1.Establishment and characterization of SAC clone system from a mouse SRS ascitic tumour

A clone system designated SAC was established from a SRS-82 mouse ascitic turnout cell line by using limited dilution method. Clones SAC-ⅡB_2, SAC-ⅡB_3 of this system were found to be different in their morphology, growth pattern, karyotypes and tumorigenicity from their parent cell line. Both the two clones and their parent cell line were X-C assay positive, and type A and type C virus particles were found in the cytoplasm and cell surface of these cells electronmicroscopically. Cell surface markers(ALS, Thy, SmIg G) of these two clones and their parent cell line were investigated by ABC immunohistochemical method and immunofluorescent staining technique, the results of which indicate that all of them belong to T cell origin.
2.Effects of Combination of SAHA and TRAIL on Proliferation of Triple-negative Breast Cancer Cell MDA-MB-231

Objective To investigate the effects of combined treatment of suberoylanilide hydroxamic acid(SAHA)and TNF?related apoptosis inducing ligand(TRAIL)on proliferation and morphology change of triple?negative breast cancer cell MDA?MB?231. Methods The effects of combination treatment of SAHA and TRAIL on proliferation and morphology change of MDA?MB?231 cells were monitored by RTCA. Morphology changes of MDA?MB?231 cells by different treatment factors were observed through time?lapse live cell imaging acquisition. Results Real?time cell proliferation assays showed that a synergistic effect were found when MDA?MB?231 cells were treated with combination of SAHA and TRAIL , and reached the best effect with 5μmol/L SAHA and 50 ng/mL TRAIL. The results of time?lapse live cell imaging acquisition showed that the growth inhibition of MDA?MB?231 cells with combined treatment of SAHA and TRAIL were more obvious than that with treatment of SAHA or TRAIL alone. Conclusion The combined treatment of SAHA and TRAIL induces a synergistic effect on growth inhibition in triple?negative breast cancer cell line MDA?MB?231.
3.Investigation and analysis of mental and physical condition of medical workers in Shandong province

Objective To investigate the physical and mental health of Shandong medical personnel.Methods 3200 medical personnel were randomly selected from hospitals of Shandong province,selfhealth,depression and sense of work stress were measured and analyzed statistically.Results 36.3% of medical staff was in depression state,and even about 22.5% of the subjects was in moderate or severe depression state.Male and female medical staff showed no difference in sense of work pressure.Senior medical staff showed higher working pressure than middle-class,junior-class and low-level medical personnel,but middleclass,junior medical staff and low-level medical staff showed no significant difference.In the aspect of self-health status,male and female medical personnel showed no significant differences,senior,middle-class and junior-class medical staff reported worse self-health status than low-level medical personnel.Conclusions Managers should pay attention to physical and mental health of medical personnel,adopt pertinent measures to reduce their mental pressure in order to promote their physical and mental health.
4.Clinical application of pancreatic amylase and lipase in acute pancreatitis

Objective To investigate the clinical value of pancreatic amylase (P-AMY) in acute pancreatitis, and perform methodical evaluation on total amylase (α-AMY), P-AMY and lipase.Methods 126 cases of patients with acute pancreatitis, 105 cases of disease controls and 95 healthy controls were involced in the investigation. Hitachi7600 automatic biochemical analyzer was applying to measuring α-AMY, P-AMY and lipase. The upper limit of the reference range of the total amylase, pancreatic amylase and lipase was 216 U/L, 115 U/L and 200 U/L respectively in our laboratory.Results The sensitivity of total amylase, pancreatic amylase, and lipase for the diagnosis of acute pancreatitis was 73.1%, 88.5% and 92.3% respectively;and while the specificity was 70.5%、81.9% and 82.9% respectively. The diagnostic accuracy, determined as the area under the curve, was 0.795 for total amylase, 0.868 for pancreatic amylase, and 0.886 for lipase. There was a significant difference between the total amylase and pancreatic amylase (P=0.045), but no significant difference was found between the pancreatic amylase and lipase (P=0.613) by ROC curve.Conclusion Pancreatic amylase has a higher sensitivity, specificity and diagnostic accuracy than α-AMY in acute pancreatitis, and shows a similar diagnostic performance as lipase. Therefore, P-AMY and lipase should be measured in combination in the diagnosis of acute pancreatitis.
5.Study on quality standard for Xiaoerhuichun Pills

