2.A Novel Genetic Polymorphism and Its Genetic Effects of Porcine Myogenin Gene in Intron 1

Single-nucleotide polymorphisms of the MyoG gene were tested using PCR-SSCP from 73 Landrace pigs, 68 Large White pigs, 57 Yimeng pigs and 83 Laiwu pigs. The effects of the MyoG gene on the birth weight, the average daily gain, the meat tenderness, and the backfat thickness were also analyzed. On the basis of the DNA sequence (M14331) of the porcine MyoG gene, primers were designed to amplify MyoG gene. One polymorphism was found in the amplified region of intron 1, in which two alleles ( A, and B) and three genotypes (AA, AB, and BB ) were examined. A G→ C transition was detected at 2 943 locus by sequencing the homozygotes. The results show that: ( 1 ) The Large White breed differed significantly ( P <0.05) in genotype distribution from the Landrace, the Laiwu and the Yimeng breeds;and the Laiwu breed differed significantly( P < 0.05) in genotype distribution from the Landrace and the Yimeng breeds; whereas no significant differences( P > 0.05) were found in genotype distribution between the Landrace and the Yimeng breeds. (2) On the basis of the fixed effect model, significant differences ( P < 0.05) were found in the birth weight and the tenderness among the different MyoG genotypes, whereas no significant differences ( P > 0.05)existed in the average daily gain and the backfat thickness. (3) Using least square analysis, it was seen that significant differences ( P < 0.05) exist in the meat tenderness between the individuals of the BB genotypes and those of the AA genotypes; and significant differences ( P < 0.05 ) were found in the backfat thickness compared the pigs of the AA genotypes with the pigs of the AB, BB genotypes. These results suggest that the MyoG genotype has significant effects on the meat quality, the growth rate, and the backfat thickness,therefore MyoG gene can be used in marker-assisted selection to improve meat quality and growth rate, and to accelerate the breeding progress.
4.Investigation on the effect of topical corticosteroids as adjunctive plan in the therapy of bacterial corneal ulcers

Objective To investigate the clinical efficacy and safety of topical corticosteroids as adjunctive plan in the therapy of bacterial corneal ulcers (BCU).Methods 62 eligible patients with confirmed BCU were screened and divided into 2 groups randomly.Both groups were treated with topical tobramycin for 3 weeks,the observation group received topical tobramycin for at least 48 hours and then treated with 0.02% fluorometholone for 3 weeks,and decrement setp by step.Best spectacle corrected visual acuity(BSCVA),fluctuation of intraocular pressure (IOP) before and after therapy,time to corneal reepithelialization and clarity,severe adverse event were observed.Results After 3 weeks,BSCVA of both groups increased compared to their baselines,however,no significant difference between the two groups.Patients whose IOP exceed 21mm Hg in the control group was statistically more than that in the observation group(x2 =7.272,P < 0.05).Follow up for 3 months,increasing of BSCVA in the observationgroup was significantly higher than the control group(t =2.388,P < 0.05) ;and lOP of both two groups almost recuperated to baselines.Although the time to corneal reepithelialization of the control group (11.3 ± 4.5) d was shorter than observation group [(14.7 ± 5.2) d] (t =2.707,P < 0.05),the recovery of corneal clarity in observation group was superior to control group (x2 =8.207,P < 0.05).In addition,no severe adverse events were observed during this period.Conclusion Although prolong the healing time of corneal epithelium,corticosteroids as an adjunctive plan in the therapy of BCU may reduce immune-mediated damage,decrease scarring,control IOP fluctuation and improve BSCVA,but with no safety concerns.

The studies on Internet public opinions in domestic library and information field from 2004 to 2013 were analyzed by subject analysis,which showed that studies on the hot spots of Internet public opinions using bibliometrics and knowledge map were rather mature while studies on how library and information science provides the theoretical support for Internet public opinions in its service were immature. The problems in studies on Internet public opinions in library and information field should be further studied, including their objectives, theoretical support, techniques and application.

Correspondence between man and nature theory is the core of TCM chronomedicine, and also plays an important role in ancient Chinese philosophy. The theory believes that distinguishing the rhythm is the foundation of treatment; complying with nature is the core of treatment; taking advantage of wane and wax is the method of treatment. This article combined the physiological and pathological characteristics of children to review the prevention and treatment of pediatric diseases based on chronomedicine, with a purpose to provide inspirations for clinical treatment.
8.Clinical observation onpreventive treatment of primary dysmenorrhea with acupuncture of regulating Qi and relieving pain

