1.Effects of Cisplatin on Proliferation and Invasion of Colorectal Cancer Cells
Tianjin Medical Journal 2014;(7):638-640
Objective To investigate the effect of cisplatin on proliferation and invasion of colorectal cancer cell line SW480. Methods SW480 cells were used as the research object, and untreated SW480 cells were used as the control group. Different concentrations of cisplatin was given at different times to intervene. The MTT, Transwell chamber and phos-phate determination methods were used to detect proliferation, invasion and Na+-K+-ATPase activity expression level in SW480 cells. Results The physiological concentration of cisplatin (70 μmol/L) inhibited the proliferation of SW480 cells at 48 h. There was no significant difference in the inhibition rate at 48 h compared with 72 h and 96 h. Treatment with 70 μmol/L cisplatin for 48 h reduced the number of cells through Matrigel membrane matrix. The Na+-K+-ATPase activity was significantly increased in 35, 70 and 140 μmol/L cells after treatment with 17.5, 35, 70 and 140 μmol/L of cisplatin in SW480 cells for 48 h, and Na+-K+-ATPase reached the highest level at 70 μmol/L of cisplatin. Conclusion The decreased activity of Na+-K+-ATPase may lead to the attenuation in proliferation and invasion of colorectal cancer cells, which may be associated with cisplatin resistance in colorectal cancer cell SW480.
2.Research advance of protein fingerprinting technology in laboratory diagnosis and clinical medicine
Basic & Clinical Medicine 2006;0(02):-
Protein fingerprinting technology(PFT) is a novel technology for laboratory diagnosis developed in recent five years. It has advantages of simple operation, testing quickly, high sensitivity and specificity. It is a revolutional progress for laboratory diagnosis. The application of PFT in medical field is mainly for the detection of diseases. The sensitivity and specificity in cancer detection are about 80%.Immunomic mass spectrometry(IMS) is a novel technology using the combined group antibodies for capture multi biomarkers and applying mass spectrometry to precisely analyze the modification or isoforms of the biomarker in single platform, whereas traditional assay could not be able to identify the variation of biomarkers.PFT and IMS have significantly influenced in cancer early detection, especially to evaluate cancers which did not express traditional tumor markers like AFP,CEA,etc.PFT and IMS have characteristics of early detection in the molecular and gene level.PFT and IMS are diagnostic technologies with bright future and potential applications.

6.Comparative study of negative emotion and behavioral problems between left-behind and non-left-behind children in rural areas during hospitalization
Chinese Journal of Practical Nursing 2012;28(29):21-22
ObjectiveTo discuss the incidence of negative emotion and behavioral problems between left-behind and non-left-behind children in rural areas during hospitalization. Methods175 participants included 90 cases of non-left-behind children patients and 85 left-behind children patients. The negative emotion and behavioral problems of all children were recorded and compared. ResultsCompared with the non-left-behind children patients,left-behind children manifested negative emotions such as anger, anxiety, depression and fear, more probability and incidence of behavioral problems such as dysphonia, special learning disabilities, sleep disorders, twitch disease, multiple tourtte's syndrome, attention deficit disorder, enuresis sickness and so on.There was significant differences between the two groups. ConclusionsCompared with the non-left-behind children,left-behind children patients are more likely to have negative emotion and behavioral prob-lems,it may have lasting influence on children and eventually influence the development of the physical and mental health, cause learning and adaptation difficulties. So more care and attention should be given to left-behind children, and given timely discovery and timely treatment of negative emotions and behavioral problems.
7.Preliminary Study of Single Cell Isolation and Single Cell PCR in Lymph Node Germinal Centers
Fudan University Journal of Medical Sciences 2000;27(6):460-463
Purpose To explore the value of single cell isolation from tissue slides and single cell polymerase chain reaction(PCR) in the study of lymphy node germinal centers. Methods By single-cell isolation and single-cell PCR,rearranged immunoglobulin(Ig) genes were amplified from single Ki67(+) cells and single CD3(+) cells. The Ⅴ-region family specific primers were designed to Ig heavy chain leader region and light chain κ and λ framework region Ⅰ. Results PCR efficiency of Ki67(+) cells were 18.7% and 50.0% respectively in 2 lymph nodes. However,in 40 CD3(+) T cells,no Ig gene rearrangements were observed, which confirmed the efficiency and specificity of single cell isolation and single cell PCR. Conclusions Our study demonstrated the feasibility of single cell isolation from tissue slides and single cell PCR.This makes a sound basis for the use of technique in the further study of lymphoid diseases.

9.Effects of taurine on platelet aggregation induced by benzo pyrene in vitro activation function
Acta Universitatis Medicinalis Anhui 2015;(1):116-118
The peripheral renous blood which were taken from healthy volunteers and platelet was isolated. Platelet samples were randomly divided into six groups. The effects of different concentration of taurine( Tau) on benzo py-rene( BAP) induced platelet aggregation in vitro activation function were investigated by detecting the maximum platelet aggregation rate, the positive expression rates of platelet membrane surface CD62P and PAC-1. The results showed that platelet maximum aggregation rate had significant differences between each concentration of BAP-Tau groups and solvent control group in adenosine diphosphate(ADP) and collagen induced (P<0. 01). The positive expression rates of platelet membrane surface CD62 P and PAC-1 in BAP-Tau high concentration group had signifi-cant differences compared with solvent control group ( P<0. 01 ) . Different concentration of Tau could antagonize platelet function damage induced by BAP with a concentration gradient change.
10.The relationship between LPS-induced iNOS mRNA expression and macrophages cytotoxic function
China Oncology 2001;0(02):-
Purpose:To study LPS induced iNOS mRNA expression and NO production by murine peritoneal macrophages, and their relationship with cytotoxic function of activated macrophages. Methods:LPS induced iNOS mRNA expression, production of NO and effects of activated macrophages on anti tumor were investigated separately by RT PCR, nitrate reductase method and 3HTdR incorporation. Results:LPS can upregulates iNOS mRNA expression in a dose dependent manner. L NIL(specific inhibitors of iNOS) pretreatment significantly reduced cytotoxic effect on the proliferation of HL 60 cells, but had little effect on the K562 tumor target cells on condition that NO production is sufficiently inhibited. Conclusions:LPS can effectively induce iNOS mRNA expression and NO production in murine peritoneal macrophages. NO is one of the important effector molecules in unspecific immunity of activated macrophages, but it plays different roles in cytotoxic effects of macrophages on different target tumor cells.
