inhibition on the growth of EAC tumor in mice.

Objective To evaluate the significance of serum matrix metalloproteinase-3 (MMP-3) and joint ultrasonography in assessing the activity of rheumatoid arthritis (RA) by comparing MMP-3 level and the ultrasonic 7 joints (US7) score in RA patients. Methods Serum MMP-3 level and US7 score were measured in 133 RA patients by immune turbidity and Doppler ultrasound.Synchronous 53 healthy subjects were recruited as controls.Clinical data were collected.Erythrocyte sedimentation rate (ESR),serum level of anti-cyclic citrullinated peptide (CCP) antibody, health assessment questionnaire (HAQ) and disease activity score 28 (DAS28) were measured. The level of disease activity is interpreted as remission(DAS28 <2.6),low(DAS 28≥2.6-<3.2),moderate(DAS 28≥3.2-<5.1),high(DAS28≥5.1).The discriminating validity of MMP-3 and US7 score in disease was evaluated using receiver operating characteristic (ROC) curve analysis with DAS28 as the reference standard. Results Compared with that in healthy controls [35.20(25.90,48.90) μg/L] and remission patients[33.40(22.60,678.40) μg/L],the MMP-3 level in moderate [105.1 (61.70,172.70) μg/L] and high [363.1 (161.50,475.90) μg/L]groups increased dramatically.US7 score in patients with high disease activity was significantly higher than that in other groups. The level of MMP-3 was significantly correlated with DAS28,HAQ,US7 score, yet did not have correlation with anti-CCP antibody.Serum level of MMP-3 was positively correlated with US7 score (r =0.566,P < 0.001).In evaluating the disease activity,US7 score combined with MMP-3 (AUC 0.863 2) was not superior to MMP-3 alone (AUC 0.854 3),but significantly better than single US7 score (AUC 0.7643,P < 0.05).Conclusions MMP-3 is an effective and simple index in evaluating RA disease activity. The combination of MMP-3 and US7 score does not further improve the efficacy to evaluate disease activity than MMP-3 alone in patients with RA.

Objective To explore whether the polymorphism of human frizzled 3 (rs2241802 ,rs2323019,rs352203) is associated with schizophrenia in Yunnan Han population. Methods PCR-RELP was used to detect the polymorphism of human frizzled 3 (rs2241802 ,rs2323019, rs352203 ) in 153 patients and 100 controls. All patients were evaluated with positive and negative symptom scale(PANSS). Following a linkage disequilibrium test in the 3 loci ,association analysis was carried out between the polymorphism, including haplotype, and schizophrenia in all samples. Results No significant difference of genotype and allele frequency of the 3 loci were observed in 2 groups. There was no difference in genotype and allele frequency of rs2241802 ,rs2323019,rs352203 in Han samples (P>0.05) . The polymorphisms of the 3 loci had been observed no difference in case-sibling control study of normal group. A-A-C haplotype and G-A-C haplotype were observed with a higher frequency in both samples (P<0.05). There was no difference in patients and controls in PANSS scores (P > 0.05 ). Conclusions There is no association between polymorphisms of human frizzled 3 ( rs2241802, rs2323019, rs352203 ) and schizophrenia in Han. A-A-C and G-A-C haplotype might possibly associate with schizophrenia in Yunnan Han population.