Objective: To establish quality standard for Xiaoerhuichun Pills(Radix Scutellariae Radix Paeoniae Rubra, Rhizoma Coptidis, Folium Isatidis, Radix Puerariae, etc.). Methods: Radix Scutellariae, Radis paeoniae Rubra, Rhizoma Coptidis, Folium Isatidis and Radix Platycodonis were identified by TLC. The content for berberine hydrochloride in Xiaoerhuichun Pills was determined by TLCS canning. Results: The methodological study showed that a good linear correlation in the range of 0.05776 to 0.34656?g of the berberine hydrochloride. The average recovery of berberine hydrochloride ( n =8) was 98.69% and RSD was 0.73%, respectively. Conclusion: The method is simple, accurate and with good repeatability and high precision. The method can be used for quality control of Xiaoerhuichun Pills.
6.ETIOLOGICAL DIAGNOSIS OF CRYPTOSPORIDIOSIS

l014 faecal specimens were collected from 4 hospitals in Nanjing to investigate the infection of Cryptosporidium. Each faecal specimen was smeared and screened by auramine-phenol staining method for oocyst, the positive and suspected specimens were identified by modified acid-fast staining method and safranin-methylene blue staining method. The result showed that 13 spccimens were positive by auramine-phenol staining method and confirmed by modified acid-fast staining method. 5 suspected specimens were negative as shown by the latter two methods.The best result could be obtained when auramine-phenol staining method was first used and folloWed by modified acid-fast staining method. This-technique is simple, sensitive and reliable. It is difficult to find oocysts by safranin methylene blue staining method when the oocysts are few (Figs. l-3).
7.Inhibition of quercetin on expressions of c-fos, c-jun mRNA, and TGF-?1 in liver tissue of hepatic fibrosis mice infected by schistosome

Objective To study the therapeutical effects and mechanisms of quercetin on hepatic fibrosis in schistosome-infected mice and compare the effects with praziquantel. Methods Eighty mice were divided into four groups: among them three groups were infected by Schistosoma japonica. After 8 weeks, one group was treated with quercetin 30 mg/(kg?d) for 8 weeks, one group was treated with praziquantel 500 mg/(kg?d) for 2 d and the other was taken as model group without any treatment. The fourth group was taken as normal group. HE staining, RT-PCR, and immunohistochemical technique were used to observe the expression of hepatic c-fos and c-jun mRNA, and the changes of hepatic TGF-?1, type Ⅰ, and type Ⅲ collagen in mice infected by S. japonica before and after treatments. Results Quercetin obviously relieved the degree of hepatic fibrosis, significantly reduced the expression of c-fos, c-jun mRNA, TGF-?1, type Ⅰ, and type Ⅲ collagen compared to the model group (P
8.Small cell carcinoma of urinary bladder (report of 6 cases)

Objective To improve the understanding of small cell carcinoma of the urinary bladder. Methods The pathological and clinical data of 6 small cell carcinoma cases (4 men and 6 women, mean age of 51 years) were analyzed.Of them 2 cases had components of transitional cell carcinoma or adenocarcinoma.Two cases underwent partial cystectomy,and 4 cases,cystectomy.After operation,5 of them underwent 2~6 courses of chemotherapy. Results All the 6 cases were followed-up for 12~60 months,and they all died of tumor relapse.Their average survival time was 28 months. Conclusions Small cell carcinoma of urinary bladder accounts for 0.44% of the primary malignant tumor of urinary bladder.It has high malignancy and easily metastasizes to lymph nodes,liver and bones,thus the prognosis is bad.Radical cystectomy in combination with systemic chemotherapy has better efficacy.
9.Neuroprotective mechanisms of histone deacetylase inhibitors in ischemic stroke

Histone deacetylases (HDACs) catalyze the deacetylation of histone and nonhistone proteins. They play important roles in regulating chromatin remodeling and gene transcription,and that aberrant chromatin remodeling and transcriptional dysregulation are associated with many neurodegenerative diseases. Preclinical studies have suggested that histone deacetylases inhibitors (HDACIs) can target diverse pathophysiologies of ischemic stroke,not only reduce neuronal damage and infarct volume,but also promote the neuronal plasticity and functional recovery after ischemia. This article mainly reviews the neuroprotective mechanisms of HDACIs in ischemic stroke.
10.Mechanism of integrin in epithelial-mesenchymal transition

Invasion and migration are distinctive features of the malignant tumors.Studies showed that epithelial-mesenchymal transition ( EMT) , a conversion process with the loss of epithelial cell features and the gain of mesenchymal phenotype has been recog-nized as a key element of invasion and migration of malignancy.When EMT occurs, the downregulation of E-cadherins and the loss of adhesion in extracellular matrix play a critical role which are regarded as important indicators in the assessment of EMT.The latest re-searches indicated that integrin, one of the cell adhesion molecules family, was involved in EMT directly or indirectly through mediating either adhesion among cells, extracellular matrix or signal pathways by activating multiple kinases tyrosine phosphorylation cascade. The relationship between EMT and integrin is summarized in this article.
