Objective To verify the targeting regulatory relationship between microRNA (miR)-21-5p and mitogen-activated protein kinase kinase 3 (MKK3).Methods miR-21-5p and MKK3 targeted relationship was analyzed by bioinformatics database predict.Constructing MKK3 3'UTR reporter vector (MKK3-3U) and mutation reporter vector (MKK3-3 U-M) was constructed by synthesizing miR-21-5p mimics (mimic),miR-2 1-5p inhibitor (inhibitor),nonsense miR (NC).NC (NC1 group),mimics (mimic1 group),inhibitor (inhibitor1 group) together with MKK3-3U cotransfected human embryonic kidney (HEK293) cells,respectively.Empty vector-pYr-MirTarget (NC2 group),MKK33U (Mt group),M KK3-3U-M (Mut group) together with mimic co-transfected HEK293,respectively.The fluorescence ratio was detected by dual luciferase assay.NC,mimics,inhibitor intervened human renal tubular epithelial (HK-2) cells,respectively.RT-PCR and Western blot were used to test cell MKK3 expression level in HK-2.Results 1.The fluorescence ratios of NC1 group,mimicsl group,and inhibitor1 group were 100.00％,67.99％,133.17％,and there was difference between mimic1 group and NC1 group(t =19.93,P ＜ 0.05).2.The fluorescence ratios of NC2 group,Mt group and Mut group were 100.00％,65.30％,98.48％,and there was difference between Mt group and NC2 group (t =14.39,P ＜ 0.05).There was no significant difference between NC2 and Mut group (t =0.63,P ＞ 0.05).3.MKK3 mRNA relative expression levels of the control group mimic2 group and inhibitor2 group were 1.36 ± 0.02,1.01 ± 0.04,1.43 ± 0.06 ; relative protein expression levels were 0.97 ± 0.05,0.62 ± 0.06,1.36 ± 0.32.MKK3 mRNA and protein levels of mimic2 group were significantly lower compared with the control group (F =85.98,118.26,all P ＜ 0.01).Conclusions MKK3 is the target gene of miR-21-5p.miR-21-5p can regulate the expression of MKK3 both in mRNA and protein levels.miR-21-5p may be an important regulatory molecule in p38 mitogen-activated protein kinase signaling pathway.

Objective To investigate the expression of visfatin in the livers of rats suffering from non-alcoholic fatty liver disease (NAFLD) and to expect the development of new diagnosis and treatment for NAFLD.Methods A total of 40 Sprague-Dawley (SD) rats was randomly divided into control and NAFLD model groups.For control group,the rats were given basic diet and subcutaneous injection of saline(0.2 ml/100 g weight)twice a week.However,rats in the model group were given high fat diet and were given subcutaneous injection of 40 ％ carbon tetrachloride (CCl4) salad oil (0.2 ml/100 g) twice a week.The levels of alanine aminotransferase (ALT),aspartate aminotransferase (AST),triglyeride (TG),total-cholesterol (TC) in serum together with the visfatin expression in hepatic tissue were analyzed at the 2nd,4th,6th,and 8th weeks after operation,respectively.Results The levels of ALT,AST,TG,and TC in model group were increased gradually as time going(P ＜ 0.05).Immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR) experiments showed the level of visfatin in model group was significantly higher than control group in each period,with a increasing trends with the modeling time (P ＜ 0.05).Conclusions (1)After high-fat feeding and intraperitoneal injection of CCL4,a model of adult SD rats suffering from NAFLD was constructed.(2)In livers of rats suffering from NAFLD,the expression of visfatin was increased while the level of liver steatosis was enhanced.Visfatin would be a new target to observe the severity of NAFLD.

Results: To obtain the repetitive results, the foundation methods of nano-dispersion systems should be recorded in detail and all the following tests should be based on it.Conclusion: The constitution and evaluation of a dispersion system in obtaining representative and reproducible results in biological evaluation is very important and should be studied case by case.

Nursing care was given to 48 HBV-related liver failure cases with abdominal distension through the application of Qi-Tou-Xiao-Gu therapy. It was showed that key points in nursing care should contain aspects of
correct selection of cases, detailed explanation, observation during application, maintain the moisture of Qi-Tou-Xiao-Gu stick, emotional regulation, and cross infection prevention. Good nursing care can improve patients' com-fort degree and satisfaction to the treatment. It can also effectively relieve abdominal distention and reduce ad-verse reactions.

The construction and management must be unified,the teaching content must be specified and the teaching methods should be improved.Integrable ware teaching library for sexual health education was constructed taking teaching and research section as unit.In the process of teaching,initial resuhs were achieved and teaching efficiency was improved.In the future,teaching material library and platform construction should be improved in order to meet the requirements of modem teaching.