Objective To study the effect of preventive treatment of primary dysmenorrhea (belonging to syndrome of stagnation of Qi and blood) by using “acupunctureof regulating Qi and relieving pain”. Methods 60 patientsfrom January 2013 to November 2013(in acupuncture and moxibustion department of Beijing Chinese Medicine Hospital affiliated to Capital Medical University), with Qi and blood stasis type of primary dysmenorrhea were randomly divided into a treatment group (formula of regulating Qi and relieving pain)and a control group(routine acupuncture group), 30 cases in each group. The treatment group was treated with acupuncturing onlieque,fenglong,ligou as the main points while the control group withhegu, diji, sanyinjiao as the main points. All patients received 3 times of acupuncture treatment at the beginning of the week before menstruation and were treated three menstrual cycles altogether. VAS scores, length of pain and CMSSⅡscores during baseline period and three menstrual cycles were observed in order to determine the clinical effects. Results VAS score, length of pain and CMSSⅡscores were significantly reduced than before(P<0.01)[the treatment group in cycle 1 was(5.05±0.98), (14.03±8.37)h, (12.77±3.64), in cycle 2 was (4.18±0.87), (12.53±7.13)h, (12.07±3.69), in cyce 3 was(3.43±0.82), (11.50±6.88)h, (11.33±3.64);the control group in cycle was 1(5.00± 1.05), (14.97±9.19)h、(14.40±3.5), in cycle 2 was(4.25±0.93), (13.83±9.12)h, (13.70±3.43), in cycle 3 was(3.73±0.91), (12.63±9.02)h, (13.03±3.41)];Compared with the control group, VAS scores intreatment groupdecreased more significantly (P<0.05), while reduced level of pain time and CMSSⅡscores showed no significant difference between the two groups (P>0.05). Conclusion Acupuncture of regulating Qi and relieving painmay effectively relieve pain, shortenlength of pain, improve symptoms of dysmenorrhea severity. It has a significant preventive treatment for Qi and blood stasis type of primary dysmenorrhea.
9.Effect of exectinib on the expression of EGFR genes 19 and 21 in patients with advanced lung cancer

Objective To investigate the effect of ectectin on the expression of EGFR gene 19 and 21 in patients with advanced lung cancer.Aim to provide scientific reference.Methods68 patients with advanced lung cancer were treated with 3 months of ectectin.Serum free DNA was extracted from patients before and after treatment.RT-PCR was performed to amplify EGFR exon 19 and exon 21, EGFR gene mutations were detected by gene sequencing, and the mutation status of exon 19 and exon 21 of EGFR gene before and after treatment were evaluated.ResultsIn 68 patients with lung cancer before and after treatment with ectatinib, Histopathological types were correlated with the efficacy of ectectin hydrochloride.DCR of adenocarcinoma was significantly higher than that of squamous cell carcinoma.18 cases of EGFR mutations, there is no CR and PD cases, PR 11 cases, SD 7 cases, ORR 61.1%, DCR 100%.In the remaining 50 patients with unknown EGFR status, PR 24, SD 4, PD 15, ORR 48.0%, DCR 56.0%.There was no significant difference in the mutation rate of exon 19 and 21 between before and after chemotherapy.The mutation status of serum EGFR was correlated with the pathological type of patients (P<0.02).The EGFR gene mutation rate was 35.4% (17/48) before chemotherapy and 50% after chemotherapy.The rate of EGFR gene mutation was 55% before and after chemotherapy.ConclusionEctectin is effective in the treatment of advanced lung cancer, and it is effective in lung adenocarcinoma and may lead to the change of serum EGFR gene mutation in lung cancer patients,for the first-line application of patients treated with ectectin hydrochloride should always observe the changes in serum EGFR gene, to avoid blindness in treatment of patients.
10.Optimization of the Forming Technology of Duanxueliu Dropping Pills by Orthogonal Experiment

OBJECTIVE:To optimize the forming technology of Duanxueliu dropping pills.METHODS:The forming technology of Duanxueliu dropping pills was optimized by orthogonal experiment with the type of matrix,the ratio of extractum to base material and the dropping temperature as factors and with RSD(weight variance of pills),the dissolution time,molding ratio of dripping pills as indexes,meanwhile,comprehensive scoring was carried out.The dripping conditions were optimized with dropping distance,dropping speed,the internal and external diameters of the burette as factors and with variation coefficient of pill weight as index.RESULTS:The optimized parameters for the forming technology were as follows:PEG6000 was used as base material with the ratio of extractum to base material at 1∶3 and the dropping temperature at 75 ℃.The optimized dripping conditions were as follows:the dropping distance was 10 cm;the dropping speed was 50~60 drops per minute;the internal diameter of the burette was 5.0 mm and the external diameter of the burette was 3.0 mm.CONCLUSIO-N:This forming technology serves as experimental basis for the preparation of Duanxueliu dropping pills.