Objective To explore the clinical features, diagnosis, treatment, and prognosis of pertussis in neonates. Methods Clinical data from 17 neonates with pertussis conifrmed by real-time polymerase chain reaction (PCR) of Bordetella pertussis, were retrospectively analyzed. Results In 17 neonates ( 8 males and 9 females), 2 neonates were premature. The age at onset was from 15 to 27 days. Thirteen cases had a history of close contact with coughing patients, 4 cases had nasal congestion and runny nose in the early stage, 5 cases had typical spastic cough, one case had cough with inspiratory“whoop”, 6 cases had lfushing in face after severe cough, 7 cases had severe cough with cyanosis, 2 cases had systemic cyanosis before hospitalized, 7 cases had regurgitation after cough, and 3 cases had fever. Fifteen cases had increased peripheral blood white blood cell count, ( 12 . 57-79 . 63 )× 109/L, with lymphocyte at 45 . 1% -75 . 2%. The PCR copy number of pertussis was 7 . 12 × 102-1 . 04 × 108/mL. Seventeen neonates were all treated with intravenous infusion of erythromycin, and all of them had a signiifcantly improvement, and no death case. Conclusions Neonates with a history of contact with coughing patients, paroxysmal cough and increased of peripheral blood leucocyte presented as lymphocyte increase, need to be alert to pertussis, and relative examination should be taken as early as possible.

To analyze the clinical features and prognosis of fulminant Wilson's disease (FWD) in patients with hepatitis B virus (HBV) infection.Twenty-seven patients were enrolled in Guangzhou Eighth People's Hospital from 2005 to 2015,including 13 FWD patients with HBV infection and 14 FWD patients without HBV infection.Clinical efficacy and survival rate were evaluated.Baseline biochemical data in two groups were comparable(P ＞ 0.05),including total bilirubin,prothrombin activity,serum albumin,alpha fetal protein,alanine transaminase,ceruloplasmin and 24 hours urine copper.Treatment in FWD group with HBV infection was ineffective,including 9(9/13) deaths and 4(4/13) patients receiveing liver transplants.However,7 (7/14) cases in the other group did not response to the treatment,including 6 (6/14) deaths and 1 (1/14)patient receiving liver transplant.The prognosis in the two groups is significantly different(P =0.006),which is much worse in FWD patients with HBV infection.

Objective To analysis pudendal nerve block anesthesia combined with noninvasive delivery on maternal vaginal delivery quality.Methods 60 primipara who were received vaginal delivery in our hospital were collected.All primipara were randomly divided into anesthesia group and no anesthesia group by the order of entry, 30 cases in each group.The no anesthesia group was given noninvasive delivery operation, and the anesthesia group received unilateral or bilateral nerve block anesthesia.Perineal laceration degree and incidence rate, time of the second stage of labor, neonatal asphyxia and two groups of maternal postpartum recovery were compared after the treatment.Results After the birth of a child, Compared with no anesthesia group, the incidence of maternal perineal laceration was lower in the anesthesia group, and the difference was statistically significant(P<0.05);the second stage of labor time of primipara was shorter in the anesthesia group(P<0.05); the pain score,getting out of bed activity time,average length of hospital stay,and incidence of postpartum complications of primipara were lower in the anesthesia group(P<0.05).Conclusions Pudendal nerve block anesthesia combined with noninvasive delivery operation can improve vaginal delivery quality, and have a guiding significance to clinical.

Objective To detect the mutation status of PIK3CA genes in breast cancer and to analyze its relation with clinico‐pathological characteristics .Methods The paraffin tissue samples in 176 cases of breast cancer were selected and the tissue DNA was extracted ,exon9 and exon20 were amplified by ARMS‐PCR .The mutation status of PIK3CA genes were detected ,other tissue samples in 20 cases of mammary gland disease were taken as the negative control .Results Among 176 samples of breast cancer tis‐sues ,45 cases of mutation were found with the mutation rate of 25 .6% ,which was dominated by H1047R mutation rate of exon20 with the mutation rate of 26 .7% (12/45);There were statistical difference between ER (+ ) and ER(-) and between PR(+ ) and PR(-) ,but no statistical difference was found between HER (+ ) and HER(-);PIK3CA gene mutation had correlation with cancer histological grade ,but no obvious correlation with tumor size ,age and lymph node status .Conclusion PIK3CA gene muta‐tion is correlated with breast cancer hormone receptor (HR) expression and tumor progression ;the PIK3CA gene mutation detection has an important significance for guiding the formulation of clinical indiidualized treatment .
